Modification of rice starch by selective degradation of amylose using alkalophilic Bacillus cyclomaltodextrinase

A cyclomaltodextrinase (CDase) isolated from alkalophilic Bacillus sp. I-5 (CDase I-5) exists in a dodecameric form, an assembly of six dimers, each catalytic site of which is located in a narrow groove at the interface of the dimeric unit. Because of the unique geometric shape of the catalytic site, the enzyme has the ability to discriminate the molecular size of substrates. An analysis of the hydrolysis reaction of the enzyme revealed that its kcat/Km value on amylose was 14.6 s(-1) (mg/mL)(-1), whereas that for amylopectin was 0.92 s(-1) (mg/mL)(-1), showing an exceptionally high preference toward amylose. CDase I-5 was applied to modify the starch structure to produce low-amylose starch products by incubating rice starch with this enzyme. We found that the amylose content of rice starch decreased from 28.5 to 9%, while the amylopectin content remained almost constant with no significant change in the side chain length distribution. When the CDase I-5-treated rice starch was stored at 4 degrees C for 7 days, the retrogradation rate was significantly retarded as compared to that in the control sample.     

Investigation of a novel, heritable bleeding diathesis of Thoroughbred horses and development of a screening assay

BACKGROUND: Bleeding in racing horses associated with exercise appears to be multifactorial, and clinical investigation into severe cases rarely occurs. Previously, we reported a severe bleeding diathesis in a Thoroughbred mare. Herein, we describe the cellular physiology of this defect, provide a diagnostic tool for identifying it, and demonstrate that the dysfunction is heritable. HYPOTHESIS: The subject has a heritable defect in platelet secretion that reduces thrombin generation in the absence of additional plasma factors and delays the onset of thrombin production even in the presence of these factors. ANIMALS: The study included 3 clinically normal Thoroughbred horses: the subject and her offspring. METHODS: Washed platelets were examined for their ability to (1) translocate phosphatidylserine to the outer leaflet of the platelet membrane as determined by annexin-V binding, (2) generate thrombin as assessed by the activity of the prothrombinase enzyme complex, and (3) bind fibrinogen and form aggregates as determined by flow cytometry. RESULTS: Subject and offspring platelets created procoagulant surfaces by translocating phosphatidylserine. The subject's platelets demonstrated reduced prothrombinase activity, resulting in decreased production of thrombin relative to control platelets. Subject and offspring platelets bound less fibrinogen than control platelets when stimulated with thrombin. CONCLUSIONS AND CLINICAL IMPORTANCE: The subject mare has a transmissible defect that involves reduced generation of thrombin by activated platelets, resulting in decreased aggregation and ineffective clotting. A flow cytometric assay of fibrinogen binding to washed platelets discriminates individuals with this platelet dysfunction and may be useful for discerning subclinical congenital or acquired platelet dysfunctions.     

miRNA and piRNA expression profiles of breeder cock testes detected by next‐generation sequencing

miRNAs are small non‐coding regulatory RNAs that play key roles in diverse biological processes. In this study, we used the Solexa sequencing technique to profile miRNAs in breeder cock testes to illustrate their functions. A total of 663 co‐expressed miRNAs and 3,180 co‐expressed piRNAs were detected in three libraries. Based on Mir‐X^? miRNA qRT‐PCR, three miRNAs representing low, medium and high expression levels according to the sequencing results were selected randomly to validate the miRNAs' expression profiles. Results suggested that the miRNA expression profiles data could represent actual miRNA expression levels. Moreover, target genes prediction of the co‐expressed miRNAs and further Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses were performed, which revealed that some candidate miRNAs were involved in the regulation of the spermatogenesis process, spermatozoa function and testicular metabolism. In conclusion, we provided a useful resource for further elucidation of the miRNAs' regulatory role in spermatogenesis, contributing to a preliminary database for functional and molecular mechanistic studies in testicular metabolism, spermatogenesis and other testes functions.

Web-based, interactive simulation of an anesthesia machine

Gases are transparent and the piping layout in anesthesia machines is not obvious, making it hard to understand anesthesia machine function, potential failure modes, and recovery from faults. We experimented with an approach that simplified the layout of the anesthesia machine piping by removing extraneous details and making gases not only visible, but also color-coded. More than 15 anesthesia machine controls were represented by icons that a user can adjust by clicking on the icon with a pointing device and subsequently observing the effect of his/her intervention on anesthesia machine function in real time. We simulated two anesthesia machine failures and also supported non-English languages and non-US gas color codes. Further, we initiated an experiment in Web philanthropy by deliberately choosing to make the educational module accessible without charge and selected the Web as the preferred medium for dissemination of the Virtual Anesthesia Machine (VAM) viewable at http://www.anest.ufl.edu/vam . Originally designed for use in human anesthesiology, a significant percentage (about 30%) of the 54 000 international visitors, as tracked by an independent third-party Web traffic monitoring service, for the period 9/00–5/02, were from the veterinary field. VAM is being used as part of the curriculum in veterinary programs in the US and overseas and has been well received by the global anesthesia community (human and veterinary).     

CD4+ and CD8+ T- lymphocytopenia in a filly with Pneumocystis carinii pneumonia

Decreased proportion of CD4+ and CD8+ T lymphocytes in peripheral blood likely contributed to susceptibility to Pneumocystis carinii in a foal. Cytological evaluation of bronchoalveolar lavage was required for identification of the pathogen and serial flow-cytometric analysis of peripheral blood lymphocytes documented transient low expression of CD4+ and CD8+ T lymphocytes. Although immunodeficiency is uncommon, it must be included in the differential diagnosis for patients suffering from chronic or opportunistic infections and may provide an indication for immunostimulant therapy.
