Rtt109 acetylates histone H3 lysine 56 and functions in DNA replication

Acetylation of histone H3 lysine 56 (H3-K56) occurs in S phase, and cells lacking H3-K56 acetylation are sensitive to DNA-damaging agents. However, the histone acetyltransferase (HAT) that catalyzes global H3-K56 acetylation has not been found. Here we show that regulation of Ty1 transposition gene product 109 (Rtt109) is an H3-K56 HAT. Cells lacking Rtt109 or expressing rtt109 mutants with alterations at a conserved aspartate residue lose H3-K56 acetylation and exhibit increased sensitivity toward genotoxic agents, as well as elevated levels of spontaneous chromosome breaks. Thus, Rtt109, which shares no sequence homology with any other known HATs, is a unique HAT that acetylates H3-K56.     

Restoration of tolerance in lupus by targeted inhibitory receptor expression

Lupus, a multigenic autoimmune condition in which a breakdown of tolerance results in the development of autoantibodies, leads to a variety of pathologic outcomes. Despite the heterogeneity of factors influencing disease susceptibility, we demonstrate that the partial restoration of inhibitory Fc receptor (FcgRIIB) levels on B cells in lupus-prone mouse strains is sufficient to restore tolerance and prevent autoimmunity. FcgRIIB regulates a common B cell checkpoint in genetically diverse lupus-prone mouse strains, and modest changes in its expression can result in either tolerance or autoimmunity. Therefore, increasing FcgammaRIIB levels on B cells may be an effective way to treat autoimmune diseases.     

Simulations of jets driven by black hole rotation

The origin of jets emitted from black holes is not well understood; however, there are two possible energy sources: the accretion disk or the rotating black hole. Magnetohydrodynamic simulations show a well-defined jet that extracts energy from a black hole. If plasma near the black hole is threaded by large-scale magnetic flux, it will rotate with respect to asymptotic infinity, creating large magnetic stresses. These stresses are released as a relativistic jet at the expense of black hole rotational energy. The physics of the jet initiation in the simulations is described by the theory of black hole gravitohydromagnetics.     

The human genus

A general problem in biology is how to incorporate information about evolutionary history and adaptation into taxonomy. The problem is exemplified in attempts to define our own genus, Homo. Here conventional criteria for allocating fossil species to Homo are reviewed and are found to be either inappropriate or inoperable. We present a revised definition, based on verifiable criteria, for Homo and conclude that two species, Homo habilis and Homo rudolfensis, do not belong in the genus. The earliest taxon to satisfy the criteria is Homo ergaster, or early African Homo erectus, which currently appears in the fossil record at about 1.9 million years ago.     

Oxygen isotopes and emerald trade routes since antiquity

Oxygen isotopic compositions of historical emerald artifacts from the Gallo-Roman period to the 18th century indicate that during historical times, artisans worked emeralds originating from deposits supposedly discovered in the 20th century. In antiquity, Pakistani and Egyptian emeralds were traded by way of the Silk Route. Together with Austrian stones, they were the only source of gem-quality emeralds. Immediately after the discovery of the Colombian mines by Spaniards in the 16th century, a new trade route was established, first via Spain to Europe and India and then directly via the Philippines to India. Since then, Colombian emeralds have dominated the emerald trade, and most of the high-quality emeralds cut in the 18th century in India originated from Colombia.     

Life sciences: microorganisms in the space environment

Preliminary results of the Spacelab 1 experiment on the response of Bacillus subtilis spores to conditions of free space are presented. Exposure to the vacuum of space on the Spacelab pallet reduced viability counts about 50 percent and increased mutation frequencies by a factor of about 10. Interpretation of apparent differences in the photobiological and photochemical data between flight and ground simulation experiments will require more statistical analyses and data from actual fluence measurements.     

Point mugu, california, earthquake of 21 february 1973 and its aftershocks

Seismological investigations show that the Point Mugu earthquake involved north-south crustal shortening deep within the complex fault zone that marks the southern front of the Transverse Ranges province. This earthquake sequence results from the same stress system responsible for the deformation in this province in the Pliocene through Holocene and draws attention to the significant earthquake hazard that the southern frontal fault system poses to the Los Angeles metropolitan area.     

Nature of "sex-ratio" agent in Drosophila

Several lines of evidence implicate small spirochetes, presumably treponemata, as etiologic agents in the production of the maternally transmitted "sex ratio" condition (SR) in Drosophila nebulosa, in D. willistoni, and in strains of D. melanogaster into which the SR condition has been artificially transferred. The presence of treponemata in the hemolymph of adult females of these species is completely correlated with the production of unisexual progenies and like this condition is dependent on the genotype of the host and of the infectious agent.
