The effects of time of feed delivery (14.00 hours; 14.00 and 20.00 hours; 16.00 and 18.00 hours) on the physiological parameters, ingestive behavior, nutrient intake and production of lactating cows maintained in a semi‐arid region during the hot season were evaluated. Regardless of treatment, all animals received the first feeding supply at 06.00 hours. Eight cows with an average body weight of 600 kg, average milk yield of 20 kg/day and 80 days in milk were utilized. The rectal temperature, respiratory rate and sweating rate were not affected (P > 0.05), with average values of 38.5°C, 53.8 movements/min and 104 g/m2/h respectively. There was no effect (P > 0.05) on the eating time (314 min/day), ruminating time (564 min/day), drinking time (61 min/day) and idle time (502 min/day). Similarly, the intake of nutrients and performance of lactating cows were not affected (P > 0.05), with average dry matter intake of 19.8 kg/day, 4% fat‐corrected milk of 20.6 kg/day and milk fat concentration of 4.03 g/100 g. Since the behavior and performance have not altered, any times of feed delivery evaluated could be used to crossbred Holstein × Zebu cows maintained on a feedlot in semi‐arid regions during the hot season.  相似文献

Three sward heights (15, 25 and 35 cm) and three supplement types (energy, energy‐protein, and a mineral mix supplement) were evaluated in a 3 × 3 factorial arrangement distributed in a completely randomized design to study changes in forage search patterns in Nellore heifers in a continuous grazing system. Pasture data were collected using two replicates (paddocks) per treatment over four periods during the rainy season. The behavior assessments were made in the first and fourth grazing seasons. It was hypothesized that supplements and pasture management would modify ingestive behavior, considering that animals would require less time grazing if they had energy requirements met through higher digestibility of better managed paddocks, or use of supplements high in energy. Total and green forage masses along with green : dead material ratio were greater in treatments managed with higher sward heights. Sward managed with 35 cm height resulted in lower leaf : stem ratio compared with 15 cm sward height treatments. The animals on the 15 cm pastures spent more time grazing overall and during each meal, but there were no differences observed in meal numbers in comparison to 35 cm treatments. Heifers fed protein and/or energy supplements spent less time grazing in the early afternoon, but overall grazing time was the same for all animals.  相似文献

We examined the potential growth of clonal Eucalyptus plantations at eight locations across a 1000+ km gradient in Brazil by manipulating the supplies of nutrients and water, and altering the uniformity of tree sizes within plots. With no fertilization or irrigation, mean annual increments of stem wood were about 28% lower (16.2 Mg ha−1 yr−1, about 33 m3 ha−1 yr−1) than yields achieved with current operational rates of fertilization (22.6 Mg ha−1 yr−1, about 46 m3 ha−1 yr−1). Fertilization beyond current operational rates did not increase growth, whereas irrigation raised growth by about 30% (to 30.6 Mg ha−1 yr−1, about 62 m3 ha−1 yr−1). The potential biological productivity (current annual increment) of the plantations was about one-third greater than these values, if based only on the period after achieving full canopies. The biological potential productivity was even greater if based only on the full-canopy period during the wet season, indicating that the maximum biological productivity across the sites (with irrigation, during the wet season) would be about 42 Mg ha−1 yr−1 (83 m3 ha−1 yr−1). Stands with uniform structure (trees in plots planted in a single day) showed 13% greater growth than stands with higher heterogeneity of tree sizes (owing to a staggered planting time of up to 80 days). Higher water supply increased growth and also delayed by about 1 year the point where current annual increment and mean annual increment intersected, indicating opportunities for lengthening rotations for more productive treatments as well as the influence of year-to-year climate variations on optimal rotations periods. The growth response to treatments after canopy closure (mid-rotation) related well with full-rotation responses, offering an early opportunity for estimating whole-rotation yields. These results underscore the importance of resource supply, the efficiency of resource use, and stand uniformity in setting the bounds for productivity, and provide a baseline for evaluating the productivity achieved in operational plantations. The BEPP Project showed that water supply is the key resource determining levels of plantation productivity in Brazil. Future collaboration between scientists working on silviculture and genetics should lead to new insights on the mechanisms connecting water and growth, leading to improved matching of sites, clones, and silviculture.  相似文献

This study was designed to determine the effect of intravenous lipopolysaccharide (LPS) administration on the secretion of interleukin (IL)‐1β and IL‐1 receptors (IL‐1Rs) gene expression in the hypothalamus of anoestrous ewes. Gene expression of IL‐1β and its receptors was assayed by the real‐time polymerase chain reaction. The expression of IL‐1β in the hypothalamus was detected using Western blot. Our results showed that IL‐1β mRNA is transcribed in the ovine hypothalamus. Lipopolysaccharide increased (p ≤ 0.01) the IL‐1β gene expression in the pre‐optic area 2.4‐fold, the anterior hypothalamus (AHA) 3.4‐fold, the medial basal hypothalamus 3.7‐fold and the medial eminence 3.9‐fold. The pro‐form and mature form of IL‐1β protein were found in the hypothalamus after endotoxin injection. In general, the endotoxin also increased more than two times (p ≤ 0.01) the expression of IL‐1 receptor type I (IL‐1R1) and type II (IL‐1R2) genes in the hypothalamus, except the AHA, where the number of IL‐1R2 mRNA was extremely low and not sufficient to the quantitative analysis. These results demonstrate that the peripheral immune/inflammatory challenge increases the IL‐1β expression in the hypothalamus. This endogenous IL‐1β seems to be involved in the modulation of processes which are regulated at the hypothalamic level. One of these processes could be a reproduction.  相似文献

Purpose

Several interactions between Al and the solid phase of soil influence Al buffering in soil solution. This work evaluated soils cultivated with Pinus taeda L. to determine Al forms in organic and mineral horizons using various extraction methods and to relate acidity with clay mineralogy.

Materials and methods

Organic and mineral horizons of 10 soil profiles (up to 2.1 m deep) in southern Brazil were sampled. Organic horizons were separated into fresh, aged, and fermented/humified litter. The following Al extraction methods were utilized: 0.5 mol L?1 pH 2.8 CuCl2–Al complexed in organic matter; 1.0 mol L?1 KCl–exchangeable Al; water–Al soluble in soil solution; HF concentrated?+?HNO3 concentrated?+?H2O2 30% (v/v)–total Al. Six sequential extractions were carried out to isolate different forms of amorphous minerals that can buffer Al on soil solution: 0.05 and 0.1 mol L?1 sodium pyrophosphate; 0.1 and 0.2 mol L?1 ammonium oxalate; 0.25 and 0.5 mol L?1 NaOH. Samples of clay were also analyzed by XRD.

Results and discussion

There was a clear effect of litter age on increasing total Al concentration. In the aged litter and fermented and/or humified litter, levels of total Al were 1.4 to 3.8 and 1.5 to 7.8 times greater than in fresh litter, respectively. The CuCl2 method had higher Al extraction capacity than the KCl method for litter. The lowest Al–pyrophosphate values were observed in the Oxisol, which also had a predominance of gibbsite and the lowest levels of Al–KCl and Al–CuCl2. There was an inverse relationship between degree of soil weathering and soluble and exchangeable Al in soils. Available Al increased with higher Si proportion in minerals of the clay fraction (2:1?>?1:1?>?0:1).

Conclusions

The worst scenario was soils with the combination of high soluble and exchangeable Al levels and high concentrations of amorphous forms of Al minerals. The best predictors of Al accumulation in the youngest litter horizon were extractions of amorphous minerals with pyrophosphate and NaOH. These extractors are normally used to predict the level of Al buffering in soils. Organic matter had less influence on Al dynamics in soils.

γδ T cell responses are induced by various viral and bacterial infections. Different γδ T cells contribute to activation and regulation of the inflammatory response and to epithelial repair. How γδ T cells respond to rotavirus infection and how the colonization of probiotics influences the γδ T cell response were unknown. In this study, we evaluated by multicolor flow cytometry the frequencies and distribution of total γδ T cells and three major subsets (CD2-CD8-, CD2+CD8- and CD2+CD8+) in ileum, spleen and blood of gnotobiotic (Gn) pigs at early (3-5 days) and late phases (28 days) after rotavirus infection. The Gn pigs were inoculated with the virulent human rotavirus Wa strain and colonized with a mixture of two strains of probiotics Lactobacillus acidophilus and Lactobacillus reuteri. In na?ve pigs, the highest frequency of total γδ T cells was found in blood, followed by spleen and ileum at the early age (8-10 days old) whereas in older pigs (32 days of age) the highest frequency of total γδ T cells was found in ileum and spleen followed by blood. Rotavirus infection significantly increased frequencies of intestinal total γδ T cells and the putatively regulatory CD2+CD8+ γδ T cell subset and decreased frequencies of the putatively proinflammatory CD8- subsets in ileum, spleen and blood at post-infection days (PID) 3 or 5. The three γδ T cell subsets distributed and responded differently after rotavirus infection and/or lactobacilli colonization. The CD2+CD8+ subset contributed the most to the expansion of total γδ T cells after rotavirus infection in ileum because more than 77% of the total γδ T cells there were CD2+CD8+ cells. There was an additive effect between lactobacilli and rotavirus in inducing total γδ T cell expansion in ileum at PID 5. The overall effect of lactobacilli colonization versus rotavirus infection on frequencies of the CD2+CD8+ γδ T cell subset in ileum was similar; however, rotavirus-infected pigs maintained significantly higher frequencies of CD8- subsets in ileum than lactobacilli-colonized pigs. The dynamic γδ T cell responses suggest that γδ T cell subsets may play important roles in different stages of immune responses after rotavirus infection and probiotic colonization. The knowledge on the kinetics and distribution patterns of γδ T cell subsets in na?ve pigs and after rotavirus infection or lactobacilli colonization provides the foundation for further mechanistic studies of their functions.  相似文献

ABSTRACT The population structure of Guignardia citricarpa sensu lato (anamorph: Phyllosticta citricarpa), a fungus of which strains pathogenic to citrus are subject to phytosanitary legislation in the European Union and the United States, was investigated. Internal transcribed spacer sequences revealed two phylogenetically distinct groups in G. citricarpa. This distinction was supported by amplified fragment length polymorphism analysis that also supported the exclusion of two isolates that had apparently been misclassified as G. citricarpa. On cherry decoction agar, but not on other media, growth rates of group I isolates were lower than those of group II isolates. Conidial dimensions were similar, but group I isolates formed conidia with barely visible mucoid sheaths, whereas those of group II formed conidia with thick sheaths. Cultures of isolates belonging to group I produced rare infertile perithecia, whereas fertile perithecia were formed by most isolates of group II. Colonies of isolates belonging to group I were less dark than those of group II, with a wider translucent outer zone and a lobate rather than entire margin. On oatmeal agar, exclusively group I isolates formed a yellow pigment. Group I harbored strains from citrus fruits with classical black spot lesions (1 to 10 mm in diameter) usually containing pycnidia. Group II harbored endophytic strains from a wide range of host species, as well as strains from symptomless citrus fruits or fruits with minute spots (<2-mm diameter) without pycnidia. These observations support the historic distinction between slowly growing pathogenic isolates and morphologically similar fast-growing, nonpathogenic isolates of G. citricarpa. The latter proved to belong to G. mangiferae (P. capitalensis), a ubiquitous endophyte of woody plants with numerous probable synonyms including G. endophyllicola, G. psidii, P. anacardiacearum, and P. theacearum. G. mangiferae occurs in the European Union and the United States on many host species including citrus, and does not cause symptoms of citrus black spot, justifying its exclusion from quarantine measures.  相似文献
