The efficacy of the pour-on formulation of eprinomectin, at a dose rate of 0.5 mg/kg bodyweight, was assessed in sheep against three main species of gastrointestinal nematodes and against the nasal bot fly, Oestrus ovis, and some pharmacokinetic parameters were determined for 21 days after the treatment. By comparison with untreated control sheep, infected experimentally with Haemonchus contortus, Teladorsagia circumcincta and Trichostrongylus colubriformis, eprinomectin was 100 per cent effective against the two abomasal species and 99.5 per cent effective against T. colubriformis. In ewes naturally infected with the nasal bot fly, the efficacy of the drug against O. ovis was 97.7 per cent. The mean (se) systemic area under the curve (AUC) was 56.0 (26.2) ng/day/ml and the mean residence time was 5.3 (1.0) days, but there were wide variations between individual sheep.  相似文献

To evaluate the efficacy with which recommendations of means to avoid the spread of anthelmintic resistance in parasitic nematode populations are communicated to farmers on dairy goat farms in France, a questionnaire survey was undertaken on their use of antiparasitic drugs. Information was collected from 73 farms in two main areas of dairy goat production. The data referred to three years. Anthelmintics were used in 69 farms, the mean number of treatments per year being 2.74. Changing the drug from one year to another was not practised. Moreover, of the 58 farms using two or more treatments per year, only 37% used anthelmintics from different classes in the lactation and drying-off periods. Benzimidazoles and probenzimidazoles were given in all except two farms and these substances represented more than 80% of all the treatments. Levamisole/pyrantel or avermectins were used in 15% and 27% of the farms, respectively. Double the ovine dose, as recommended in goats to ensure efficacy of benzimidazoles, was applied in 55% of the farms. In addition, in all the flocks, the substances were given on the basis of a mean estimated live weight and not by reference to the heaviest animal. These results indicate that errors in the use of anthelmintics are still frequent in dairy goat farms in France, with probable consequences for the spread of anthelmintic resistance in the populations of parasites.  相似文献

The level of dietary protein is known to alter the establishment and the pathophysiology of gastrointestinal nematodes in sheep. On the other hand, high-producing dairy goats are less resistant and/or resilient than low-producing ones to experimental nematode infection. During a 2-year study, we have investigated the course of a natural nematode infection (mainly T. colubriformis) in a high-producing dairy goat flock. In year 1, 50 grazing goats divided in high (HP) or low producer (LP) were compared from April to October for parasitological and milk parameters. In year 2, the 25 HP goats were only considered and were allocated to two levels of dietary protein, high level (HPr) with a protein coverage of 125% or normal level (NPr) with a protein coverage of 106%. They were monitored as above. In year 1, HP goats showed a greater nematode egg output (1856 vs. 1000epg) associated with higher values in T. colubriformis IgG in autumn than LP ones whereas the decrease in serum phosphate concentration was similar in both groups. In year 2, HPr goats exhibited lower egg output in autumn (2219 vs. 2817) vs. NPr ones. Moreover, milk yield and fat content were significantly higher in HPr vs. NPr in the 2nd part of the study. HP goats are less resistant to nematode infection in natural conditions. Resistance and resilience of HP goats may partially be improved by a protein supplementation in the diet.  相似文献

Bacteremia is an important cause of morbidity and mortality in humans. In this study, we focused on the development of an animal model of bacteremia induced by non-typhoidal Salmonella. New Zealand White rabbits were inoculated with a human isolate of non-typhoidal Salmonella strain CVD J73 via the intra-peritoneal route. Blood samples were collected at specific time points and at euthanasia from infected rabbits. Additionally, tissue samples from the heart, lungs, spleen, gastrointestinal tract, liver and kidneys were obtained at euthanasia. All experimentally infected rabbits displayed clinical signs of disease (fever, dehydration, weight loss and lethargy). Tissues collected at necropsy from the animals exhibited histopathological changes indicative of bacteremia. Non-typhoidal Salmonella bacteria were detected in the blood and tissue samples of infected rabbits by microbiological culture and real-time PCR assays. The development of this animal model of bacteremia could prove to be a useful tool for studying how non-typhoidal Salmonella infections disseminate and spread in humans.  相似文献

Infections with gastrointestinal nematodes (GIN) remain a major threat for ruminant production, health and welfare associated with outdoor breeding. The control of these helminth parasites has relied on the strategic or tactical use of chemical anthelmintic (AH) drugs. However, the expanding development and diffusion of anthelmintic resistance in nematode populations imposes the need to explore and validate novel solutions (or to re-discover old knowledge) for a more sustainable control of GIN. The different solutions refer to three main principles of action. The first one is to limit the contact between the hosts and the infective larvae in the field through grazing management methods. The latter were described since the 1970s and, at present, they benefit from innovations based on computer models. Several biological control agents have also been studied in the last three decades as potential tools to reduce the infective larvae in the field. The second principle aims at improving the host response against GIN infections relying on the genetic selection between or within breeds of sheep or goats, crossbreeding of resistant and susceptible breeds and/or the manipulation of nutrition. These approaches may benefit from a better understanding of the potential underlying mechanisms, in particular in regard of the host immune response against the worms. The third principle is the control of GIN based on non-conventional AH materials (plant or mineral compounds). Worldwide studies show that non conventional AH materials can eliminate worms and/or negatively affect the parasite's biology. The recent developments and pros and cons concerning these various options are discussed. Last, some results are presented which illustrate how the integration of these different solutions can be efficient and applicable in different systems of production and/or epidemiological conditions. The integration of different control tools seems to be a pre-requisite for the sustainable management of GIN infections. This new era of GIN management requires a new paradigm: to achieve enough control to reduce the negative impact of GIN infections enabling an optimum level of production, health and welfare.  相似文献

The present trial aimed at comparing the sensitivity of two in vitro methods, i.e. the larval migration inhibition assay (LMIA) and the larval exsheathment inhibition assay (LEIA), to evaluate the anthelmintic (AH) properties of tannin-rich plant extracts against Haemonchus contortus infective larvae. The two assays were applied on the same batch of H. contortus infective larvae exposed to water/acetonic extracts obtained from four tropical plants with different tannin contents: Acacia gaumeri, Brosimum alicastrum, Havardia albicans and Leucaena leucocephala. Increasing concentrations (0, 75, 150, 300, 600, 1200 μg/ml PBS) of lyophilized extracts were used in both in vitro assays. A general lineal model test was used to determine the dose-effect in the LMIA or the difference in the percentage of exsheathed larvae between the respective control and treated groups. The LMIA showed a dose-dependent AH effect for H. albicans (P<0.001) and A. gaumeri (P<0.05), but not for L. leucocephala and B. alicastrum. In contrast, the exsheathment process was significantly affected by all doses of H. albicans and A. gaumeri extracts and a significant dose-dependent effect was found for B. alicastrum and L. leucocephala. Calculation of lethal dose (LD) was possible with LEIA using B. alicastrum and L. leucocephala but not with H. albicans and A. gaumeri as the lowest tested concentration was achieving more than 50% inhibition. Calculation of LD with the LMIA results was not feasible. These results suggest that tannin-rich plant extracts are more potent inhibitors of the exsheathment of H. contortus L(3) larvae than their motility. This information underlines the difference of sensitivity between methodological procedures to evaluate the AH properties of plant extracts on the same nematode stage.  相似文献

The MHC (BoLA) type has been determined for cattle from three breeds in West Africa. Seventy Baoule, 50 N'Dama and 30 Zebu cattle from the centre and north of the Ivory Coast were tested. Lymphocytes from these cattle were tested in a lymphocytotoxicity test with alloantisera detecting all of the internationally recognised BoLA sera. 78 sera prepared in Edinburgh and 57 in Jouy-en-Josas were used in the study. The results showed that sera prepared in Europe detect similar specificities in West Africa. Although with some specificities the frequencies differ from those seen in Europe. The frequency of null alleles is higher than in Europe in the Ndama and Zebu animals indicating the existence of additional specificities which will require the production of alloantisera in these breeds. However in the Baoule the null allele frequency is lower even then in some European breeds. The population data in which no animals have more than two workshop specificities is consistent with a single locus control in West Africa as in Europe.  相似文献

Resistance to anthelmintics in nematode parasite of the digestive tract is a major concern in small ruminants and particularly in goats. One possible solution to limit the development and spread of resistance is to give treatments on a selective basis, i.e. by targeting the most susceptible animals within a flock rather than treating all the animals. In dairy goats, epidemiological studies have shown that, within a flock, goats in first lactation and the multiparous ones with the highest level of milk production were highly receptive to parasite infection. The objective of the study was to assess whether selective treatments could achieve a level of control of nematodes similar to systematic drenching. A similar experimental design was applied for two successive years. An experimental flock of 120 dairy goats was divided into two groups. All the goats from group SYS (systematic treatment) were drenched at mid-grazing season (July). In contrast, anthelmintics in group SEL (selective treatment) were restricted to the goats in first lactation plus the high producers. Overall, these treated goats represented 48% of the flock in year 1 and 66% in year 2. After treatments, both groups grazed on separate pastures. Parasitological and pathophysiological measurements were performed monthly. The results indicate a similar level of egg excretion in the two groups. The pathophysiological parameters (pepsinogen and phosphate concentrations) were also similar in the two groups, as well as the milk production for the 2 years. These results suggest that a targeted use of anthelmintics may allow efficient control of gastrointestinal nematodes whilst resulting in a predicted reduction in the selection pressure for the development of anthelmintic resistance.  相似文献

Since usual processes involve water as absorbent, they appear not always really efficient for the treatment of hydrophobic volatile organic compound (VOC). Recently, absorption and biodegradation coupling in a two-phase partitioning bioreactor (TPPB) proved to be a promising technology for hydrophobic compound treatment. The choice of the organic phase, the non-aqueous phase liquid (NAPL) is based on various parameters involved in both steps of the process, hydrophobic VOC absorption in a gas?Cliquid contactor, and biodegradation in the TPPB. VOC solubility and diffusivity in the selected NAPL, as well as NAPL viscosity, seems to be the main parameters during the absorption step, while biocompatibility, namely the absence of toxic effect of the NAPL towards microorganisms, non-biodegradability and VOC partition coefficient between NAPL and water were revealed as the key factors during the biodegradation step. The screening of the various NAPL available in the literature highlighted two families of compounds matching the required conditions for the proposed integrated process, silicone oils and ionic liquids.  相似文献
