Keratinocytes from explants of the oral mucosa of dogs were grown in culture for five passages. The ultrastructure of primary cultures and fully developed subcultures passaged 1, 3, and 5 times was examined. At every stage, the cells had the morphologic characteristics of epithelial cells and formed a multilayered squamous epithelium. The basal cells had the characteristics of metabolically active cells, whereas the suprabasal cells and the cells at the media interface expressed many, but not all, of the organelles and cell surface characteristics associated with keratinocyte differentiation. Keratohyalin granules were located in the suprabasal and superficial cells. Cell size and shape and the relationship between cells in the layers also reflected the morphologic characteristics of the parent tissue. Cells maintained this typical structure through all passages and the cultures changed minimally for up to a week after development.  相似文献

Plasma membranes were isolated from cultured canine keratinocytes by paraformaldehyde-induced membrane vesiculation. The isolated plasma membrane vesicles retained cell surface antigens (eg, a pemphigus vulgaris antigen). These membrane vesicles were used as an antigen source for the production of monoclonal antibodies. Eight antibodies that had specific reactivity to the cytoplasmic membrane of keratinocytes on frozen sections of canine esophagus were identified by use of an indirect immunoperoxidase method. The stratified squamous epithelium of the esophageal mucosa had 4 staining patterns. When applied to frozen sections of canine skin, lip, and tongue, the antibodies had different tissue specificities for differing stratified squamous epithelia. Using sodium dodecyl sulfate polyacrylamide-gel electrophoresis and the western blot technique, one of the antibodies was specific for a 60-kD cell surface molecule. Therefore, such monoclonal antibodies may be useful in defining heterogeneity between different stratified squamous epithelia, in identifying biologically important surface antigens, or in the diagnosis of tumors.  相似文献

The cytotoxic immune response of puppies to feline sarcoma virus induced tumors was studied. Neonatal puppies were compared with adolescent dogs. Three different types of cytotoxicity were investigated: complement dependent cytotoxicity, T-cell-mediated cytotoxicity and antibody dependent cellular cytotoxicity. The relationship between the spontaneous regression of the sarcoma and the development of the immune system of the puppies is discussed.  相似文献

Specialized secretion systems are used by many bacteria to deliver effector proteins into host cells that can either mimic or disrupt the function of eukaryotic factors. We found that the intracellular pathogens Legionella pneumophila and Coxiella burnetii use a type IV secretion system to deliver into eukaryotic cells a large number of different bacterial proteins containing ankyrin repeat homology domains called Anks. The L. pneumophila AnkX protein prevented microtubule-dependent vesicular transport to interfere with fusion of the L. pneumophila-containing vacuole with late endosomes after infection of macrophages, which demonstrates that Ank proteins have effector functions important for bacterial infection of eukaryotic host cells.  相似文献

Esophageal neoplasms were diagnosed in 8 of 49,229 dogs seen over the last 11 years at the Veterinary Medical Teaching Hospital of the University of California at Davis. The neoplasms were primary in 2 dogs and metastatic to the esophagus in 6 dogs, with thyroid carcinoma being the most common site of the primary tumor. The most common clinical signs were regurgitation, dysphagia, weight loss, development of neck masses, and respiratory difficulties. It was concluded that the clinical signs often can be misleading. The interpretation of survey radiographs, barium contrast studies, or fluoroscopic studies often provide the initial data base. The final diagnosis requires histologic examination. Retention of air in the esophagus (with or without esophageal displacement) and motor dysfunction (with or without gross morphologic changes) are the most important criteria for radiographic diagnosis.  相似文献

Neospora caninum, Toxoplasma gondii and Eimeria bovis are coccidian parasites of veterinary importance. Tachyzoites of N. caninum and T. gondii and sporozoites of E. bovis are able to invade and replicate in endothelial cells in vivo and in vitro. As it holds true for all eukaryotic cells, the survival of parasitized host cells and the parasites themselves should be dependent on ion balances, especially on extra- and intracellular calcium concentrations. Addition of the calcium ionophore A23187 reliably did release merozoites from mature N. caninum and T. gondii meronts grown in cultured primary bovine umbilical vein endothelial cells (BUVEC). Extent and time course of merozoite release depended on both, maturity of the meronts and concentration of the calcium ionophore. Attempts, however, to achieve synchronous release of merozoites from E. bovis first generation meronts by ionophore treatment failed, suggesting a different biological behaviour of this parasite. According to microscopical observations, the quite variable time of E. bovis macromeront maturation and a hampered merozoite exit owing to dense parasite-induced cytoskeleton elements surrounding the meront may be a reason for the lack of inducible synchronous release. In rangelands, grazing management is a main driver of rangeland condition. Due to masking effects of seasonal climate fluctuations, little is known about (dis)similarity of management effects on rangeland condition and forage provision across major dryland biomes. Taking a macro-ecological perspective, we analysed if management effects differed between South Africa’s central grassland and Kalahari savanna biomes. We recorded proxies of forage provision (phytomass, vegetation cover and their ratio) over five seasons, annual rainfall to account for seasonal climate fluctuations, and rangeland condition (through relative abundances of increaser and decreaser species). Regarding forage provision, we found effects of management for the savanna, where, irrespective of rainfall, rotational grazing management resulted in higher phytomass and phytomass–cover ratios than management with continuous grazing. In the grassland, however, this difference was only discernible for phytomass–cover ratio in two years with above-average antecedent rainfall. This suggests that management effects are biome-dependent and that modulating effects of annual rainfall are stronger in the grassland. In either biome, management effects on the dominance of increaser and decreaser species were negligible, i.e. rangeland condition did not differ across management types in either biome. We conclude that investigations on management effects should account for interactions with biome and rainfall.  相似文献
