The Mars Exploration Rover Spirit has traversed a fairly flat, rock-strewn terrain whose surface is shaped primarily by impact events, although some of the landscape has been altered by eolian processes. Impacts ejected basaltic rocks that probably were part of locally formed lava flows from at least 10 meters depth. Some rocks have been textured and/or partially buried by windblown sediments less than 2 millimeters in diameter that concentrate within shallow, partially filled, circular impact depressions referred to as hollows. The terrain traversed during the 90-sol (martian solar day) nominal mission shows no evidence for an ancient lake in Gusev crater.  相似文献

Intraperitoneal administration of ICSM18 and 35, monoclonal antibodies against prion protein (PrP), has been shown to significantly delay the onset of prion disease in mice, and humanized versions are candidate therapeutics for prion and Alzheimer's diseases. However, a previous report of severe and widespread apoptosis after intracerebral injection of anti-PrP monoclonal antibodies raised concerns about such therapy and led to an influential model of prion neurotoxicity via cross-linking of cell surface PrP by disease-related PrP aggregates. In extensive studies including ICSM18 and 35, fully humanized ICSM18, and the previously reported proapoptotic antibodies, we found no evidence of apoptosis, thereby questioning this model of prion neurotoxicity.  相似文献

Low-molecular-weight fucoidan (LMWF) is a sulfated polysaccharide extracted from brown seaweed that presents antithrombotic and pro-angiogenic properties. However, its mechanism of action is not well-characterized. Here, we studied the effects of LMWF on cell signaling and whole genome expression in human umbilical vein endothelial cells and endothelial colony forming cells. We observed that LMWF and vascular endothelial growth factor had synergistic effects on cell signaling, and more interestingly that LMWF by itself, in the absence of other growth factors, was able to trigger the activation of the PI3K/AKT pathway, which plays a crucial role in angiogenesis and vasculogenesis. We also observed that the effects of LMWF on cell migration were PI3K/AKT-dependent and that LMWF modulated the expression of genes involved at different levels of the neovessel formation process, such as cell migration and cytoskeleton organization, cell mobilization and homing. This provides a better understanding of LMWF’s mechanism of action and confirms that it could be an interesting therapeutic approach for vascular repair.  相似文献

The Afrotropical mosquito Anopheles gambiae sensu stricto, a major vector of malaria, is currently undergoing speciation into the M and S molecular forms. These forms have diverged in larval ecology and reproductive behavior through unknown genetic mechanisms, despite considerable levels of hybridization. Previous genome-wide scans using gene-based microarrays uncovered divergence between M and S that was largely confined to gene-poor pericentromeric regions, prompting a speciation-with-ongoing-gene-flow model that implicated only about 3% of the genome near centromeres in the speciation process. Here, based on the complete M and S genome sequences, we report widespread and heterogeneous genomic divergence inconsistent with appreciable levels of interform gene flow, suggesting a more advanced speciation process and greater challenges to identify genes critical to initiating that process.  相似文献

The effect of feed intake level (.6, 1.0, and 1.6 x maintenance energy and protein requirements, M) on splanchnic (portal-drained viscera [PDV] plus liver) metabolism was evaluated in six multicatheterized beef steers (398 +/- 27 kg), using a double 3 x 3 Latin square design. On the last day of each 21-d experimental period, six hourly blood samples were collected from arterial, portal, and hepatic vessels. Due to catheter patency, PDV fluxes were measured on five steers, and liver and splanchnic fluxes on four steers. Increasing intake elevated (P < .01) splanchnic release of total (T) amino acids (AA), through increases (P < .01) in PDV release of both essential (E) and nonessential (NE) AA, in spite of a tendency (P < .20) for increased liver removal of NEAA. The PDV release of AA N represented 27 and 51% of digested N for 1.0 and 1.6 x M, respectively. At 1.0 and 1.6 x M, the liver removed 34% of total AA released by the PDV. For individual AA, portal flux of most EAA increased (P < .05) with feed intake, and the increase (P < .10) in splanchnic flux was accompanied by increased arterial concentration for all EAA except histidine, lysine, and methionine. This suggests that these might be limiting AA for this diet. On a net basis, most individual NEAA were released by the PDV except glutamate and glutamine, which were removed by the digestive tract. There was a net removal of NEAA by the liver, except for aspartate and especially glutamate, which were released. Ammonia release by the PDV tended (P < .20) to increase with intake and represented 69, 53, and 45% of digested N at .6, 1.0, and 1.6 x M, respectively. Urea removed by the PDV, unaffected by intake, represented 32, 33, and 21% of the digested N. Arterial glucose concentration increased linearly (P < .01) with greater intake, whereas net liver and splanchnic glucose release increased in a quadratic (P < .05) manner. Net PDV glucose release represented 26% of net glucose hepatic release at 1.6 x M. Intake elevated (P < .10) both insulin and glucagon arterial concentrations, resulting from a larger increment of portal release (P < .01) than hepatic removal (P < .05). Intake-based variations in IGF-I and NEFA arterial concentrations (P < .05) were not related to changes in splanchnic metabolism. These results clearly show the crucial role of the splanchnic tissues in regulating the profile and quantity of AA and concentrations of glucose and pancreatic hormones reaching peripheral tissues.  相似文献

Background

One of the challenges faced by equine breeders is ensuring delivery of good quality semen doses for artificial insemination when the mare is due to ovulate. Single Layer Centrifugation (SLC) has been shown to select morphologically normal spermatozoa with intact chromatin and good progressive motility from the rest of the ejaculate, and to prolong the life of these selected spermatozoa in vitro. The objective of the present study was a proof of concept, to determine whether fertilizing ability was retained in SLC-selected spermatozoa during prolonged storage.

Findings

Sixteen mares were inseminated with SLC-selected sperm doses that had been cooled and stored at 6°C for 48 h, 72 h or 96 h. Embryos were identified in 11 mares by ultrasound examination 16–18 days after presumed ovulation.

Conclusion

SLC-selected stallion spermatozoa stored for up to 96 h are capable of fertilization.  相似文献

Previous studies in lowland tropical rainforests of French Guiana showed that, among non-N2-fixing trees, two groups of late successional species contrasting in their leaf 15N natural abundance coexist, suggesting two different main ways of nitrogen acquisition. Two abundant late-successional species typically co-occurring in rainforests in French Guiana, namely Eperua falcata and Dicorynia guianensis, were chosen as representative of each group. Stable isotope techniques and measurements of potentials of microbial N transformation were performed to assess to what extent leaf 15N natural abundance of these species could be related to (i) δ15N signatures of soil mineral N sources and (ii) the capacity of soil to express nitrification and denitrification (both processes being directly involved in the balance between NH4+ and NO3). Soil δ15N-NH4+ was roughly similar to leaf δ15N of D. guianensis (around 3.5‰), suggesting a preferential use of NH4+, whereas in E. falcata, leaf δ15N values were closer to root δ15N-NO3? values (0.2 and ?2.0‰, respectively), suggesting a preferential use of NO3?. These differences in N source utilization were not accompanied by differences in availability in soil NO3? or in intensity of microbial functions responsible for soil N mineral evolution. However, (i) under both tree species, these functions showed clear spatial partitioning, with denitrification occurring potentially in soil and nitrification in the litter layer, and (ii) E. falcata fine roots colonized the litter layer much more strongly than D. guianensis fine roots. This strongly suggests that (i) the contrasted leaf δ15N values found in the two late-successional species reveal distinct N acquisition strategies and (ii) the ability of roots to predominantly exploit the litter layer (E. falcata) or the soil (D. guianensis) may constitute an important explanation of the observed differences. A complementarity between tree species, based on mineral N resource partitioning (itself resulting from a spatially structured location of the microbial functions responsible for the balance between NH4+ and NO3), can thus be supposed.  相似文献

Summary Genetic analysis of resistance to PVY in androgenetic doubled haploid lines, F1, F2 and backcross progenies of the Mexican pepper line, CM 334 (Capsicum annuum L.), was performed. Three reaction types were observed when seedlings were inoculated with several PVY strains of different pathotypes and with an American PeMV strain. Resistant genotypes never showed systemic symptoms although some individuals sporadically developed necrotic local lesions on inoculated cotyledons. Susceptible genotypes exhibited either a typical systemic mosaic or a systemic necrosis that caused the death of the inoculated seedlings. Segregation analyses indicated that resistance to pepper potyviruses in CM 334 is conferred by two genes. The first one, tentatively named Pr4, is dominant and confers the resistance to all now known pathotypes of PVY and to PeMV. The second one, tentatively named pr5, is recessive; it confers only the resistance to common strains of PVY. The systemic necrotic response is conferred by an independent dominant gene, tentatively named Pn1.  相似文献

A postcolumn derivatization method is described for determination of reducing sugars and phosphorylated reducing sugars from chicken meat and other foods using high-performance liquid chromatography (HPLC). Reducing sugars are extracted with ethanol/water, separated on a Kromasil amine-bonded column by isocratic analysis using acetonitrile/water as the mobile phase, and, after postcolumn reaction with tetrazolium blue, are determined by the resulting absorbance at 550 nm. Phosphorylated sugars are first dephosphorylated using alkaline phosphatase and then determined by the same method.  相似文献

Tiltmeters on the Arctic Ocean were used to measure flexure of the ice forced by an energetic packet of internal waves riding the crest of diurnal internal bores emanating from the Yermak Plateau, north of the Svalbard Archipelago. The waves forced an oscillatory excursion of 36 microradians in tilt of the ice, corresponding to an excursion of 16 micrometers per second in vertical velocity at the surface and of 3.5 millimeters in surface displacement. Strainmeters embedded in the ice measured an excursion of 3 x 10(-7) in strain, consistent with ice flexure rather than compression. The measured tilt is consistent with direct measurements of excursions in horizontal current near the surface (12 centimeters per second) and in vertical displacement (36 meters) of the pycnocline 100 meters below the surface.  相似文献
