Heading date 1 (Hd1) is one of the major determinants of flowering time and regional adaptability in rice (Oryza sativa L.), because there is a significant correlation between Hd1 allelic diversity and phenotypic differences in rice cultivars. In this study, we isolated a novel rice mutant with a Tos17 retrotransposon insertion upstream of Hd1. The Tos17 insertion mutant of Hd1 [designated as Tos17(Hd1)] had decreased photoperiod sensitivity and mRNA expression of Hd1, as compared with wild‐type Nipponbare. Between Tos17(Hd1) and Nipponbare, expression levels of OsPRR1 and OsGI were similar, whereas those of two florigen genes Hd3a and RFT1, were significantly different under both short and long‐day conditions. Tos17(Hd1) showed photoperiod sensitivity intermediate between that of Nipponbare and a near‐isogenic line having a non‐functional allele of Hd1, indicating that Tos17(Hd1) has an attenuated but functional Hd1 allele. Our results revealed that allele mining of Hd1 may provide good opportunities to develop novel rice cultivars showing different levels of photoperiod sensitivity.  相似文献

We aimed to determine the effects of nutritional status on concentrations of somatotropic axis hormones (growth hormone (GH) and insulin‐like growth factor 1 (IGF‐1)), insulin and metabolites (glucose, total protein and nonesterified fatty acids (NEFA)) in the plasma and colostrum in late antepartum cows. Eight pregnant Japanese Black cows were randomly assigned to two experimental groups (n = 4 per group). Control cows (CON) received 100% of their nutritional requirements until parturition, whereas restricted group cows (RES) received 60% of their nutritional requirements. Blood samples were taken during the antepartum period, and blood and colostrum samples were collected on days 0, 1, and 3 after calving. Compared to the CON group, the RES group had higher concentrations of GH and NEFA in plasma, but significantly lower concentrations of glucose and insulin in plasma. The concentrations of GH in plasma after calving were significantly higher, but total plasma protein was significantly lower in RES than in CON cows. Compared to the CON group, the RES group had significantly higher concentrations of GH in colostrum, but significantly lower total concentrations of protein in colostrum. Concentrations of IGF‐1 were not different between the two groups. These findings suggest that maternal nutritional status during late gestation influences concentrations of GH and total protein in the blood and colostrum of Japanese Black cows.  相似文献

When exposed to light, the Sekiguchi lesion (sl) rice mutant has an enhanced resistance to Magnaporthe grisea infection responsible for Sekiguchi lesion formation and tryptamine accumulation. Glyphosate [N-(phosphonomethyl) glycine] pretreatment suppressed Sekiguchi lesion formation and tryptamine accumulation in the sl mutant after M. grisea infection even under light. This inhibition by glyphosate was blocked by the supply of exogenous tryptophan, but not by exogenous phenylalanine. In glyphosate-pretreated leaves, 5-enol-pyruvyl-shikimate-3-phosphate synthase gene expression and tryptophan biosynthesis were significantly suppressed. During tryptophan starvation, catalase activity was maintained at a high level even under light, leading to the suppression of H2O2 generation and DNA fragmentation. These results show a strong relationship between the tryptophan and tryptamine pathways in the induction of light-enhanced resistance to M. grisea infection in the sl mutant.  相似文献

Nasunin, delphinidin-3-(p-coumaroylrutinoside)-5-glucoside, an antioxidant anthocyanin isolated from eggplant peels, was demonstrated as an angiogenesis inhibitor. Nasunin at higher 10 microM suppressed microvessel outgrowth in an ex vivo angiogenesis assay using a rat aortic ring. The effect of nasunin was examined in various in vitro angiogenesis models using human umbilical vein endothelial cells (HUVECs). Nasunin suppressed HUVEC proliferation in a dose-dependent manner (50-200 microM); however, it had no significant effect on HUVEC chemotaxis in a Boyden chamber assay and HUVEC tube formation on a reconstituted basement membrane. These results imply that nasunin with both antioxidant and antiangiogenic activities might be useful to prevent angiogenesis-related diseases.  相似文献

Seven of 92 lactating Holstein cows on a dairy farm developed urticaria with alopecia and decreased milk production, and three of the seven died over the course of 7 to 18 days. Pathologic examination of the three cows, including the two dead and one euthanized cow, revealed that the skin, liver, spleen, kidneys, heart, salivary glands, pancreas, adrenal glands, mammary glands, lymph nodes, and trigeminal ganglia had lymphocytic to lymphogranulomatous inflammation. Inflammation predominated by lymphocytic infiltration was prominent in the heart, pancreas, mammary glands, adrenal gland, and trigeminal ganglia. Severe granulomatous inflammation with multinucleated giant cells was present in the spleen and kidneys. These lesions and their distributions were most similar to those seen in suspected cases of citrus pulp toxicosis and hairy vetch toxicosis. The outbreak of this disease resolved with the elimination of Citrus pulp from the feed. Immunohistochemical detection of lymphocytes and macrophage markers confirmed dramatic hyperplasia of CD3-positive T lymphocytes in these lesions. This strongly suggested that a type 4 hypersensitivity reaction played a role in the development of the lesions.  相似文献

1. In order to introduce exogenous DNA into gonads of chick embryos, stage X blastoderms of freshly laid and unincubated eggs were transfected by lipofection and electroporation in vivo. 2. The introduced DNA, green fluorescence protein (GFP) gene, was efficiently expressed in the blastoderms incubated for 24 h (78.8%, 78/99). 3. The GFP gene was present in most of the embryonic bodies and extra-embryonic membranes died by d 10 of incubation, when analysed by polymerase chain reaction. On d 16 to 20 of incubation, the GFP gene was detected in 7.0 to 20.9% of embryos in the heart, liver, stomach and brain, but not in the sartorius muscle. For the gonads, the GFP gene was detected in 22.2% (6/27) of the testes and 6.3% (1/18) of the ovaries examined. 4. These results suggest that it is possible to introduce exogenous DNA into gonads of chick embryos by lipofection and electroporation of stage X blastoderms in vivo.  相似文献

Secretory IgA was isolated from feline bile, using ammonium sulphate precipitation, gel-filtration chromatography and affinity chromatography. It formed a precipitating line between anti-cat whole serum or anti-cat bile serum by immunoelectrophoresis and double diffusion. It consisted of three subunits that molecular weight were 80 kd, 62 kd and 27 kd by SDS-PAGE analysis. The 80 kd protein was equivalent to secretory component of human and other animals. Immunoglobulin levels were observed on bile collected continuously by gallbladder cannulation in growing cats. Although immunoglobulins were not detected in bile of feline fetus, after birth its levels increased gradually. Biliary IgA levels reached the adult levels earlier than that of their serum.  相似文献

We evaluated the postsurgical outcomes of cutaneous or subcutaneous mast cell tumors (MCTs, n=25) in 23 dogs, resected with lateral surgical margins proportional to the widest tumor diameter, including at least one facial plane. The tumor diameter range was 0.3–2.6 cm (median: 0.9 cm), and all were histologically diagnosed as Kiupel’s low-grade MCT. Resection was histologically complete in 20, close (deep margin) in three, and incomplete (deep margin) in two. No dogs developed local recurrence at the site of initial surgery during follow-up of 161–2,219 days (median: 976 days). These results suggest that resection of low-grade, relatively small MCTs with surgical margins proportional to the tumor diameter is a practical procedure with high success rates.  相似文献
