Soybean has been considered a cold intolerant species; based largely upon seed germination and soil emergent evaluations. This study reports a distinct acquisition of cold tolerance, in seedlings, following short acclimation periods. Diversity in cold responses was assessed in eight cultivars of Glycine max and six accessions of G. soja. All varieties of soybean significantly increased in freezing tolerance following acclimation. This study indicates soybean seedlings are indeed capable of sensing cold and acquiring cold tolerance. Germination rates after cold imbibition were negatively correlated with maturity group, but positively correlated with cold acclimation potential in G. soja. Seed fatty acid composition was varied between the species, with Glycine soja accessions containing about 2‐times more linolenic acid (18:3) than G. max. Furthermore, high levels of linoleic acid (18:2) in seeds were positively correlated with germination rates following cold imbibition in G. soja only. We suggest that domestication has not impacted the overall ability of soybean to cold acclimate at the seedling stage and that there is little variation within the domesticated species for ability to cold acclimate. Bacterial wilt or brown rot is one of the most devastating diseases of potato caused by a bacterium Ralstonia solanacearum (Smith 1986) Yabuuchi et al. (Microbiol Immunol 39:897–904 1995). Traditionally, R. solanacearum is classified into five races (r) on the basis of differences in host range and six biovars (bvs) on the basis of biochemical properties. Recently using molecular methods, R.?solanacearum has been classified into phylotypes based on the intergenic transcribed sequence of the ribosomal RNA genes 16S and 23S and into sequevars based on the endoglucanase gene (egl) sequence. In the present study, 75 bacterial strains, isolated from wilt infected potatoes from various potato growing regions of India, were classified by traditional and molecular methods. The identity of all the strains was confirmed as R. solanacearum as expected single 280-bp fragment resulted in all the strains following PCR amplification using R. solanacearum specific universal primer pair 759/760. Biovar (bv) analysis, based on utilization of disaccharide sugars and hexose alcohols, categorised the 75 strains into bv2 (78.7 %), 2 T (5.3 %), 3 (5.3 %) and 4 (10.7 %). The phylotype specific multiplex PCR assigned 78.7 % strains to phylotype II, 16.0 % to phylotype I and 5.3 % to phylotype IV. Phylogenetic analysis of egl gene sequences clustered all fifty nine phylotype II (bv2) strains with reference strain IPO1609 (IIB-1), all four phylotype IV (bv2T) strains with reference strain MAFF301558 (IV-8), three phylotype I (bv3) strains with reference strain MAFF211479 (I-30) and all eight phylotype I (bv4) and one phylotype I (bv3) strain with reference strain CIP365 (I-45). The study concluded that the Indian potato strains of R. solanacearum belong to three out of four phylotypes namely: the Asian phylotype I, the American phylotype II, and the Indonesian phylotype IV. This is the first study to address the diversity of R. solanacearum from potato in India using phylotype and sequevar scheme. We also report here for the first time the occurrence of phylotype IV sequevar 8 (bv2T) strain of R. solanacearum causing potato bacterial wilt in mid hills of Meghalaya in India. 相似文献

Four Bt cotton hybrids, each with one of four different events, viz., MRC 6301 Bt (cry1Ac gene), JKCH 1947 Bt (modified cry1Ac gene), NCEH 6R Bt (fusion cry1Ac/cry1Ab gene) and MRC 7017 Bollgard II (cry1Ac and cry2Ab genes) were compared for survival and development of Earias vittella (Fabricius) along with their isogenic non-Bt genotypes. None of the neonates were able to complete the larval period and reach pupal stage on squares of 90, 120 and 150 days old crop of all Bt hybrids. Likewise, on bolls also, zero per cent larval survival was observed in all Bt hybrids except JKCH 1947 Bt where 0.67 per cent larvae could manage to reach pre-pupal stage at 120 and 150 days old crop but failed to form cocoon and enter pupal stage. The surviving larva took more development time (3.7 to 5.4 days) as compared to larvae fed on bolls of JKCH 1947 non-Bt. The average survival period (ASP) of larvae was in order of 150 > 120 > 90 days old crop among the crop ages; JKCH 1947 Bt > MRC 6301 Bt > NCEH 6 R Bt > MRC 7017 Bollgard II among Bt hybrids; and bolls > squares between fruiting bodies. However, reverse was true for speed index of toxic effect. The concentration of Cry toxin varied significantly in squares and bolls and also among the crop ages. The amount of Cry toxin in squares and bolls had significant negative correlation with ASP of the E. vittella larvae. 相似文献

1. The study investigated the extent of hyperpigmentation (a trait fixed in native Indian Kadaknath chickens), bodyweight, carcase quality and leanness at 12 weeks of age in F(1) and back-crosses of Kadaknath with White Leghorn, White Plymouth Rock and Aseel Peela chickens. 2. The objective of the study was to determine if hyperpigmentation was affected by the major gene Fibromelanosis (Fm) and to evaluate the effects of different proportions of Kadaknath genes on growth and carcase quality. 3. The pigmentation pattern of skin indicated that Fm behaved as the primary locus affecting dermal-hyperpigmentation and that the sex-linked Id locus produced an epistatic effect. 4. The results suggested that variable allelic forms of Id were acting in different crosses, which resulted in variation in melanosis of the host. However, no conclusive pattern for shank pigmentation could be explained through genotyping of the Id and Fm loci. 5. Analysis of quantitative traits indicated the positive impact of a Kadaknath genomic proportion of 50% or more on meat texture and carcase leanness. Improvement in leanness occurred in White Rock crosses but not in White Leghorn and Aseel Peela crosses. 6. Thigh-meat texture was influenced more by enhanced Kadaknath genomic proportions than the breast-meat. It was concluded that introgression of Kadaknath genomic proportion beyond 50% in a cross with meat-type chickens, irrespective of the impact Fm, brought improvement in meat quality whereas no such advantage was obtained for growth traits. 7. The beneficial impact of the Kadaknath genome on meat quality calls for further studies to identify causative genes for their selective use to improve meat quality in Kadaknath crossbred chickens. 相似文献

A captive adult female bottlenose dolphin presented with stillbirth. The placenta appeared oedematous. No other gross lesions were evident in the placenta or the stillborn calf. Histopathology revealed mild multifocal placentitis and foetal encephalitis. Brucella sp. was isolated from lung, liver, spleen and kidney. Sequence and phylogenetic analysis demonstrated this organism to be most similar to Brucella ceti sequence type (ST) 27. Brucella sp. DNA was detected in formalin-fixed paraffin-embedded placenta and brain by real-time PCR using primers targeting the IS711 gene. Immunohistochemical staining revealed Brucella sp. antigen in placental inflammation. This is the first report of isolation of Brucella sp. from a marine mammal in the Southern Hemisphere and the first report of marine Brucella-associated disease in Australia. 相似文献

Among the treatments of exogenous foliar spray application of sulphates of cobalt (Co) and cadmium (Cd) at 0, 250, 500, 1000, and 1500 ppm, and nickel (Ni) at 0, 100, 200, and 400 ppm prior to flower bud differentiation in the first week of October, cobalt sulphate (1000 ppm) or cadmium sulphate (1500 ppm) significantly (P=0.05) reduced floral malformation of mango (Mangifera indica L) cv. Langra by 65 to 71% and increased the fruit yield by 35 to 37%. The physical characteristics of the fruit were not affected significantly at the lower concentrations but higher concentrations of Co and Cd caused reduction. Total and non‐reducing sugars in the fruit were adversely affected by the higher rates of Co and Ni applied, but exhibited no effect on total soluble solids percentage (TSS%) or reducing sugars. The foliar spray application of the sulphates of Co, Ni, and Cd increased their respective concentrations in the leaf tissue without any phytotoxic symptoms. Most of the desirable effects were achieved with the 1000 ppm cobalt sulphate treatment applied in the first week of October prior to flower bud differentiation. 相似文献

The present investigation was carried on a 8 parent diallel of mungbean involving 28 F1's and 28 F2's excluding reciprocals grown at three different locations including Meerut, Ludhiana and Hissar. Three different analyses including combining ability, graphical and component analysis were utilized.From the combining ability analysis over different environments it was reported that g.c.a. effects are relatively more consistent over environments than s.c.a. effects. The parents P3 and P5 were ranked as the best general combiners and the cross P5 × P7 was the good specific combination in both the diallel sets. The graphical analysis suggested that high protein content is governed by dominant genes. Further all the analyses revealed that non-additive variance was mostly responsible for this character. Thus two methods, including biparental mating system among the selected F2/F3 plants from P5×P7 cross; and modified diallel selective mating system for the selected parents with good or average general combining ability were suggested. The study was conducted on eleven healthy non-anaemic adolescent girls of 16 to 18 years of age. Balance studies were conducted in two trials of three weeks each on low and high fibre diets. High fibre diet contained 25 g Isabgol husk in addition to low fibre diet. The mean diet and nutrient intakes of the subjects were approximately the same during both trials. Addition of Isabgol husk to low fibre diet significantly (P0.05) increased faecal excretion of zinc, copper and manganese and lowered their apparent retention. The serum levels of these trace minerals decreased significantly (P0.05). Thus the high level of Isabgol has undesirable effect on trace minerals. 相似文献
