Upland heather moors are high priority biotopes for conservation in Britain, but are under threat from a number of sources. One such threat is a change from a vegetation dominated by dwarf shrubs, mainly Calluna vulgaris, to a grassland dominated by Molinia caerulea. Restoration of a dwarf shrub community has proved problematic with studies showing that herbicide use may be necessary to control Molinia. Previously the non-selective herbicide, glyphosate, has been recommended, but this herbicide may damage Calluna and other moorland species severely. Here, we tested a range of selective herbicides (graminicides) for use in an herbicide-based conservation strategy under field conditions. Two selective herbicides, propaquizafop and quizalofop-ethyl, gave a short-term check to Molinia and another, cycloxydim, provided a reduction for at least 1 year, but this effect disappeared after 3 years. Damage to Calluna was less than that caused by glyphosate, and the selective herbicides had little effect on other moorland species present. Our findings suggest that selective herbicides have a potential role in reducing Molinia during moorland restoration, although follow-up applications, and appropriate moorland management, will also be needed. 相似文献

Purpose
The application of organic and inorganic fertilizers to soil can result in increased gaseous emissions, such as NH3, N2O, CO2, and CH4, as well as nitrate leaching, contributing to climate warming and ground and surface water pollution, particularly in regions with hot climates, where high temperatures and high soil nitrification rates often occur. The use of nitrification inhibitors (NIs) has been shown to effectively decrease nitrogen (N) losses from the soil-plant system.Materials and methods
Non-disruptive laboratory incubation experiments were conducted to assess the extent to which temperature (20 and 30 °C) and nutrient source (mineral and organic fertilizers) influence the rate of carbon (C)- and N-related microbial processes in soil in response to the NI 3,4-dimethylpyrazole phosphate (DMPP). Furthermore, short-term changes in the ability of microbes to degrade C substrates were evaluated in disruptive soil microcosms using microbial community-level physiological profiling and the abundance of the bacterial 16S rRNA gene as a measure of total bacterial population size.Results and discussion
DMPP reduced net nitrification after 2 and 4 weeks of incubation at 30 and 20 °C by an average of 78.3 and 84.5 %, respectively, and with similar dynamics for mineral or organic fertilization. The addition of labile organic matter with cattle effluent led to a rapid increase in C mineralization that was significantly reduced by DMPP at both temperatures, whereas no changes could be detected after the addition of mineral fertilizer. The culturable heterotrophic microorganisms showed metabolic diversification in the oxidation of C sources, with organic fertilizer playing a major role in the substrate utilization patterns during the first week of incubation and the DMPP effects prevailing from day 14 until day 28. Furthermore, the copy number of the bacterial 16S rRNA gene was reduced by the application of DMPP and organic fertilizer after 28 days.Conclusions
Our results show the marked efficiency of DMPP as an NI at elevated temperatures of incubation and when associated with both mineral and organic fertilization, providing support for its use as a tool to mitigate N losses in Mediterranean ecosystems. However, we also observed impaired C respiration rates and bacterial abundances, as well as shifts in community-level physiological profiles in soil, possibly indicating a short-term effect of DMPP and organic fertilizers on non-target C-related processes and microorganisms.983.

A novel system was investigated, finalized to reduce the impact of highly polluting wastewaters, and based on combined actions of catalytic oxidations and microbial biotechnologies. Olive oil mill wastewaters (COD 10,000-100,000 mg O(2)/L) were oxidized up to 80-90% by stoichiometric amounts of dilute hydrogen peroxide (35%) and in the presence of water soluble iron catalysts, either Fe(II) or Fe(III), at concentrations up to 1% w/w and more, i.e., much larger than those reported for conventional Fenton processes. In the combined action, the mineralization activity of a selected microbial consortium was used to degrade residual volatile and nonvolatile organic compounds into CO(2) and biomass. The concentrations of ambient gas-phase hydrogen peroxide were measured during the summer of 1998, 1999 and 2000. The experiments were performed in the city of Wroclaw and in the vicinity of Mount Szrenica, 1362 m a.s.l., Poland. Analysis was carried out by the chemiluminescence method. Typical mean ranges of 30 min H2O2 concentrations measured were 1.4–6.0 μg m-3 at Mount Szrenica, whereas in the urban atmosphere H2O2 concentrations were in the range of 2.7–11.7 μg m-3. In the case of the urban atmosphere, H2O2 concentrations were well correlated only with solar radiation and temperature. In the mountain air, H2O2 concentrations increased along with the increase of temperature, O3, CO and the decrease of humidity. Bioassay-guided fractionation of the ethyl acetate extract of Ruta graveolens (common rue) leaves led to the isolation of the furanocoumarins 5-methoxypsoralen (5-MOP), 8-methoxypsoralen (8-MOP), and the quinolone alkaloid graveoline as phytotoxic constituents. Graveoline and 8-MOP substantially inhibited growth of Lactuca sativa (lettuce) seedlings and reduced chlorophyll content at 100 microM; this effect was not due to a direct effect on chlorophyll synthesis. Radical growth of L. sativa was inhibited by 10 microM 8-MOP. Graveoline inhibited growth of Lemna paucicostata (duckweed) at 100 microM. This is the first report of the phytotoxic activity of graveoline. Growth of Agrostis stolonifera (bentgrass) was inhibited by 5-MOP at 30 microM. All three compounds substantially reduced cell division in Allium cepa (onion) at or below 100 microM. None of the compounds caused significant cellular leakage of Cucumis sativus (cucumber) cotyledon disks at 100 microM. All three compounds inhibit plant growth, at least partially through inhibition of cell division. 相似文献

Ammonia-oxidising bacteria (AOB) in limed and non-limed acidic coniferous forest soil were investigated using real-time PCR. Two sites in southern Sweden were studied, 244 Åled and Oxafällan. The primers and probe used earlier appeared to be specific to the 16S rRNA gene of AOB belonging to the β-subgroup of the Proteobacteria [Appl. Environ. Microbiol. 67 (2001) 972]. Plots treated with two different doses of lime, 3 or 6 t ha−1, were compared with non-limed control plots on two occasions during a single growing season. Three different soil depths were analysed to elucidate possible differences in the density of their AOB communities. The only clear effect of liming on the AOB was recorded in the beginning of the growing season at 244 Åled. In samples taken in April from this site, the numbers of AOB were higher in the limed plots than in the control plots. At the end of the growing season the AOB communities were all of a similar size in the different plots at both sites, irrespective of liming. The number of AOB, determined using real-time PCR, ranged between 6×106 and 1×109 cells g−1 soil (dw) at the two sites, and generally decreased with increasing soil depth. The results showed no correlation between community density and potential nitrification. This may indicate a partly inactive AOB community. Furthermore, more than 107 cells g−1 soil (dw) were recorded using real-time PCR in the control plot at 244 Åled, although Bäckman et al. [Soil Biol. Biochem. 35 (2003) 1337] detected no AOB like sequences in the same plots using PCR followed by DGGE. Taken together our results strongly suggest that the primers and probe set used are not well suited for quantifying AOB in acidic forest soils, which is probably due to an insufficient specificity. This shows that it is extremely important to re-evaluate any primers and probe set when used in a new environment. Summary Phaseolin variability was analyzed in 86 wild Phaseolus lunatus accessions, representing a wide geographical distribution from Mexico to Argentina, to determine geographical patterns of variability. The collection can be divided into two major groups. One group consists of accessions from Mexico, Central America, Cuba, Columbia, Argentina showing two Mesoamerican (M) phaseolin pattern types, already described in cultivated materials, as well as a variety of additional banding patterns. This suggests that at least two genotypes in the Mesoamerican gene pool were involved in a domestication process and spread throughout the world. The other group, consisting of wild Andean (A) forms from Northern Peru and belonging to the Andean gene pool, showed, among others, two A pattern types comparable to those found in cultivated large-seeded accessions. 相似文献

Atmospheric deposition was sampled at six stations located at different latitudes in north western Italy; chemical analyses showed mineral acidity and the prevalence of sulphate, nitrate, ammonium and calcium. A comparative examination of the data collected in 1985-93 at the different stations showed a regular decrease of concentrations from south to north; the gradient is linear and not dependent on altitude. This pattern is clearly correlated with the meteorology, as the study area receives masses of air mostly from the S.E. and S.W., i.e. from the Po Plain, one of the most densely populated and industrialized areas in Italy. On the other hand, the loads of ions from the atmosphere show maxima in a band covering the subalpine area, because of the higher amounts of precipitation.
