The selenium status of three different classes of goats ((i) female lactating, (ii) female non-lactating, and (iii) male goats) grazing semi-arid pasture in the southern part of the Punjab province, Pakistan and that of selenium concentration of soil and dietary sources, ingested by those animals were investigated during two different seasons of the year (winter and summer). Soil, forage, feed, water from the pasture and blood plasma, urine, faeces, and (if applicable) milk from these goats were collected fortnightly. The samples were analyzed for selenium concentrations. Soil selenium showed both seasonal and sampling periods effect on its concentration while forage selenium was affected only by the seasonal changes. No significant effect of seasons or fortnights on feed selenium level was observed. In fecal samples selenium concentration in lactating and non-lactating and plasma of male goats were affected by sampling periods. While fecal selenium in male goats showed significant effect on its concentration both seasonal and within fortnights. Severe deficient level of soil selenium during both seasons and marginal deficient level of forage selenium during summer were observed. Selenium concentrations in feed slightly exceeded the requirements of ruminants in feed during both seasons of the year. Plasma selenium concentrations in all goat classes were higher in winter than that in summer showing no seasonal or fortnight variation and its concentration was slightly lower in lactating goats as compared to other classes. On the bases of these results, it is concluded that overall selenium status of the goats based on plasma selenium concentration may be considered adequate mainly due to the mineral supplement provided all over the year, since soil and forage selenium concentrations were low to deficient. 相似文献

Accurate diagnosis of disease is the major step between the cause and cure of disease. An economical, reliable, and rapid diagnostic tool is fundamental for the management of udder health. The earlier the disease is identified, the less will be the damage; keeping this in mind, many efforts are being made to develop reliable diagnostic tools for use on farm. However, traditional gold standard methods including somatic cell count and microbial culturing are still in use. They are partially being replaced with polymerase chain reaction and sequencing-based tests. Nanotechnology and protein-based tests have also gained lot of attention and some of them are potential candidate of future diagnostic tests for bovine mastitis. Research laboratories are struggling to develop simple, economical, and user-friendly biosensor-based methods that can be performed on farm for rapid diagnosis. The combination of both genomic and proteomic approaches, together with further involvement of nanotheranostic technologies and other sensors, will assist in the quest of better mastitis diagnostic tools. 相似文献

Cauliflower (Brassica oleracea L. var. botrytis) is widely used as a vegetable in many countries; however, productivity is adversely affected in areas affected by drought. To investigate the role of exogenously applied ascorbic acid (AsA) in mitigating the negative effects of drought on cauliflower, a pot experiment was performed using two cultivars of cauliflower ‘Local’ and ‘S-78’. Seeds of both cultivars were soaked in 75 mg l–1, or in 150 mg l–1 AsA, or in water (control) for 16 h. Water-deficit stress suppressed plant growth, reduced leaf chlorophyll concentrations, relative water contents (RWC), shoot and root P and K+ ion concentrations, and total soluble protein concentrations, significantly. However, significant drought-induced increases were observed in relative membrane permeability (RMP), the accumulation of total phenolic compounds, leaf free proline, glycinebetaine (GB), endogenous AsA, and hydrogen peroxide concentrations, and in the activity of superoxide dismutase (SOD). Seed treatment with 75 or 150 mg l–1 AsA resulted in lower accumulations of H2O2, while increasing shoot and root fresh weights and dry weights, RWC, total phenolic compound, free proline, GB, and endogenous AsA concentrations, and the activities of SOD and catalase (CAT). No changes were observed in leaf chlorophyll concentrations or in peroxidase (POD) activities, RMP, shoot and root P and K+ ion accumulation, or in total soluble protein concentrations under water stress or non-stress conditions following seed treatment with AsA. ‘Local’ had higher proline concentrations and SOD activities; however, ‘S-78’ had higher RWC values, GB, and AsA concentrations. Overall, a pre-sowing treatment of cauliflower seed with 75 or 150 mg l–1 AsA improved seedling tolerance to drought stress in both cultivars, which could be attributed to AsA-induced decreases in RMP and H2O2 concentrations, increases in the activities of CAT and SOD, increased RWC, and higher total phenolic compound, proline, GB, and AsA concentrations. The exogenous application of AsA therefore offers an effective strategy to minimise the adverse effects of drought stress on vegetable crops, including cauliflower. 相似文献

Monoclonal antibody-based competitive ELISA (C-ELISA) has been used for the specific measurement of antibodies to peste des petits ruminants (PPR) viruses in sheep, goats, cattle and Buffalo. Serum samples from sheep (n = 232), goats (n = 428), cattle (n = 43), buffalo (n = 89) were tested. The animals had not been vaccinated against rinderpest or PPR. Findings suggested that the sero-positive cases were significantly higher in sheep (51.29%) than in goats (39.02%) (P = 0.002). The overall sero-prevalence of PPRV in small ruminants was 43.33%. The PPR antibodies seroprevalence was 67.42% in buffalo and 41.86% in cattle which was significantly higher in buffalo (P = 0.005). The overall sero-prevalence of PPRV in large ruminants was 59.09%. Cattle and buffalo sera showed a high prevalence of antibody against PPR virus which may explain the difficulty experienced in achieving high post-vaccination immunity levels against rinderpest. Because antibodies against PPR virus are both cross-neutralizing and cross-protective against rinderpest virus, further vaccination in the presence of antibodies against PPR virus may be a waste of national resources. It was also suggested that antibodies to PPR virus could prevent an immune response to the rinderpest vaccine. This paper presents serological evidence for the transmission of PPR virus from sheep and goats to cattle and buffalo and highlights the need to include PPR serology in the sero-monitoring programme to give a better indication of national herd immunity of sheep and goats against PPR. 相似文献

Influence of sudden changes in management program on physiological and behavioral parameters in hens
The present study was designed to test the effects of sudden changes of food access and light duration on the physiological and behavioral parameters of hens. The physiological parameters investigated were heart rate (HR), body temperature (BT), and locomotor activity (LA) using a radiotelemetry system. After implantation of the telemetry transmitters, six hens were housed individually in cages under constant environmental conditions for 10 days with a photoperiod of 15 h light (04.00–19.00 hours), and food was available ad libitum at all times. After that, the same hens were subjected to a feed withdrawal trial, from 12.00 to 08.30 hours, followed by a lighting hour reduction trial by changing the time of lights‐off from 19.00 to 14.00 hours. The physiological and behavioral data were recorded for 2 days before each trial, as control data. With the feed withdrawal trial, during the light and dark periods, HR and BT were significantly lower in the hens without food access than in the control. Whereas, LA was significantly lower only during the light period in the hens without food access than in the control. Further, the time spent resting increased significantly, but the time spent feeding decreased significantly in the hens without food access than in the control. Also, the number of times the cage was pecked and pecking of the feeder occured at a significantly higher level, while the number of times beaks were wiped occured at a significantly lower level in the hens without food access than in the control. With the lighting hour reduction trial, during the light period, HR and BT were significantly higher, whereas LA was significantly lower in the ’sudden light‐off’ treated hens than in the control. In addition, during the dark period, HR, BT and LA were significantly higher in the sudden light‐off treated hens than in the control. Moreover, in the sudden light‐off treated hens, the time spent preening and feeding decreased significantly, but the time spent resting increased significantly than in the control. It is concluded that sudden changes in a management program might result in many significant differences as were found in the physiological and behavioral parameters of hens in the present study. 相似文献

This study was conducted to isolate novel lactose utilizing Xanthomonas campestris mutants. Such a mutant will assist the utilization of whey as the sole carbon source for xanthan gum production, lower costs of fermentation process and set a precise application for whey as a waste. In this study, a mutant strain (NA1) was isolated from Xanthomonas campestris cells exposed to nitrous acid mutagenesis Environmental conditions were optimized and maximum activity of the beta-galactosidase enzyme was obtained at pH 5.5 and 38 degrees C following which the beta-galactosidase activity in NA1 culture was increased 9.5 folds, compared to that of the wild type culture (336.1 U vs. 35.4 U). Xanthan gum production by NA1 using whey as carbon source was also studied. Using the experimental design of Plackett-Burman and statistical analysis, whey, as the main substrate and pH were the first factors affecting gum production among the seven parameters tested. Gum production using significant factors (such as substrate concentration and pH) was carried out in a lab-scale fermentor and 10 g L(-l) xanthan was obtained. 相似文献

A new bacterial disease of Russell prairie gentian (Eustoma grandiflorum) was found in Fukuoka Prefecture, Japan, in 1997. This disease was characterized by wilting and yellowing of the foliage.
A cross section of the stem of a diseased plant revealed a tan to yellow-brown discoloration of the vascular tissue. A nonfluorescent,
aerobic, Gram-negative bacterium was consistently isolated from infected plants. The bacteriological characteristics of 10
isolates of the bacterium coincided with those of the reference strains of Burkholderia caryophylli that were isolated from carnations. The bacterium, as well as the reference strains, attacked Russell prairie gentian and
carnation after artificial inoculation and reproduced the symptoms similar to those after natural infections. On the basis
of bacteriological characteristics and pathogenicities, the bacterium was identified as B. caryophylli. This is the first report of a disease caused by B. caryophylli on Russell prairie gentian ; therefore, bacterial wilt of Russell prairie gentian is proposed as the name of the disease.

A total of 210 European seabass fry with an average body weight of 0.51 ± 0.03 g were divided into seven experimental treatments (three replicates per treatment). The seven experimental diets (45.64% CP) were formulated to contain synthetic astaxanthin (SA; 0.1 g kg?1), marigold flower meal (MFM; 0.2 g kg?1) or crab waste meal (CM; 10 g kg?1), with or without sodium taurocholate (ST; 0.4 g kg?1); the control diet contained no supplementation. The experiment was conducted for 8 weeks. The results show that seabass fry fed the diets containing different sources of carotenoids, especially with ST, demonstrated higher growth efficiency, survival (%), feed efficiency, protein and ether extract contents of fish body. The best results were found with DietMFM+ST. Whole‐body carotenoid levels increased significantly with all supplementation treatments; the highest level was observed with SA, followed by fish fed DietMFM and DietCM, especially with ST co‐supplementation. Moreover, thiobarbituric acid‐reactive substances were significantly reduced in fish fed the diets containing different sources of carotenoids compared to the control diet. The total antioxidant status significantly increased in fish fed DietSA and DietCM alone or with ST compared to the control group. In conclusion, these results reveal that DietMFM+ST is the most suitable treatment for improving growth, feed efficiency, and the protein and ether extract contents of seabass fry compared to other sources of carotenoids. The best oxidant/antioxidant balance and carotenoid content were achieved using SA, CM and MFM with ST supplementation. 相似文献
