Background

Salmonella enterica serovar Enteritidis is a zoonotic pathogen. Human infections are associated with contaminated eggs and egg products. In Japan, since 1989, the incidence of food-borne disease caused by S. Enteritidis has increased and a pandemic has occurred; however, little is known about changes that occurred before and after this pandemic event in the dominant lineage of isolates from food-producing animals. This study aimed to determine the S. Enteritidis lineages in Japan over the last few decades by using multilocus variable-number tandem repeat analysis (MLVA).

Findings

MLVA was used to analyse 79 S. Enteritidis isolates collected from chickens (n = 63), cattle (n = 12), pigs (n = 2), and goats (n = 2) during 1975–2009. The S. Enteritidis isolates showed 14 different MLVA allele combinations, which were classified into two major clusters (A and C) and a minor cluster (B). All the 62 isolates in cluster A were isolated after 1988, whereas 13 of the 17 isolates belonging to cluster B and C were isolated before 1989.

Conclusions

The MLVA results showed that cluster C was predominant before 1989, and isolates in cluster A disseminated since 1989 and replaced the previous dominant clone, suggesting that isolates of cluster A originated from imported S. Enteritidis infection.  相似文献

Sequences of the internal transcribed spacer (ITS) region 1 were used to examine the phylogenetic relationships among races of 19 isolates of Phytophthora vignae f. sp. adzukicola and between this forma specialis and three isolates of the closely related P. vignae f. sp. vignae. The ITS 1 sequences were highly conserved (> 98.7% similarity) among representatives of both formae speciales groups. The results of this study indicate that P. vignae is a monophyletic group. Photosystems harvest light energy, yet this energy cannot be efficiently employed for CO(2) assimilation at the below-freezing temperatures to which plants are typically exposed during winter in the temperate and boreal zones. To elucidate the mechanisms whereby this energy is dissipated, I evaluated performance of photosystems in winter needles of the evergreen tree Taxus cuspidata Sieb. et Zucc. Chloroplasts were localized adjacent to plasma membranes in needle cells in summer, whereas they congregated together in the centers of the cells during winter. When winter needles were acclimated to a temperature of 20 degrees C, their chloroplasts gradually dispersed to the edges of the cells, as in the summer. Acclimation-dependent relocalization coincided with changes in CO(2) uptake. Examination of photosystem II fluorescence kinetics in winter needles indicated that the quinone electron acceptor (Q(A)) reduction rate exceeded the Q(A) oxidation rate at low temperatures. The majority of Q(A) remained reduced even when winter needles were subjected to a temperature of -5 degrees C at low irradiance.  相似文献

We investigated morphological and photosynthetic responses of current-year seedlings of oak (Quercus crispula Blume) under high-light conditions. Quercus crispula seedlings were grown from seed in a relative photosynthetically active photon flux density (RPPFD) of 100, 10 or 2%. There was no difference in total dry mass between 100 and 10% RPPFD. At the end of the growing season, plants grown in 2% RPPFD had a lower total dry mass than those grown in 100 or 10% RPPFD. Seedlings grown in 100% RPPFD showed morphological acclimation, i.e., high root/shoot ratios and high leaf mass per area. De-epoxidation level in the xanthophyll cycle and activity of an antioxidant enzyme were highest in 100% RPPFD, but total chlorophyll concentration and photosynthetic rate were highest in 10% RPPFD. These results indicate that excess photons were generated in 100% RPPFD, leading to increased capacities for dissipation of received light energy through the xanthophyll cycle and for scavenging of reactive oxygen species through the water-water cycle. Nevertheless, a midday decrease in dark-adapted quantum yield of photosystem II (F(v)/F(m)) indicated that seedlings grown in 100% RPPFD were suffering from photoinhibition. We conclude that Q. crispula current-year seedlings have high morphological acclimation to high light but that photosynthetic efficiency cannot be maintained under high-light conditions even with a photoprotection system.  相似文献

Stream macroinvertebrate assemblages are expected to be affected by the abundance and constitution of litter from surrounding forests. We compared forest floor cover, overland flow, stream environment, and stream macroinvertebrate assemblages between the catchments of a Japanese cedar plantation (CP) and a primary deciduous forest (DF). Both systems experience excessive deer browsing. Understory vegetation cover was higher in the DF than in the CP in summer, although cover was low (<20 %), possibly because of excessive deer browsing. Litter cover was much higher in the CP than in the DF in summer as a result of the long abscission period, slow breakdown, and low rate of dispersal of Japanese cedar litter compared to deciduous litter. Monthly overland flow was always lower in the CP than in the DF, and substrate size was smaller in the DF stream. In the CP, cedar litter accumulated in the stream, probably because of its low breakdown rate and morphology, and abundant shredder taxa characterized the macroinvertebrate assemblage. In contrast, abundant burrower taxa characterized the macroinvertebrate assemblage in the DF stream. These results imply that Japanese cedar litter functions in structuring the macroinvertebrate assemblage by supplying persistent food resources for detritivores, and by buffering fine sedimentation via overland flow under excessive deer browsing.  相似文献

Previous studies have demonstrated a clear relationship between diameter at breast height (DBH) and tree transpiration (Q T) in multi-specific broadleaved forests. However, these studies were conducted with a limited range of tree sizes and species, and thus many multi-specific broadleaved forests fall outside these conditions. Therefore, this study examined the relationship between DBH and Q T in a warm-temperate multi-specific broadleaved forest (n = 12 species) with a wide range of tree sizes (5.0–70.0 cm DBH) using the Granier-type heat dissipation method. The results showed that, although sap flow density varied between individual trees and species, there was a significant relationship between log Q T and log DBH (r 2 = 0.66, P < 0.001) because of the strong dependence of sapwood area on DBH. This study confirmed the applicability of the relationship for the stand transpiration (E C) estimates even in a multi-specific broadleaved forest with a wide variation in DBH. Our results also revealed that selecting the sample trees in descending order of DBH effectively reduced potential errors in E C estimates for a specific sample size, as larger trees contribute more to E C. This information should be useful for future studies investigating the transpiration of multi-specific broadleaved forests, reducing errors during the scaling-up procedure.  相似文献

Decentralized forest policy has been moderately successful in delivering resource-use rights to local people. At the same time, it is possible that decentralization leads to recentralization because governments never give their authority over forest resources. Recentralization studies have paid little attention to the potential of local dynamics to lead to institutional arrangements that affect forest outcomes. This paper uses a case study of Community-based Forest Management (CBFM) in the Philippines to explore how local realities lead to the development of effective institutions for forest management. In this case study, local informal regulations of forest resource use were created through the process of settling local conflicts among competing CBFM interests, including members and non-members of people’s organizations, and frontline foresters who are working at local level. Frontline foresters played a role as coordinator of institutional arrangements that regulate local forest exploitation within the CBFM implementation process. The behavior of frontline foresters affected by their own personalities and existing social relations among residents, can deter recentralization in some ways. More attention is needed on the role of frontline foresters and non-members of people’s organization as influential negotiators in state-society relations concerning forests.  相似文献

Histidine decarboxylase (HDC) from Staphylococcus epidermidis TYH1, a halotolerant histamine-producing bacterium isolated from Japanese fermented fish-miso, was purified to homogeneity for the first time. The enzyme was purified 182-fold from cell-free extracts by ammonium sulfate precipitation, anion exchange chromatography and gel filtration chromatography. The N-terminal amino acid sequences of two polypeptide chains of 27–30 and 7–9 kDa were highly homologous with those of α- and β-chains of other staphylococcal HDCs. The optimum pH and temperature for the enzyme were 6.0 and 60 °C, respectively. This enzyme did not decarboxylate lysine, arginine, tyrosine, tryptophan or ornithine. The enzyme activity decreased with the addition of NaCl. At pH 4.8, the V max and K m values were 45.5 μmol histamine min?1 mg?1 and 1.10 mmol/L, respectively. Moreover, this enzyme was resistant to heat treatment (80 °C for 15 min) and was stable upon freezing at ?30 °C for 7 days. The very similar physiological properties of this enzyme and the almost identical N-terminal amino acid sequence to that of the HDC from S. capitis indicated that this enzyme may be evolutionally highly conserved in the genus Staphylococcus. The biophysical properties of staphylococcal HDC were elucidated using native purified enzyme.  相似文献

The antifungal glycoalkaloid -tomatine accumulates in tomato plants and may protect plants from fungal infection. Fusarium oxysporum f. sp. lycopersici, the causal agent of vascular wilt of tomato, produces a tomatinase (FoToml) that degrades -tomatine to the nontoxic compounds tetrasaccharide lycotetraose and tomatidine. Induction of tomatinases and the distribution of FoToml homologs were examined among 30 strains belonging to 16 formae speciales of F. oxysporum. Tomatinase activity was found in 27 strains belonging to 15 formae speciales, but FoToml homologs (>98% sequence identity) were detected in only six strains belonging to four formae speciales. To identify tomatinases other than FoToml, -tomatine-inducible proteins of another tomato pathogen F. oxysporum f. sp. radicis-lycopersici were analyzed by two-dimensional gel electrophoresis. A protein with a molecular mass of 64kDa accumulated in the -tomatine-induced culture filtrates, and the protein had tomatinase activity, degrading -tomatine to lycotetraose and tomatidine.  相似文献

Lysophosphatidic acid (LPA) is a bioactive phospholipid that stimulates cell proliferation and migration, and protects cells from apoptosis. It interacts with specific G protein-coupled transmembrane receptors. Recently, frequent mutations of the LPA receptor-1 (LPA1) gene were detected in rat lung adenocarcinomas induced by N-nitrosobis(2-hydroxypropyl)amine (BHP). In this study, to evaluate the involvement of other LPA receptor gene alterations during lung carcinogenesis, we investigated mutations of the LPA2, LPA3, LPA4 and LPA5 genes in lung adenocarcinomas induced by BHP in rats. Fifteen male Wistar rats, 6 weeks of age, were given 2000 ppm BHP in their drinking water for 12 weeks and then maintained without further treatment until sacrifice at 25 weeks, and 15 adenocarcinomas were obtained. Genomic DNAs were extracted from frozen tissues, and the LPA2, LPA3, LPA4 and LPA5 genes were examined for mutations, using polymerase chain reaction (PCR)-single strand conformation polymorphism (SSCP) analysis. No mutations of LPA2, LPA3, LPA4 and LPA5 were detected in the 15 adenocarcinomas. These results suggest that alterations due to LPA2, LPA3, LPA4 and LPA5 gene mutations might not be involved in the development of lung adenocarcinomas induced by BHP in rats.  相似文献
