Background: The cytoreductive efficacy of the individual components of multidrug chemotherapy for canine lymphoma is difficult to evaluate after complete remission. Objectives: To compare the cytoreductive efficacy of vincristine (VCR), cyclophosphamide (CPA), and doxorubicin (DXR) in dogs that received a 6‐month modified version of the University of Wisconsin‐Madison chemotherapy protocol (UW‐25). Animals: Twenty‐nine dogs with high‐grade B‐cell multicentric lymphoma. Methods: Rearranged immunoglobulin heavy chain gene fragments from lymphoma cells were amplified by polymerase chain reaction (PCR) and sequenced to prepare clone‐specific primers and probes for real‐time PCR. The number of lymphoma cells in peripheral blood was measured from diagnosis to week 11 of UW‐25. Results: The number of lymphoma cells after the 1st administration of VCR, CPA, and DXR in weeks 1–4 was decreased in 29/29 (100%), 15/29 (51.7%), and 26/27 (96.3%) dogs, respectively. The cytoreductive efficacy of CPA was less than that of VCR and DXR. VCR, CPA, and DXR administered in weeks 6–9 were effective in 5/26 (19.2%), 5/20 (25.0%), and 14/19 (73.7%) dogs, respectively, indicating the sustained cytoreductive efficacy of DXR. CPA nonresponders were heavier and exhibited a shorter 1st remission than CPA responders. Conclusion and Clinical Importance: When using UW‐25 for treatment of canine lymphoma, CPA was found to have less cytoreductive efficacy than VCR and DXR. Real‐time PCR‐based quantification of tumor cells is an objective marker of the efficacy of chemotherapeutic agents.  相似文献

To identify factors that control coat color in Akita-inu dogs, we sequenced all the exons of the melanocortin 1 receptor (MC1R), β-defensin103 (CBD103) and agouti signaling protein (ASIP) genes of dogs with four distinct coat colors, namely, brindle, sesame, red and white. Then we examined correlations among specific alleles and coat color. In the case of the MC1R gene, all white dogs were homozygous for a nonsense mutation, R306ter, while brindle, sesame, and red dogs had at least one R306 allele. In the case of the CBD103 gene, all brindle dogs were heterozygous for the G23del mutation (deletion of codon 23, encoding glycine), while all sesame and red dogs were homozygous for G23. In the case of the ASIP gene, all dogs, regardless of coat color, had at least one S82 H83 allele. A missense mutation in the ASIP gene, P87L, was identified for the first time in some Akita-inu dogs but was not associated with any specific coloration. Our results indicate that the 2 key mutations, R306ter in the MC1R gene and G23del in the CBD103 gene, are associated with the phenotypic discriminations among brindle, red/sesame, and white coats, while no mutation that might potentially be associated with the discrimination of a sesame coat from a red coat is present in the coding sequences of these three genes.  相似文献

We evaluated age-related changes in serum osteocalcin concentrations in non-periparturient cows and variations in serum osteocalcin concentration in periparturient primiparous and multiparous cows. The serum osteocalcin levels were evaluated in 144 non-periparturient Holstein dairy cows aged 11 days to 10 years; these levels were the highest in the youngest cows, appeared to steadily decrease with age until the time of the first calving, and were subsequently maintained at low levels. Between 14 days before calving and 21 days after calving, the serum osteocalcin levels were significantly higher in the primiparous cows than in the multiparous cows. A comparison between age-matched non-periparturient and periparturient cows showed that serum osteocalcin levels were significantly lowered during late gestation in both primiparous and multiparous cows. These results suggest that serum osteocalcin measurement might be useful for the detection of mineral imbalances at the time of parturition in cows.  相似文献

Circadian rhythms of behavior are driven by oscillators in the brain that are coupled to the environmental light cycle. Circadian rhythms of gene expression occur widely in peripheral organs. It is unclear how these multiple rhythms are coupled together to form a coherent system. To study such coupling, we investigated the effects of cycles of food availability (which exert powerful entraining effects on behavior) on the rhythms of gene expression in the liver, lung, and suprachiasmatic nucleus (SCN). We used a transgenic rat model whose tissues express luciferase in vitro. Although rhythmicity in the SCN remained phase-locked to the light-dark cycle, restricted feeding rapidly entrained the liver, shifting its rhythm by 10 hours within 2 days. Our results demonstrate that feeding cycles can entrain the liver independently of the SCN and the light cycle, and they suggest the need to reexamine the mammalian circadian hierarchy. They also raise the possibility that peripheral circadian oscillators like those in the liver may be coupled to the SCN primarily through rhythmic behavior, such as feeding.  相似文献

The objective of the present study was to record the seed yield and to examine visually the quality of soybean seeds cultivated under different types and placements of urea fertilizers. In addition to the conventional fertilizer application (including ammonium sulfate 16 kg N ha-1 broadcasting (100 kg N ha-1 of urea (0B) and X00-d type coated urea CU-100 (CUB), and deep placement (100 kg N ha-1) of urea (UD) and 100-d type coated urea CU-100 (CUD) was conducted in separate plots in a paddy field converted to an upland field located at Shindori Experimental Station of Niigata University. Soybean plant growth was periodically analyzed and the quality of harvested seeds was also visually examined (hereafter referred to as “visual quality”). It was found that the deep placement treatments were more conducive 1o nitrogen (N2)fixation, based on the relative mreide N concentration in the xylem sap, which is a good indicator of N~fixation by soybean. Also the total seed yield was the highest in CUD (82 g plant-1) and 0D (81 g plant-1), compared to the control (62 g plant-1), UB (68 g plant-1), and CUB (68 g plant-1). The visual quality of harvested seeds showed that CUD enhanced the quality of seeds compared to the other treatments, in which the percentage of good quality seeds, hereafter referred to as "good seeds," based on the dry weight was 51 (control), 65 (K3B), 61 (CUB), 61 (0D), and 6696 (CUD). In terms of diseased seeds, the percentage of turtle wrinkle and broken seed coats was found to decrease by N application compared to the control. Thus, it is suggested that N fertilization management is important for maximum yield of soybean as well as for the enhancement of seed quality.  相似文献

ABSTRACT

The accident at Fukushima 1 Nuclear Power Plant in 2011 caused contamination by radioactive cesium (134Cs and 137Cs) in surrounding areas. After this accident, concerns about Cs contamination, including food safety, have limited industrial activities and reconstruction in Fukushima. Sorghum (Sorghum bicolor (L.) Moench) is an annual C4 crop that can be used as biofuel feedstock due to its high biomass. Use of Cs-contaminated fields to produce biofuel feedstock would be more acceptable than use for food or feed crops due to the lower risk of human internal exposure to radioactive Cs. In addition, high-biomass sorghum might be suitable for removal of Cs from fields (phytoremediation). For both applications, it is important to use accessions showing the appropriate level of accumulation of radioactive Cs (low for biofuel feedstock, high for phytoremediation). Here, we examined the accumulation of radioactive Cs in the aerial parts of 56 sorghum accessions grown in Fukushima. Accessions were cultivated in a low-level-contaminated field in 2011 and in a highly contaminated field in a planned evacuation zone in 2012. After cultivation, activity concentrations from 134Cs and 137Cs were measured in the aerial plant parts. In 2011, the activity concentrations of 134Cs and 137Cs were 58.2–350 and 58.6–450 Bq kg?1 dry weight, respectively. In 2012, the concentrations of 134Cs and 137Cs were 54.2–1320 and 57.1–1960 Bq kg?1 dry weight, respectively. Relative to the median values of the accessions grown each year, 3 showed lower activity concentrations and 2 showed higher activity concentrations of radioactive Cs under both cultivation conditions. In contrast to a previous report, there was no significant correlation between biomass and Cs activity concentration. Because both biomass and Cs concentration are important in classifying accessions for use in phytoremediation, we also calculated the Cs accumulation index (single-plant biomass × Cs activity concentration) for each accession. The accession AKLMOI WHITE showed the highest values in both years, suggesting that this accession has the high per-plant accumulation capacity for radioactive Cs. Our data collected from actual contaminated fields is irreplaceable for choosing sorghum accessions for cultivation in Cs-polluted sites such as Fukushima.  相似文献

Abstract

Varietal differences in the resistance to bacterial wilt and nutrient uptake of tomato (Lycopersicon esculentum Mill.) seedlings were examined in pot experiments. Twenty-three tomato cultivars were classified into three groups based on the degree of resistance to the disease. Differences in nutrient uptake among cultivars were observed for all the elements tested, and highly resistant cultivars were characterized by a high calcium uptake.  相似文献

SUMMARY: This experiment was conducted to investigate the effect of astaxanthin supplemented steam-dry pellets on the spawning of striped jack Pseudocaranx dentex . Five months before spawning, 7-year-old fish were divided into three groups, each consisting of five females and five males. The control group was fed a raw fish mix (RF) and the test diets consisted of steam-dry pellets (DP) and steam-dry pellets supplemented with 10 p.p.m. astaxanthin (a-DP). Feeding was carried out once per day, three times a week in floating net cages. The RF and the a-DP groups produced more than three times the amount of eggs produced by the DP group. The buoyancy, fertilization, and hatching rates were higher and similar for the RF and the DP groups. Total lipid contents of the diets, eggs, and larvae were similar among the groups. Highly unsaturated fatty acids (n-3 HUFA) and polar and non-polar lipids in eggs and larvae were also related to their dietary availability. These results indicate that astaxanthin, due to its various physiological functions, improved the condition of the broodstock and enhanced total egg production but not egg quality as striped jack do not incorporate carotenoids into their eggs. Therefore we confirm that the nutritional quality of diets given to striped jack prior to spawning plays a fundamental role in egg production, quality and egg composition.  相似文献

ABSTRACT:   The present study aimed to investigate functional changes of the kidney in mature masu salmon. The urine flow rate (UFR), kidney-somatic index (KSI), and epithelial cell height of second proximal convoluted segment of renal tubules were examined in mature and immature masu salmon Oncorhynchus masou . Two methods of calculating UFR were made as either the units per body weight, or body weight without gonads. The former calculation using body weight may be used for comparison between data obtained from immature fish, and the latter method was suitable when comparing mature sexes and assessing the transition from immature to finally mature stages. In both calculations of UFR, KSI, and epithelial cell height of the second proximal segment, the values for mature fish were significantly higher than for immature fish. These results first suggest that functional transition of the kidney occurs in maturing fish.  相似文献

The secretion rhythms of plasma cortisol (CORT) and prolactin (PRL), hormones related to stress responsiveness and biological rhythm and controlled by light and temperature, were investigated under varying external environments and different management techniques. Serial blood samples were collected from female cattle reared in free‐stall and freely fed (FF) conditions (n = 4) or in tie‐stall and restricted feeding (RF) conditions (hay and concentrate twice daily, n = 4). Plasma CORT and PRL concentrations, eating behavior, and environmental parameters were analyzed. Cyclic patterns for each parameter were examined using spectral analysis, and correlations between CORT, PRL and other parameters were investigated using cross‐spectral analysis. Under FF conditions, CORT secretion was not related to the lighting intensity and eating behavior. However, under RF conditions, the CORT secretion rhythm showed a distinct correlation with lighting intensity and eating behavior. Under FF conditions, the PRL secretion rhythm was similar in all seasons. However, under RF conditions, the PRL rhythm oscillated with high frequency in summer and low frequency in winter, indicating a seasonal change in rhythm. The present study demonstrates that hormone secretion rhythms change under different environments and management techniques.  相似文献
