The concentrations of selected metals—Cr, Ni, Cu, Zn, Cd, and Pb—were determined in the samples of Hypogymnia physodes lichen and Pleurozium schreberi moss collected in Polish and Czech Euroregions Praded and Glacensis. More specifically, the samples were collected in Bory Stobrawskie, Bory Niemodlińskie, and Kotlina K?odzka (Poland) and in Jeseniki (Czech Republic). The concentration of metals in the samples was measured using the atomic absorption spectrometry (flame AAS technique and electrothermal atomization AAS technique). The results were used to calculate the comparison factor (CF) that quantifies the difference in concentration of a given bioavailable analyte × accumulated in lichens and mosses: CF?=?2 (c x,lichen ? c x,moss) (c x,lichen + c x,moss)?1. The values of CF greater than 0.62 indicate the most probable location of heavy metals deposited in the considered area. In this work, the method was used to show a significant contribution of urban emissions to the deposition of heavy metals in the area of Bory Stobrawskie and in the vicinity of K?odzko City. 相似文献

Escherichia coli is one of the major pathogens in humans and animals causing localized and systemic infections, which often lead to acute inflammation, watery diarrhea, and hemorrhagic colitis. Bacterial lipopolysaccharide (LPS) and Shiga exotoxins (Stx) are mostly responsible for such clinical signs. Therefore, highly effective treatment of E. coli infections should include both eradication of bacteria and neutralization of their toxins. Here, for the first time, we compared the in vitro ability of common antibiotics to decrease LPS‐ and Stx‐mediated cytotoxicity: colistin, amoxicillin (used separately or combined), enrofloxacin, and its metabolite ciprofloxacin. Three experimental scenarios were realized as follows: (a) the direct effect of antibiotics on endotoxin, (b) the effect of antibiotic treatment on LPS‐mediated cytotoxicity in an experiment mimicking “natural infection,” (c) the effect of antibiotics to decrease Stx2e‐mediated cytotoxicity. Two cell lines, A549 and Vero cells, were used to perform cytotoxic assays with the methyl tetrazolium (MTT) and lactate dehydrogenase leakage (LDH) methods, respectively. Colistin and amoxicillin, especially used in combination, were able to attenuate LPS toxic effect, which was reflected by increase in A549 cell viability. In comparison with other antibiotics, the combination of colistin and amoxicillin exhibited the highest boster or additive effect in protecting cells against LPS‐ and Stx2e‐induced toxicity. In summary, in comparison with fluoroquinolones, the combination of colistin and amoxicillin at concentrations similar to those achieved in plasma of treated animals exhibited the highest ability to attenuate LPS‐ and Stx2e‐mediated cytotoxicity. 相似文献

Acetaminophen (paracetamol) is used in dogs to manage fever and mild pain. The aim of this study was to assess the pharmacokinetics of acetaminophen in both fed and fasted Labrador Retrievers after a single intravenous and oral administration (20 mg/kg). Six healthy dogs underwent three treatments in a randomized block study (a, n = 2; b, n = 2; c, n = 2). In phase one, group a received acetaminophen intravenously, group b and c orally after being fasted and fed, respectively. In phase two and three, groups were swapped, and the experiment was repeated. At the end of the trial, each dog received the same treatment. Acetaminophen plasma concentrations were detected using a validated HPLC‐UV method. The pharmacokinetic analysis was performed using a noncompartmental model. Clearance, volume at steady state and half‐life of acetaminophen in Labrador Retrievers were 0.42 L/kg hr, 0.87 L/kg and 1.35 hr, respectively. No significant statistical differences were found between fasted and fed dogs regarding maximum plasma concentration, time at maximum concentration and bioavailability as measured by the AUC. Background: Interleukin-1β(IL-1β) is important mediator of inflammatory-induced suppression of reproductive axis at the hypothalamic level. At the beginning of inflammation, the main source of cytokines in the cerebrospinal fluid(CSF) is peripheral circulation, while over time, cytokines produced in the brain are more important. Melatonin has been shown to decrease pro-inflammatory cytokines concentration in the brain. In ewes, melatonin is used to advance the onset of a breading season. Little is known about CSF concentration of IL-1β in ewes and its correlation with plasma during inflammation as well as melatonin action on the concentration of IL-1β in blood plasma and the CSF, and brain barriers permeability in early stage of lipopolysaccharide(LPS)-induced inflammation.Methods: Systemic inflammation was induced through LPS administration in melatonin-and sham-implanted ewes. Blood and CSF samples were collected before and after LPS administration and IL-1β and albumin concentration were measured. To assess the functions of brain barriers albumin quotient(QAlb) was used.Expression of IL-1β(Il1B) and its receptor type Ⅰ(Il1r1) and type Ⅱ(Il1r2) and matrix metalloproteinase(Mmp) 3 and 9 was evaluated in the choroid plexus(CP).Results: Before LPS administration, IL-1β was on the level of 62.0 ± 29.7 pg/mL and 66.4 ± 32.1 pg/mL in plasma and 26.2 ± 5.4 pg/mL and 21.3 ± 8.7 pg/mL in the CSF in sham-and melatonin-implanted group, respectively.Following LPS it increased to 159.3 ± 53.1 pg/mL and 197.8 ± 42.8 pg/mL in plasma and 129.8 ± 54.2 pg/mL and139.6 ± 51.5 pg/mL in the CSF. No correlations was found between plasma and CSF IL-1β concentration after LPS in both groups. The QAlb calculated before LPS and 6 h after was similar in all groups. Melatonin did not affected m RNA expression of Il1B, Il1r1 and Il1r2 in the CP. The m RNA expression of Mmp3 and Mmp9 was not detected.Conclusions: The lack of correlation between plasma and CSF IL-1β concentration indicates that at the beginning of inflammation the local synthesis of IL-1β in the CP is an important source of IL-1β in the CSF. Melatonin from slow-release implants does not affect IL-1β concentration in plasma and CSF in early stage of systemic inflammation. 相似文献

Metamizole (dipyrone, MET) is a nonopioid analgesic drug commonly used in human and veterinary medicine. The aim of this study was to assess two major active metabolites of MET, 4‐methylaminoantipyrin (MAA) and 4‐aminoantipyrin (AA), in goat plasma after intravenous (IV) and intramuscular (IM) administration. In addition, metabolite concentration in milk was monitored after IM injection. Six healthy female goats received MET at a dose of 25 mg/kg by IV and IM routes in a crossover design study. The blood and milk samples were analyzed using HPLC coupled with ultraviolet detector and the plasma vs concentration curves analyzed by a noncompartmental model. In the goat, the MET rapidly converted into MAA and the mean maximum concentration was 183.97 μg/ml (at 0.08 hr) and 51.94 μg/ml (at 0.70 hr) after IV and IM administration, respectively. The area under the curve and mean residual time values were higher in the IM than the IV administered goats. The average concentration of AA was lower than MAA in both groups. Over 1 μg/ml of MAA was found in the milk (at 48 hr) after MET IM administration. In conclusion, IM is considered to be a better administration route in terms of its complete absorption with long persistence in the plasma. However, this therapeutic option should be considered in light of the likelihood of there being milk residue. 相似文献

Tree structural diversity is assessed by modelling stand diameter at breast height (DBH) distribution. The aim of this study
was to verify the suitability of a mixture of two- and three-component Weibull and gamma models for describing irregular and
multimodal DBH distributions. Investigations were carried out in natural Abies alba Mill. and Fagus sylvatica L. stands, representing the growing-up stage, in the Świętokrzyski National Park (Central Poland) and in the Pieniny National
Park (Southern Poland). Sample plots (0.25–0.5 ha in area) were selected from a database used for forest dynamics analysis.
The parameters of the mixture model were estimated using maximum likelihood estimation (MLE) and a combination of an expectation–maximisation
(EM) algorithm with a Newton-type (NT) method. The multistart method (MM) of choosing initial values was used. The DBH distributions
in mixed Abies–Fagus forests of complex structure were accurately approximated by two- and three-component mixture models. The root mean square
error (RMSE) values varied from 3.4515 to 7.6682 for the Weibull mixture and from 3.6457 to 7.1419 for the gamma mixture.
The results indicated that the mixture Weibull and gamma models were, in general, equally suitable for modelling irregular
and multimodal DBH distributions. Finite mixture models may be used to characterise tree structural diversity, especially
in uneven-aged stands of complex structure. 相似文献

Increasing demand for renewable energy and limiting CO2 emissions have stimulated much interest in wood-based biofuels. Unfortunately, expanding the utilization of forest biomass may cause nutrient depletion in forested environments. This study investigates the element content from various parts of the tree. Comparisons were made between different harvesting scenarios and their impact on the amount of nutrients removed from the forest environment. The harvesting scenarios were: stem-only harvesting (SOH), branch and stem harvesting (BSH), and two variants of whole-tree harvesting (WTH). The elements taken into account were: carbon (C), nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), sodium (Na), sulphur (S), copper (Cu), iron (Fe), nickel (Ni), manganese (Mn), zinc (Zn), cadmium (Cd), chromium (Cr) and lead (Pb). To make the results comparable, the same amount of removed biomass was taken into calculation. The differences between harvesting scenarios were significant. The amounts of removed elements formed similar pattern: the lowest level was found in SOH, average in BSH, and highest in both variants of WTH. This confirms that the application of WTH is connected with increased risk of nutrient depletion, even when the volume of harvest would be equal to other variants. 相似文献

The oak (Quercus robur L.) regeneration intensity was assessed in the core area of the Białowieża National Park (BNP) in Poland with respect to
the selected ecological factors. The emphasis was placed on the response of oak regeneration to disturbances, including the
large-scale dieback of spruce stands. Defining their effect could help predicting the role of oak in naturally developing
lowland forest ecosystems in the European hemiboreal zone. The results of the study challenge the opinion that the ‘lime-oak-hornbeam
forest’ is a ‘climax’ community, confirming a very poor regeneration represented by only two saplings taller than 0.5 m per
hectare. By contrast, in spruce-dominated communities, from 49 to 848 taller saplings per hectare were found. The occurrence
of saplings was associated with discontinuous canopy of late seral stage of stands, as well as with large gaps. Most of the
best quality grown-up oak saplings developed in the immediate neighbourhood of spruce logs. The results of the research indicate
that ‘lime-oak-hornbeam forest’ (Tilio-Carpinetum) should be rather perceived as a transient community, evolved from relict, culturally modified, oak woodlands. Because spruce
had become a dominating species only after abandoning in mid-1800s the historical regime of anthropogenic disturbances (involving
frequent forest fires), the observed phenomena related to the disintegration of spruce stands had probably no precedent over
the last 500 years. To confirm whether the massive decline of spruce stands will finally result in the successful establishment
of the new canopy oak generation, both in Białowieża and other forests of hemiboreal zone, further research is needed. 相似文献
