Two- to four-week stratification of red alder seeds slightly increased the speed of germination but did not improve total germination under alternating warm temperatures (30°C/20°C), yet significantly improved speed and total germination under cool temperatures (15°C/5°C) that simulated early spring outdoor nursery bed conditions. Only seeds that received four-week stratification achieved complete germination at the cooler temperatures. Strong seed source variation in response to seed treatments was observed at the cool regime. The Yacolt source germinated well regardless of seed treatments, even without stratification, while five other sources needed two to four weeks of stratification to achieve above 50% germination. Isolation of Shiga-toxin (Stx) positive Escherichia coli O157:H7 from commercially grown pigs has been reported. Furthermore, experimental infection studies have demonstrated that Stx-positive E. coli O157:H7 can persist in 12-week-old experimentally orally inoculated conventional pigs for up to 2 months and that persistence was not dependent upon intimin. We have shown that the flagellum of Stx-negative E. coli O157:H7 does not have a role to play in pathogenesis in ruminant models whereas, in poultry, the flagellum of E. coli O157:H7 was important for long-term persistent infection. The contribution of the flagellum of Stx-negative E. coli O157 in the colonisation of pigs was investigated by adherence assays on a porcine (IPI-21) cell line, porcine in vitro organ culture (IVOC) and experimental oral inoculation of conventional 14-week-old pigs. E. coli O157:H7 NCTC12900nal(r) and isogenic aflagellate and intimin deficient mutants adhered equally well to IPI-21 cells. In porcine IVOC association assays, E. coli O157:H7 NCTC12900nal(r) was associated in significantly higher numbers to tissues from the caecum and the terminal rectum than other sites. The aflagellate and intimin deficient mutants significantly adhered in greater numbers to more IVOC gastrointestinal tissues than the parent. Groups of 14-week-old pigs were dosed orally with 10(10)CFU/10ml of either E. coli O157:H7 NCTC12900nal(r) or isogenic aflagellate and intimin deficient mutants and recovery of each test strain was similar. Histological analysis of pig tissues at post mortem examination revealed that E. coli O157 specifically stained bacteria were associated with the mucosa of the ascending and spiral colon. These data suggest that colonisation and persistence of Stx-negative E. coli O157:H7 in pigs, involves mechanisms that do not require the flagellum or intimin. 相似文献

Juvenile wood (JW) of conifers is often associated with compression wood (CW), with which it is sometimes believed to be identical. To determine whether JW and CW can be distinguished metabolically, we compared gas chromatographic profiles of 25 polar metabolites from rooted cuttings of a single loblolly pine (Pinus taeda L.) clone raised in controlled environment chambers and subject to three treatments: (1) grown erect with minimal wind sway (control); (2) swayed by wind from oscillating fans; and (3) with 30-cm growth increments successively bent at an angle of 45 degrees to the vertical. Profiles were compared by principal component analysis. Substantial increases in abundances of coniferin and p-glucocoumaryl alcohol separated immature JW-forming xylem tissues of the control trees from the CW-forming xylem of the bent and swayed trees. 相似文献

The stable propagation of genetic material during cell division depends on the congression of chromosomes to the spindle equator before the cell initiates anaphase. It is generally assumed that congression requires that chromosomes are connected to the opposite poles of the bipolar spindle ("bioriented"). In mammalian cells, we found that chromosomes can congress before becoming bioriented. By combining the use of reversible chemical inhibitors, live-cell light microscopy, and correlative electron microscopy, we found that monooriented chromosomes could glide toward the spindle equator alongside kinetochore fibers attached to other already bioriented chromosomes. The movement of dislocations in a crystal is the key mechanism for plastic deformation in all materials. Studies of dislocations have focused on three-dimensional materials, and there is little experimental evidence regarding the dynamics of dislocations and their impact at the atomic level on the lattice structure of graphene. We studied the dynamics of dislocation pairs in graphene, recorded with single-atom sensitivity. We examined stepwise dislocation movement along the zig-zag lattice direction mediated either by a single bond rotation or through the loss of two carbon atoms. The strain fields were determined, showing how dislocations deform graphene by elongation and compression of C-C bonds, shear, and lattice rotations. 相似文献

Half of the world's land base is grazed by domesticated livestock. Because of the important functional role of ants in grasslands, it is important to understand the effect of livestock grazing on ant abundance and diversity. The objectives of this study were to examine the effect of cattle grazing and site productivity on the abundance, species richness, and species diversity of ants in Lac du Bois Grasslands Provincial Park, British Columbia, Canada. We hypothesized that the measured ant variables would be lowest in grazed areas and at low site productivity. Pitfall trapping was conducted at four sites: two at each low and high site productivity levels. At each site an ungrazed (fenced exclosure) and grazed transect was sampled during May, July, and August of 2008. Captured ants were preserved in ethanol and identified. Eight genuses of ants were collected: Tapinoma, Camponotus, Formica, Lasius, Aphaenogaster, Myrmica, Solinopsis, and Temnotharox. The mean number of ants per pitfall was higher at high site productivity sites that were grazed (15.10 ± SE) compared to high productivity sites ungrazed (3.28 ± SE); grazing at low productivity reduced numbers of ants from 5.07 (± 0.70 SE) to 2.20 (± 0.39 SE) (F = 21.806; P [ 0.001). Tapinoma sessile and A. occidentalis had the greatest numbers in the pitfall traps. Species richness (F = 23.330, P [ 0.001) and diversity (F = 11.764, P = 0.001) followed a similar trend. Because productivity and cattle grazing affect ant diversity and abundance, and ants impact ecosystem functioning, these factors should be considered in management of grasslands. 相似文献

In order to monitor lead pollution from urban areas to coral reefs in the Gulf of Thailand, linear and two-dimensional distributions of Pb in corals from Khang Khao Island, Thailand and Rukan-sho, Okinawa, were analysed with high resolution. Laser ablation inductively coupled plasma mass spectrometry was applied to measure Pb content in coral skeletons using synthetic Pb standards in a CaCO3 matrix as calibration materials. Linear and two-dimensional ablation schemes were applied to determine the Pb content in corals collected from Khang Khao Island, Thailand and Rukan-sho, Okinawa. The coral skeleton was ablated by Nd:YAG laser (wavelength of 266 nm, beam diameter of 155 µm, scanning speed of 10 µm s?1 and frequency of 10 Hz) along the growth axis, and ion counts for 208Pb were normalised to 42Ca. Lead content in the corals was determined using a calibration curve obtained from the synthetic Pb standards (0–141 µg g?1). The linear ablation track of the sample from Khang Khao showed over 30 peaks of Pb with an average value of 3.55 µg g?1, while Pb content in the coral from Rukan-sho showed relatively small variation with an average value of 0.132 µg g?1. Two-dimensional imaging of Pb in the coral skeletons was also carried out for an area of 7?×?20 mm on the sample from Khang Khao. The distribution patterns of Pb and Ba in the Thailand coral co-varied. These observations from the linear and imaging analyses suggest discontinuous inputs of anthropogenic Pb from rivers to the Gulf of Thailand. 相似文献

Although data quality and weighting decisions impact the outputs of reserve selection algorithms, these factors have not been closely studied. We examine these methodological issues in the use of reserve selection algorithms by comparing: (1) quality of input data and (2) use of different weighting methods for prioritizing among species. In 2003, the government of Madagascar, a global biodiversity hotspot, committed to tripling the size of its protected area network to protect 10% of the country’s total land area. We apply the Zonation reserve selection algorithm to distribution data for 52 lemur species to identify priority areas for the expansion of Madagascar’s reserve network. We assess the similarity of the areas selected, as well as the proportions of lemur ranges protected in the resulting areas when different forms of input data were used: extent of occurrence versus refined extent of occurrence. Low overlap between the areas selected suggests that refined extent of occurrence data are highly desirable, and to best protect lemur species, we recommend refining extent of occurrence ranges using habitat and altitude limitations. Reserve areas were also selected for protection based on three different species weighting schemes, resulting in marked variation in proportional representation of species among the IUCN Red List of Threatened Species extinction risk categories. This result demonstrates that assignment of species weights influences whether a reserve network prioritizes maximizing overall species protection or maximizing protection of the most threatened species. 相似文献
