Homegardens are traditional food systems that have been adapted over generations to fit local cultural and ecological conditions. They provide a year-round diversity of nutritious foods for smallholder farming communities in many regions of the tropics and subtropics. In southwestern Uganda, homegardens are the primary source of food, providing a diverse diet for rural marginalized poor. However, national agricultural development plans as well as economic and social pressures threaten the functioning of these homegardens. The implications of these threats are difficult to evaluate, because the structure and functions of the homegardens are not well understood. The aim of the study was to identify patterns and influencing factors in the diversity of homegardens by documenting the floristic diversity and its interactions with spatial, environmental and socio-economic factors. A geographically and socially focused assessment of floristic diversity in 102 randomly selected homegardens in three districts of southwest Uganda was conducted along a deforestation gradient following a human ecology conceptual framework and testing multiple quantitative hypotheses regarding the above mentioned factors. A merged mixed-method approach was followed to provide context and feedback regarding quantitative findings. Results show a high total richness of 209 (mean 26.8 per homegarden) crop species (excluding weeds and ornamentals) dominated by food species, which constituted 96 percent of individuals and 44 percent of all species. Forest-edge homegardens maintained higher plant diversity compared to homegardens in deforested areas and near degraded wetlands. Multiple linear regression models indicated elevation, location, homegarden size, distance to market, additional land ownership (outside the homegarden) and livestock ownership as significant predictors of crop diversity. Cluster analysis of species densities revealed four garden types: ‘diverse tree gardens’, ‘small forest-edge gardens’, ‘large, old, species-rich gardens’, and ‘large, annual-dominated herb gardens’, with 98% correct classification. Location, elevation, and garden size were also important determinants in the cluster assignment. We conclude that the diversity of the studied homegardens may be changing as part of adaptive traditional practices and in response to external drivers. The identified patterns illustrate the importance of homegardens for rural livelihoods and may offer some ways to support farmers to maintain these systems as relevant mechanisms for development in Uganda.  相似文献

Background

Plant biomass consists primarily of carbohydrates derived from photosynthesis. Monitoring the assimilation of carbon via the Calvin-Benson cycle and its subsequent utilisation is fundamental to understanding plant growth. The use of stable and radioactive carbon isotopes, supplied to plants as CO2, allows the measurement of fluxes through the intermediates of primary photosynthetic metabolism, long-distance transport of sugars in the vasculature, and the synthesis of structural and storage components.

Results

Here we describe the design of a system for supplying isotopically labelled CO2 to single leaves of Arabidopsis thaliana. We demonstrate that the system works well using short pulses of 14CO2 and that it can be used to produce robust qualitative and quantitative data about carbon export from source leaves to the sink tissues, such as the developing leaves and the roots. Time course experiments show the dynamics of carbon partitioning between storage as starch, local production of biomass, and export of carbon to sink tissues.

Conclusion

This isotope labelling method is relatively simple to establish and inexpensive to perform. Our use of 14CO2 helps establish the temporal and spatial allocation of assimilated carbon during plant growth, delivering data complementary to those obtained in recent studies using 13CO2 and MS-based metabolomics techniques. However, we emphasise that this labelling device could also be used effectively in combination with 13CO2 and MS-based techniques.

The production of new bioactive oligosaccharides is currently garnering much attention for their potential use as functional ingredients. This work addresses the enzymatic synthesis and NMR structural characterization of 2-α-D-glucopyranosyl-lactose derived from sucrose:lactose and sucrose:cheese whey permeate mixtures by using a Leuconostoc mesenteroides B-512F dextransucrase. The effect of synthesis conditions, including concentration of substrates, molar ratio of donor/acceptor, enzyme concentration, reaction time, and temperature, on the formation of transfer products is evaluated. Results indicated that cheese whey permeate is a suitable material for the synthesis of 2-α-D-glucopyranosyl-lactose, giving rise to yields around 50% (in weight respect to the initial amount of lactose) under the optimum reaction conditions. According to its structure, this trisaccharide is an excellent candidate for a new prebiotic ingredient, due to the reported high resistance of α-(1→2) linkages to the digestive enzymes in humans and animals, as well as to its potential selective stimulation of beneficial bacteria in the large intestine mainly attributed to the two linked glucose units located at the reducing end that reflects the disaccharide kojibiose (2-α-D-glucopyranosyl-D-glucose). These findings could contribute to broadening the use of important agricultural raw materials, such as sucrose or cheese whey permeates, as renewable substrates for enzymatic synthesis of oligosaccharides of nutritional interest.  相似文献

A new method has been developed and validated for the simultaneous determination of four biogenic (putrescine, cadaverine, histamine, and tyramine) and three volatile amines (trimethylamine, triethylamine, and tripropylamine) in anchovy. Separation and determination of the selected compounds were carried out by ultra-high-performance liquid chromatography coupled to tandem mass spectrometry (UHPLC-MS/MS), using an electrospray ionization source (ESI) in positive ion mode. Chromatographic separation was carried out using an aqueous solution of formic acid (0.1%) and methanol as mobile phase in gradient mode. The method was validated, and mean recoveries were evaluated at three concentration levels (75, 150, and 250 μg/kg), ranging from 70 to 110% at the three levels assayed. Intra- and interday precision, expressed as relative standard deviation (RSD), were lower than 15% and 20%, respectively. Limits of quantitation (LOQs) were 25 μg/kg for all cases, except for that of TMA, which was set at 60 μg/kg. The developed procedure was applied to determine the target compounds in anchovy samples stored during 7 days at 4 °C, observing the increasing in the concentration of these compounds at longer storage time.  相似文献

Olive fruits are seriously deteriorated by pre and postharvest damage due to the attack of insects, such as Bactrocera olaea, which strongly alters the quality of olives. Defence response in olive fruits injured both by pathogens and by mechanical damages has been associated with the enzyme β-glucosidase, which specifically hydrolyses oleuropein, producing highly reactive aldehyde molecules. In situ detection of ~-glucosidase activity in olive fruit tissues following injury, which simulates Bactrocera oleae punctures, is reported. The assay was performed in two cultivars showing different degrees of susceptibilities to fly infestation. In both cultivars, the histochemical assay for β-glucosidase showed that within 20 min after the injury, a strong ~-glucosidase activity could be observed in the damaged tissues. Thereafter a progressive enzyme inactivation occurred starting from tissues around the boundary of the injury with decrease of the enzyme activity and stopped after 3 h. Whereas the mass of active cells reached a distance of (300±50) μm from the edge of the injury. Biochemical analyses showed that in extracts of the injured fruit, β-glucosidase activity rapidly increased within 20 min from injury, thereafter decreasing and reaching values comparable with those in intact fruits. Following puncture, the oleuropein contents did not change significantly in the high susceptibility cultivar, whereas it rapidly decreased in the cultivar showing low susceptibility. The results strongly suggest that olive fruits susceptible towards fly infestation could be related to the ability of the oleuropein-degrading-β-glucosidase to produce the highly reactive molecules in the damaged tissues. As a consequence of puncture, high level of peroxidase activity was detected. This feature also suggested that this enzyme could play a key role in the defence response against insect injuries.  相似文献

Commercial embryo transfer (ET) has unprecedented productive and economic implications for the pig sector. However, pig ET has been considered utopian for decades mainly because of the requirements of surgical techniques for embryo collection and embryo deposition into recipients, alongside challenges to preserve embryos. This situation has drastically changed in the last decade since the current technology allows non‐surgical ET and short‐ and long‐term embryo preservation. Here, we provide a brief review of the improvements in porcine ET achieved by our laboratory in the past 20 years. This review includes several aspects of non‐surgical ET technology and different issues affecting ET programmes and embryo preservation systems. The future perspectives of ET technology are also considered. We will refer only to embryos produced in vivo since they are the only type of embryos with possible short‐term use in pig production.  相似文献

A domestic shorthair kitten was presented for evaluation and further treatment of seizures. Magnetic resonance imaging of the brain revealed a large multilobulated mass in the third ventricle extending into the right lateral ventricle with secondary obstructive hydrocephalus. The mass was homogeneously isointense to gray matter on T2W, T2‐FLAIR, T2*W, T1W, and ADC images, and hyperintense on DW‐EPI. There was no appreciable contrast enhancement. Seizures were managed medically and with subsequent ventriculoperitoneal shunt placement. Clinical status later deteriorated and the cat was euthanized. Histopathology confirmed that the mass was the result of neuronal heterotopia. To the authors’ knowledge this is the first report of neuronal heterotopia in a cat.  相似文献

Surface active cellulose particles have been prepared for use as foam stabilizing agents in foods. Various sources of cellulose were broken down by combinations of milling, acid dissolution and treatment with cellulase. The most efficient and simple method was hammer and freezer milling of dry crystalline α-cellulose (Tencel). The resultant Tencel particles were made partially hydrophobic through precipitation of ethyl cellulose (EC) onto them in acetone-water dispersions. The optimum ratio of EC to cellulose and the optimum solids concentration (C(x)) at which to form the complexes were 1:1 and C(x) ≈ 1 wt %, respectively. Complexes combined at low concentrations (e.g., C(x) ≈ 0.1 wt %) with caseins or whey proteins gave significant improvements in stability of foams and bubbles to coalescence and disproportionation compared to either component alone. As such, the complexes could be a useful ingredient in improving the quality of various food foams.  相似文献

A rapid, one‐step agglutination assay has been developed, based on latex particles sensitized with antibodies against vitellogenin (Vtg), aimed at Atlantic bluefin tuna, Thunnus thynnus (Linnaeus) (ABFT), gender identification. The egg‐yolk precursor protein Vtg was used as a gender marker for the assay as it is a female‐specific protein synthesized during reproductive maturation. The presence of Vtg in the plasma was revealed in 60–120 s through an agglutination reaction by mixing small volumes of ABFT plasma and an anti‐Vtg antibody‐latex suspension on a microscope slide. The effectiveness of the present test was restricted to the months of May and June, concomitant with high circulating Vtg levels. Because of its rapidity and ease of performance in the field, the present gender identification assay could be useful for broodstock management in the aquaculture industry as well as in tagging studies on wild populations.  相似文献
