The inheritance of distichiasis was analyzed in the dog breed Elo using complex segregation analyses.The different hypotheses of monogenic, mixed-monogenic-polygenic and polygenic inheritance as well as a pure environmental model were tested. In further analyses the number of examined Elo dogs per ophthalmologist and the proportion of genes of Eurasian and Old English Sheepdogs were included in the models as covariates. Seven families were randomly collected for the analyses.The seven families comprised a total of 296 animals with 218 examined and 65 affected dogs. A genetic component was verified for the prevalence of distichiasis. Mixed models with a recessive, dominant or arbitrary major gene effect and the polygenic model represented likely hypotheses of inheritance. The prevalence of distichiasis observed was insufficiently explained by non-genetic factors and the breed composition. Therefore, a model only including non-genetic factors and the breed composition could be excluded. 相似文献

OBJECTIVE: To determine the susceptibility of strains of Pasteurella multocida subsp multocida isolated from lung specimens of pigs with pneumonia to 20 antimicrobials and to evaluate the emergence of resistance to those antimicrobials in Spain during the past 2 decades. SAMPLE POPULATION: 63 isolates recovered from 1987 to 1988 and 132 isolates recovered from 2003 to 2004. PROCEDURE: A broth microdilution method was used to determine minimal inhibitory concentration (MIC) range and values for MIC50 and MIC90. Resistance of a strain to an antimicrobial agent was determined by use of the breakpoint value when available. RESULTS: Isolates were generally susceptible to penicillin, ampicillin, ceftiofur, gentamicin, apramycin, neomycin, spectinomycin, chlortetracycline, erythromycin, tilmicosin, enrofloxacin, and florfenicol, and most isolates were resistant to clindamycin, tylosin tartrate, and tiamulin regardless of the time period. A substantial increase in resistance to sulfa-chlorpiridazine, sulfadimethoxine, sulfathiazole, and trimethoprim-sulfamethoxazole was observed, and a minor increase in resistance to oxytetracycline was also detected. Several multiresistance patterns were observed, most frequently among isolates recovered in the 2003 to 2004 interval. CONCLUSIONS AND CLINICAL RELEVANCE: Ceftiofur, florfenicol, and enrofloxacin are recommended for treatment of infections caused by P multocida subsp multocida in Spain. OBJECTIVE: To determine whether inflammation of the jejunum of horses decreases the number of motilin receptors and amounts of motilin receptor mRNA and alters erythromycin lactobionate binding affinity to the motilin receptor in jejunal tissues. SAMPLE POPULATION: Jejunal segments in 6 adult horses. PROCEDURE: Each horse was anesthetized, and a ventral median celiotomy was performed; 2 segments of jejunum underwent a sham operation, 2 segments underwent ischemic strangulation obstruction (ISO), and 2 segments underwent intraluminal distension (ILD). Treatments were maintained for 120 minutes. From each segment, full-thickness biopsy samples were collected and smooth-muscle homogenates were prepared. Affinity and distribution of motilin binding to these preparations were determined by use of iodine 125 (125I)-labeled synthetic porcine motilin. Via displacement experiments, competition between 125I-labeled motilin and erythromycin lactobionate for binding to motilin receptors in the different segments was investigated. A quantitative real-time PCR technique was used to assess motilin receptor mRNA content in the muscle preparations. RESULTS: Compared with the ISO or ILD segments, the number of motilin receptors was significantly higher in the sham-operated segments; ILD segments contained the lowest number of motilin receptors. The expression of motilin receptor mRNA was significantly decreased in ILD segments but not in ISO segments. Erythromycin lactobionate displacement of 125I-labeled motilin from motilin receptors did not differ significantly among the jejunal segments. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that downregulation and decreased production of motilin receptors in inflamed jejunal tissue contribute to the altered prokinetic response to erythromycin in horses with gastrointestinal disease. 相似文献

OBJECTIVE: To evaluate mRNA expression of several proinflammatory and anti-inflammatory cytokines and chemokines in equine unstimulated and interleukin-1beta (IL-1beta)-stimulated chondrocytes. STUDY DESIGN: In vitro experiment using equine chondrocyte cultures. SAMPLE POPULATION: Whole articular cartilage from metacarpophalangeal joints (n=5 horses; 10 fetlocks). METHODS: Chondrocyte monolayer cultures were established from digested adult equine articular cartilage and stimulated with 5 ng/mL of recombinant human IL-1beta. RNA was extracted from the cells 24 hours after stimulation. IL-1beta, IL-4, IL-6, IL-8, tumor necrosis factor-alpha (TNF-alpha), and ubiquitin (house keeping gene) mRNA expression were investigated by real-time RT-PCR. RESULTS: IL-1beta, IL-6, and IL-8 mRNA were expressed in unstimulated chondrocytes from macroscopically normal joints and were significantly up-regulated after stimulation (5/5 horses). IL-4 mRNA was not detected in any samples (0/5 horses). TNF-alpha mRNA, by comparison, was expressed in 2/5 unstimulated samples and in all stimulated samples but a considerable sample variation in response to IL-1beta stimulation was observed. CONCLUSIONS: Equine chondrocytes express mRNA for several proinflammatory cytokines and chemokines and IL-1beta modulates their expression. CLINICAL RELEVANCE: Chondrocytes express proinflammatory cytokines and chemokines capable of modulating a local inflammatory cascade in articular cartilage, which could potentially lead to focal degradation and osteoarthritis. 相似文献

The objectives of this study were: the chemical characterization of extracts from seven plants (Larrea tridentata, Flourensia cernua, Lippia graveolens, Agave lechuguilla, Yucca filifera, Opuntia ficus-indica, and Carya illinoensis) which are acclimated to the Mexican semi-desert. The extracts were obtained using Soxhlet method by water, ethanol and an infusion method using alternative organic solvents (lanolin and cocoa butter), in addition it was evaluated the antibacterial activity of semi-desert plant extracts against Enterobacter aerogenes, Escherichia coli, Salmonella typhi and Staphylococcus aureus. Chemical characterization of plant extracts showed that they are rich in secondary metabolites; cocoa butter was the non-conventional solvent which it was possible to obtain the highest content of total tannins. It was not possible to identify saponins in those extracts where non conventional solvents were used. While in extract where non conventional solvents were used, it was only possible to detect the presence of terpenes in creosote bush and prickly pear extracts. S. aureus was the bacterial strain that showed the highest growth inhibition as consequence of the plant extracts. The use of semi-desert plant extracts obtained using organic solvents are a good alternative for food-borne pathogen bacteria control because all the bacterial growth decreased with the tested extracts. 相似文献

A defining feature of inflammation is the accumulation of innate immune cells in the tissue that are thought to be recruited from the blood. We reveal that a distinct process exists in which tissue macrophages undergo rapid in situ proliferation in order to increase population density. This inflammatory mechanism occurred during T helper 2 (T(H)2)-related pathologies under the control of the archetypal T(H)2 cytokine interleukin-4 (IL-4) and was a fundamental component of T(H)2 inflammation because exogenous IL-4 was sufficient to drive accumulation of tissue macrophages through self-renewal. Thus, expansion of innate cells necessary for pathogen control or wound repair can occur without recruitment of potentially tissue-destructive inflammatory cells. 相似文献
