ObjectiveTo establish a safe, reliable and reversible immobilization protocol for captive juvenile crocodiles.Study designProspective, randomized, clinical study.AnimalsThirty male estuarine crocodiles (body mass 1–12.1 kg) and 10 male Australian freshwater crocodiles (body mass 4.1–12.8 kg).MethodsAn optimized dose of medetomidine (0.5 mg kg?1) was administered intramuscularly (IM) into the tail (Group 1; n = 5), pelvic limb (Group 2; n = 5) and thoracic limb (Groups 3 and 4; n = 5 in each group) of estuarine crocodiles weighing 3–12.1 kg. Their heart and respiratory rates and degree of immobilization were monitored every 15 minutes until recovery and daily thereafter for 3 subsequent days. In Group 4 (n = 5), medetomidine was antagonized with an optimized dose of atipamezole (2.5 mg kg?1) given IM into the thoracic limb and time to recovery recorded. The effects of increasing doses of medetomidine given IM in the thoracic limb (n = 4) and intravenously (n = 6) were determined in 1–2 kg estuarine crocodiles. Australian freshwater crocodiles (4.1–12.8 kg) were administered medetomidine IM into the thoracic limb in divided doses at 0.5 mg kg?1 (n = 5) and 0.75 mg kg?1 (n = 5) and similarly monitored.ResultsImmobilization was achieved only in the estuarine crocodiles >3 kg and when medetomidine was administered into the thoracic limb. Immobilization was achieved within 30 minutes and the duration of immobilization lasted approximately 90 minutes. Immobilization in estuarine crocodiles was readily reversed with atipamezole. A dose of 0.75 mg kg?1 was required to immobilize Australian freshwater crocodiles and the onset of immobilization was longer and the duration shorter than seen in the estuarine crocodiles. The heart and respiratory rates of all immobilized animals decreased significantly and arterial blood pressure became undetectable in the animals in which it was measured.Conclusions and clinical relevanceMedetomidine administered in the thoracic limb of captive estuarine and Australian freshwater crocodiles, ranging from 3 to 12.8 kg, provides a predictable onset and duration of immobilization sufficient for physical examination, sample collection, short minor procedures and translocation of the animals. Atipamezole administered in the thoracic limb results in complete reversal of the effects of medetomidine in the estuarine crocodile and a rapid return to normal behaviour. 相似文献

Purpose
At the global scale, soil contamination with persistent metals such as lead (Pb), zinc (Zn), and copper (Cu) induces a serious threat of entering the human food chain. In the recent past, different natural and synthetic compounds have been used to immobilize metals in soil environments. However, the mechanisms involved in amendment-induced immobilization of metals in soil remained unclear. The objective of the present work was therefore to determine the mechanisms involved in metal-phosphates formation in the rhizospheric soils of pea and tomato currently cultivated in kitchen gardens.Materials and methods
Pea and tomato were cultivated on a soil polluted by past industrial activities with Pb and Zn under two kinds of phosphate (P) amendments: (1) solid hydroxyapatite and (2) KH2PO4. The nature and quantities of metal-P formed in the rhizospheric soils were studied by using the selective chemical extractions and employing the combination of X-ray fluorescence micro-spectroscopy, scanning electron microscopy, and electron microprobe methods. Moreover, the influence of soil pH and organic acids excreted by plant roots on metal-P complexes formation was studied.Results and discussion
Our results demonstrated that P amendments have no effect on metal-P complex formation in the absence of plants. But, in the presence of plants, P amendments cause Pb and Zn immobilization by forming metal-P complexes. Higher amounts of metal-P were formed in the pea rhizosphere compared to the tomato rhizosphere and in the case of soluble P compared to the solid amendment. The increase in soil-metal contact time enhanced metal-P formation.Conclusions
The different forms of metal-P formed for the different plants under two kinds of P amendments indicate that several mechanisms are involved in metal immobilization. Metal-P complex formation in the contaminated soil depends on the type of P amendment added, duration of soil-plant contact, type of plant species, and excretion of organic acids by the plant roots in the rhizosphere. 相似文献43.

Payments for Ecosystem Services (PES) is a powerful economic tool that gives positive conditional incentives for the provision of additional ecosystem services over the status quo, which has been used widely in terrestrial conservation. Interest in the concept of marine PES has recently emerged, but the fluid, transboundary and often common pool nature of marine ecosystems presents challenges for PES design and implementation. Here, we consider the potential role of PES in addressing current gaps in fisheries management. Used in combination with conventional regulatory approaches, PES may increase private sector engagement and generate more sustainable financing for fisheries management whilst spreading accountability throughout the supply chain. The approach is most likely to be feasible and effective in commercially valuable fisheries with: (i) demand for one or more ecosystem service and a threat to supply; (ii) suitable baseline data available and potential management actions underpinned by robust science; (iii) clarity and security of property rights; (iv) capacity for hybrid multi‐level governance; (v) capacity for rigorous monitoring, control and surveillance; and (vi) potential for financial sustainability of the scheme. An examination of four contrasting fisheries – Namibian hake, Mozambican shallow‐water shrimp, Western and Central Pacific skipjack tuna and Bangladesh hilsa – demonstrates that a developing world fishery will rarely fulfil each of these preconditions a priori, but that the potential for successful application of PES still exists. In practice, PES design will depend on the institutional context and require creative and innovative approaches to the maintenance of conditionality and additionality. 相似文献

Infection by Oestrus ovis is common in Lacaune dairy ewes of Roquefort cheese area (Aveyron, France). It is believed by local breeders that there is a close relationship between nasal myiasis and the incidence of enzootic nasal tumour. In order to check these anecdotal reports, a serological survey was done on 658 breeding ewes before turn-out and 897 breeding and primiparous (hoggets) ewes at the end of the grazing season. By the time of sampling, it was clear whether the sheep were infected at the end of the winter or had been re-infected over summer.In April and September, 40.7 and 26.3%, respectively, were free of O. ovis infection, indicating that the autumn treatment was not completely effective and that O. ovis adult flies were circulating during the summer in many flocks. There were no differences in the incidence of adenocarcinoma between the groups indicating that there is no relationship between O. ovis infection and the presence of the cancer.Differences in milk production between the three groups were not statistically significant (Anova test P>0.05). In flocks where 1-5% of the ewes were infected or in non-infected flocks, ewes produced 3.6 and 8.56%, respectively, more milk than ewes from flocks where more than 5% of animals were infected. The purpose of this experiment was to determine if an earlier infection with Oestrus ovis would down regulate an infection with Trichostrongylus colubriformis when the larvae of O. ovis were expelled from the nasal cavities of sheep by a specific treatment. Three groups of five lambs were used: group 1 was artificially infected with O. ovis larvae and later with T. colubriformis, group 2 received O. ovis larvae and later was treated with ivermectin 14 days before being infected with T. colubriformis. Group 3 was infected with T. colubriformis only. The criteria examined were: the effects on nematode egg excretion, worm fecundity, nematode burdens and the kinetics of blood eosinophils. Significant decreases of nematode egg excretion, worm fecundity, nematode burdens were observed in group 1 compared to group 3. However, no changes were observed in either group 2 or 3. In group 2 it was noted that antiparasitic treatment induced a rapid decrease in blood eosinophils to a range close to the non-infected control group and this was associated with the removal of the down regulation effects of nematode burdens. This experiment showed that there is no cross immunity between O. ovis and T. colubriformis and that eosinophils may act against any parasite without specific priming. 相似文献

Seriously escalating global anthelmintic resistance in gastrointestinal nematodes of small ruminants has spawned a variety of alternatives to anthelmintics for worm management, based on the need for sustainable Integrated Parasite Management (sIPM). Pivotal to the sIPM approach is the concept of refugia, the proportion of a given parasite population that escapes exposure to control measures. By balancing drug applications with the maintenance of refugia, the accumulation of anthelmintic resistance alleles in worm populations can be considerably delayed, while still providing good levels of control. The over-dispersed nature of parasitic infections provides an opportunity to achieve this balance, by targeting treatments to the members of a flock or herd that are least tolerant to nematode infection. However, implementation of this strategy has only recently become feasible, with the development of the FAMACHA((c)) system for clinical evaluation of anaemia due to haemonchosis. Subsequently, the use of milk yields has proven an effective indicator in dairy goats infected predominantly with nematodes other than Haemonchus contortus. In addition, short-term weight changes and perhaps also body condition scoring may provide indices of parasitism, permitting the rapid identification of animals likely to benefit from treatment. However, sIPM and refugia-based approaches are more complex than whole-flock treatments in conventional programs, and adoption by farmers is most likely where the theoretical basis is understood. As close communication with informed advisors is generally limited, there is a danger that sIPM will remain a theoretical concept without alternative modes of communication. The development of computer-based decision support programs, which use epidemiological, seasonal and clinical information to provide recommendations for specific situations, should be accorded high priority in the future development of worm management systems. 相似文献

Numerous studies have examined the interactions between protein nutrition and the response to nematode parasitism in sheep, but very few in goats. Compared with other ruminants, goats are less resistant to nematode infection. In addition, in dairy goats, high producing animals have been shown to be less resistant and less resilient to infection compared to low producing ones. The objective of the present study was to examine the consequences of protein supplementation on both resistance and resilience of dairy goats to nematode trickle infection, taking into account the initial level of milk production of the animals. During a 14-week period, 40 milking goats received a high protein (HP) diet supplying 130% of the protein requirements, and 38 goats were fed a intermediate protein (IP) diet (120% of the protein requirements). In addition, half of each group was given a weekly trickle infection with Trichostrongylus colubriformis larvae, the other part of the flock remained non-infected. Faecal egg counts (FEC), eosinophil counts and pathophysiological data (urea, albumin and inorganic phosphate concentrations in the serum) were measured twice a month. Milk production data (milk yield, protein and fat contents) were also recorded every 15 days. The results showed that FECs were lower (p < 0.05) and eosinophil counts higher (p < 0.05) in the animals receiving the HP diet suggesting that resistance was enhanced by protein supplementation. Meanwhile, milk parameters (related to resilience) were not affected by the level of protein in the diet when considering the whole groups. In contrast, in the high producing goats, the milk production and milk composition parameters were improved with the HP diet. To conclude, we have seen that the expression of both resistance and resilience did not appear when the coverage of the protein requirements was insufficient. Because the milk production is dependent on the protein supply, we suggest that there is a competition in the use of the protein between the development of resistance and the milk production. 相似文献

Infections of the gastrointestinal tract with parasitic nematodes, and particularly Haemonchus contortus, represent one of the main pathological constraints on the breeding of small ruminants in tropical countries. Owing to the constant development of anthelmintic resistance in worm populations, alternative control methods are being evaluated, including an assessment of the value of the traditional plants. The effect of the consumption of Fagara leaves on sheep infected with gastrointestinal nematodes was therefore measured both under controlled conditions and in a field survey. In sheep experimentally infected with 2500 third-stage larvae of H. contortus, a three-day administration of Fagara was associated with a decrease in egg excretion and a significant reduction in the fertility of female worms, without changes in the worm number. A reduction in egg output was also confirmed from the results obtained of a survey or naturally infected sheep performed in smallholder farms. This survey also indicated that the repeated consumption of small amounts of Fagara leaves was more effective than a single three-day administration. Results of both studies suggest that Fagara leaves might be an alternative option to chemotherapy to modulate the epidemiology of H. contortus in tropical countries. 相似文献

Mixed parasitic infection of animals is a common phenomenon in nature. The existence of one species often positively or negatively influences the survival of the other. Our experimental study was started with the objectives to demonstrate the interaction of Haemonchus contortus and Oestrus ovis in relation to cellular and humoral immune responses in sheep. Twenty-two sheep of Tarasconnais breed (France) were divided into four groups (O, OH, H and C) of five or six animals. Group O and OH received 5 weekly consecutive inoculations with O. ovis L1 larvae (total = 82 L1) in the first phase of the experiment between days 0 and 28. On the second phase, groups OH and H received 5000 L3 of H. contortus on day 48 while group C served as our control throughout the experimental period. Parasitological, haematological, serological and histopathological examinations were made according to standard procedures and all animals were slaughtered at day 95. There was no significant variation in the number and degree of development of O. ovis larvae between the two infected groups. Furthermore, in tissues examined in the upper respiratory tract (nasal septum, turbinate, ethmoide and sinus), group O and OH has responded similarly on the basis of cellular inflammatory responses (blood and tissue eosinophils, mast cells and globule leucocytes (GL)) and serum antibody responses against the nasal bots. This may indicate that the presence of H. contortus in the abomasa of group OH had no marked influence over the development of O. ovis larvae in the upper respiratory tract. On the other hand, we have observed a significantly lower H. contortus female worm length, fecal egg count (FEC) and in utero egg count in animals harbouring the nasal bot (OH) than in the mono-infected group (H). This was significantly associated with higher blood eosinophilia, higher packed cell volume (PCV) and increased number of tissue eosinophils and globule leucocytes. We conclude that, the establishment of O. ovis larvae in the upper respiratory tract has initiated higher inflammatory cellular activity in group OH there by influencing the development and fecundity of H. contortus in the abomasum. 相似文献

The pestivirus bovine viral diarrhea virus (BVDV) is known to bind to the CD46 molecule, which subsequently promotes entry of the virus. Mapping of the BVD-virion-binding site has shown that two peptides, 66EQIV69 and 82GQVLAL87, located on antiparallel beta sheets in the most distal complement control protein module (CCP1), provide the attachment platform. In the present study, we reveal the existence of ten distinct allelic versions of the CCP1 module, varying significantly in frequency among taurine and indicine races. A complex mRNA splicing pattern was also evidenced for bovine CD46, generating three different serine-threonine-proline segments and five different cytoplasmic domains. The four most frequent allelic variants and the six splice variants were then expressed in BVDV-nonpermissive porcine cells and the quantity of progeny virions generated by each cell preparation was measured 48 h post-infection. As expected, ectopic expression of the 10 bovine CD46 isoforms rendered the PK15 cells permissive to BVDV, as attested by the 100,000-fold greater recovery of virions from these cells than from non-transfected cells. This permissivity increase was significantly lower (-33%, P<0.001) when the canonical CCP1 was replaced with the variant most frequent in zebus, suggesting positive or negative selection of this allele in the latter and in the former, respectively. The predicted secondary structure of this variant suggests that the measured loss of function is due to the disappearance of one of the two beta sheets constituting the BVDV attachment platform. On the other hand we showed that for a given CCP1, the titer recovered at 48 hpi also depended on the nature of the CD46 cytoplasmic domain (P<0.001). This result implies that virus binding generates a cytoplasmic-tail-dependent outside-in signal that determines permissivity to BVDV. 相似文献
