Aeromonas salmonicida was isolated from ulcerations in common dab (Limanda limanda). An experiment was performed to pinpoint its role in ulceration development, considering the importance of the skin barrier and the pigmented and non-pigmented sides. The skin of dab was treated in three zones, one where scales and epidermis were removed, one where mucus was discarded and one non-treated zone. Fish were tagged to allow individual identification and challenged with A. salmonicida. Mortality and severity of the developing lesions were recorded for 21 days post-inoculation. Starting 12 days post-inoculation, mortality occurred gradually in challenged fish; however, no direct cause could be established. Both control fish and challenged fish developed ulcerations containing A. salmonicida. Sequencing of vapA gene revealed that isolates retrieved from both groups were distinct, suggesting the presence of A. salmonicida prior to the trial. Most ulcerations developed in zones where skin was removed, suggesting that abrasion might be a predisposing factor in ulceration development. Ulcerations were also observed at the insertion site of the tag, where exposed muscle tissue might have favoured the development of ulcerations. In conclusion, A. salmonicida seems to be involved in the development of skin ulcerations in dab, although the exact pathogenesis needs to be elucidated.  相似文献

Equine viral arteritis (EVA) is an infectious disease with variable clinical outcome. Outbreaks, causing important economic losses, are becoming more frequent. Currently, there is a shortage of pathogenesis studies performed with European strains. In the present study, eight seronegative ponies were experimentally inoculated with the Belgian strain of equine arteritis virus (EAV) 08P178 (EU-1 clade) and monitored daily for clinical signs of EVA. Nasopharyngeal swabs, ocular swabs, bronchoalveolar cells and blood were collected for virological and serological testing. Two ponies were euthanized at 3, 7, 14, and 28 days post infection (DPI). After necropsy, specimens were collected for virus titration and immunofluorescence. EVA symptoms such as fever and lymphadenomegaly were evident from 3 to 10 DPI. Virus was isolated in nasal secretions from 2 to 9 DPI and in bronchoalveolar cells from 3 to 7 DPI. A cell-associated viraemia was detected from 3 to 10 DPI. After replication in the respiratory tract and draining lymph nodes, EAV reached secondary target organs (high virus titers in internal organs sampled at 7 DPI). At 14 DPI, virus titers dropped drastically and, at 28 DPI, only tonsils were positive. Immunofluorescence revealed both individual and clustered EAV-infected cells. Antibodies were detected starting from 7 DPI. It can be concluded that the Belgian strain 08P178 is a European mildly virulent subtype. At present, most European EAV strain infections were thought to run a subclinical course. This study is a proof that mildly virulent European EAV strains do exist in the field.  相似文献

Southeastern Tanzania serves as a typical example of soil degradation and soil organic carbon (SOC) losses on the African continent. Although sequestration of SOC through aforestation or reforestation proved favorable, these measures are restricted by the ability to produce rapid, cost-effective and precise sampling schemes. The aim of this study is to contribute to a better knowledge of the spatial distribution of soil C in tropical natural and plantation forest. This paper presents sampling strategies for estimating mean SOC values as well as for SOC mapping, based on different methods for SOC determination and on different precision levels. To do so we conducted a carbon variability study in five common forest types of Southeastern Tanzania (coastal dry forest, Miombo woodland, teak plantation, pine plantation and cashew plantation) using conventional statistical methods, as well as geostatistics. In the 5 forest types of this study, SOC stocks in the upper 5 cm ranges between 5 (in the cashew plantation) and 13 (in the coastal forest) t ha− 1. The optimal sampling distance for measuring mean SOC stocks varies between 36 m (in the patchy miombo woodland) and 422 m (in the homogenized cashew plantation). Sample sizes fluctuate between 6 and 72 (1 t ha− 1 precision) for respectively cashew plantation and coastal forest. A rectangular grid with a sample interval of 25 m can be used for SOC mapping with a point kriging estimation error of 3.0 t ha− 1 in the coastal forest, 2.6 t ha− 1 in miombo woodland, 2.2 t ha− 1 in the teak plantation and 1.1 t ha− 1 in the cashew plantation. Since the pine plantation has no spatial structure; samples can be arranged randomly and its best soil map has an average C content attributed over the whole field. Refining the sampling strategy with a new spatial variability study in other forest types can be based on a regular grid with sampling distances of half the range identified in this study. This paper proves that the optimal sampling scheme varies strongly as a result of the different spatial behavior of SOC in forests and depends on the required precision and research question. Only when the right strategy is followed, high standards of precision can be met without economic loss or risk of statistical misinterpretation.  相似文献

In Patagonia, arid and semiarid lands are being affected both by inappropriate management practices, which are leading to degradation, and by volcanic activity, whose effects are still unclear. This study aimed to test whether superficial deposition of volcanic tephra could benefit two of the most prominent Patagonian forage grass species (Poa ligularis and Pappostipa speciosa var. speciosa). Pots with P. ligularis and P. speciosa were kept under wet (W) and dry (D) conditions in the presence (T+) or absence (T?) of tephra for 105 days, and then were all well-watered. We determined the effects of tephra on soil water retention and conservation, soil moisture content (% v/v), plant growth, stomatal conductance (gs), and gs recovery capacity. The water regime significantly affected both species performance and gs. The presence of tephra increased soil water conservation, soil moisture content in wet conditions, and P. ligularis gs in wet conditions, and decreased senescence in dry conditions (9% in P. ligularis and 16% in P. speciosa). The presence of tephra allowed roots to grow in 8/10 and 2/10 pots in W conditions for P. ligularis and P. speciosa, respectively, and in only 1/10 pots in D conditions, only for P. ligularis. Tephra was also associated with gs recovery after dry conditions. Poa ligularis was more positively affected by tephra than P. speciosa, probably because P. ligularis has higher phenotypic plasticity. The positive effects of tephra may increase the resilience and resistance of P. speciosa and P. ligularis to periods of water shortage.  相似文献

The equine blood oxyhemoglobin dissociation curve has been traced in its entirety in standard conditions and the effects of temperature, pH and 2,3-diphosphoglycerate on this curve have been measured. When compared to that of human blood, the curve showed a higher oxygen affinity of hemoglobin (23.8 +/- 0.8 versus 26.6 mm Hg). The effect of the pH, expressed by d log P50/dpH, was found to be identical in man and horse (-0.47). The effect of temperature, however, expressed by d log P50/dT, proved to be lower in the horse (0.016 versus 0.024). The P50 showed an increase of 1 mm Hg each time 2,3-diphosphoglycerate was experiencing an elevation of 4 mumol/gHb in the horse. The results obtained from this experiment are intended for the replacement of the values related to the human blood which have been found to be somewhat inadequate for the horse blood, when the data of blood gases are given in algorithms.  相似文献

The fate of 4-chlorophenylurea in soils was studied with two preparations: one labelled with 14C in the phenyl ring and the other in the carbonyl group. The initial dose of 1 mg kg?1 decreased to 50% in about 5 weeks in aerobic sandy clay and in about 16 weeks in anaerobic hydrosoil. Soil treatment with each of the preparations resulted in the release of [14C]carbon dioxide, pointing to decarbonylation and ring opening. The fraction of non-extractable (soil-bound) radioactivity increased during incubation. Quantities of ring-14C-labelled and carbonyl-14C-labelled bound residues differed strongly in the aerobic soil but only slightly in the anaerobic hydrosoil. It is assumed that two sorts of bound residues are formed from 4-chlorophenylurea: one is fairly stable and might consist of bound 4-chloroaniline or its transformation products, whereas the other is presumed to be a degradable derivative of 4-chlorophenylurea.  相似文献

Candida isolates (n=23) derived from Testudinidae were investigated by multiplex-polymerase chain reaction (PCR). The isolates comprised 13 Candida (C.) tropicalis, nine C. albicans and one C. parapsilosis. In addition, five reference strains C. albicans (ATCC 10231), C. tropicalis (DSM 4238), C. parapsilosis (DSM 4237), C. glabrata (ATCC 70614) and C. krusei (ATCC 70075) were investigated. PCR revealed easily distinguishable species-specific amplicons of the chs1-gene and resulted in a clear identification in all cases. No discrepancies between conventional identification techniques and the PCR-based system were seen. The described PCR offers a rapid alternative to conventional techniques for the identification of C. albicans, C. glabrata, C. tropicalis, C. parapsilosis and C. krusei.  相似文献
