A simple method for the analysis of major wine volatiles and semivolatiles by stir bar sorptive extraction in combination with thermal desorption and gas chromatography-mass spectrometry (SBSE-TD-GC-MS) was developed. Significant experimental parameters such as extraction time, temperature, salt addition, pH, and thermal desorption parameters were optimized to provide a sensitive and robust analytical method. The method provided good repeatability (%RSD < 10%) for 38 major wine volatile compounds, including alcohols, acids, esters, phenols, aldehydes, ketones, and lactones. Quantitative data for 62 South African red and white wines were used to study the suitability of major volatile data for the differentiation of wine samples according to grape variety or cultivar. Principal component analysis (PCA) and cluster analysis (CA) showed that most of the variation in volatile composition between wine samples could be ascribed to differences in wine age, wood contact, and fermentation practices. Commercial cultivation of Bt cotton produced higher boll load which led to stiff inter-original competition for photosynthates, resulting in early cessation of growth (premature senescence) due to more availability of sink and less sources. To overcome this problem, field experiment was conducted during 2011 and 2012 using five treatments of plant growth manipulation viz. no fruiting branch removal (F1), removal of first fruiting branch (F2), removal of first and second fruiting branch (F3), removal of all squares from first fruiting branch (F4), removal of all squares from first and second fruiting branches (F5), and three potassium (K) application rates viz. 50 kg ha?1 (K1), 100 kg ha?1 (K2), and 150 kg ha?1 (K3). More nodes above white flower were recorded in F5, followed by F3, while minimum were recorded in F1. Among potassium levels, maximum nodes above white flower were recorded in K3 followed by K2 and K1 during both years of study. Plant height recorded at physiological cutout stage or at maturity stage showed that plants gained more height with removal of all squares from first tosecond fruiting branches with higher potassium dose. Leaf K increased with increasing applied potassium and also with square/branch removal. So early removal of squares/fruiting branches along with higher potassium dose helped in delaying canopy senescence in Bt cotton. 相似文献
Nutrients constrain the soil carbon cycle in tropical forests, but we lack knowledge on how these constraints vary within the soil microbial community. Here, we used in situ fertilization in a montane tropical forest and in two lowland tropical forests on contrasting soil types to test the principal hypothesis that there are different nutrient constraints to different groups of microorganisms during the decomposition of cellulose. We also tested the hypotheses that decomposers shift from nitrogen to phosphorus constraints from montane to lowland forests, respectively, and are further constrained by potassium and sodium deficiency in the western Amazon. Cellulose and nutrients (nitrogen, phosphorus, potassium, sodium, and combined) were added to soils in situ, and microbial growth on cellulose (phospholipid fatty acids and ergosterol) and respiration were measured. Microbial growth on cellulose after single nutrient additions was highest following nitrogen addition for fungi, suggesting nitrogen as the primary limiting nutrient for cellulose decomposition. This was observed at all sites, with no clear shift in nutrient constraints to decomposition between lowland and montane sites. We also observed positive respiration and fungal growth responses to sodium and potassium addition at one of the lowland sites. However, when phosphorus was added, and especially when added in combination with other nutrients, bacterial growth was highest, suggesting that bacteria out-compete fungi for nitrogen where phosphorus is abundant. In summary, nitrogen constrains fungal growth and cellulose decomposition in both lowland and montane tropical forest soils, but additional nutrients may also be of critical importance in determining the balance between fungal and bacterial decomposition of cellulose. 相似文献
Estimating the relative importance of habitat loss and fragmentation is necessary to estimate the potential benefits of specific
management actions and to ensure that limited conservation resources are used efficiently. However, estimating relative effects
is complicated because the two processes are highly correlated. Previous studies have used a wide variety of statistical methods
to separate their effects and we speculated that the published results may have been influenced by the methods used. We used
simulations to determine whether, under identical conditions, the following 7 methods generate different estimates of relative
importance for realistically correlated landscape predictors: residual regression, model or variable selection, averaged coefficients
from all supported models, summed Akaike weights, classical variance partitioning, hierarchical variance partitioning, and
a multiple regression model with no adjustments for collinearity. We found that different methods generated different rankings
of the predictors and that some metrics were strongly biased. Residual regression and variance partitioning were highly biased
by correlations among predictors and the bias depended on the direction of a predictor’s effect (positive vs. negative). Our
results suggest that many efforts to deal with the correlation between amount and fragmentation may have done more harm than
good. If confounding effects are controlled and adequate thought is given to the ecological mechanisms behind modeled predictors,
then standardized partial regression coefficients are unbiased estimates of the relative importance of amount and fragmentation,
even when predictors are highly correlated. 相似文献
Potato psyllid, Bactericera cockerelli (?ulc), causes economic damage to potato crops throughout the major potato growing regions of western North America. When cultivated crops are not available, potato psyllid often occurs on non-crop hosts. In the southern U.S. and northern Mexico, native species of Lycium (Solanaceae) are important non-crop hosts for the psyllid. We determined whether Old World species of Lycium now widespread in the Pacific Northwest are reservoirs of potato psyllid in this growing region. We examined Lycium spp. across a wide geographic region in Washington, Oregon, and Idaho at irregular intervals during three growing seasons. Potato psyllids were present at all locations. To determine whether Lycium is also a host during intervals of the year in which the potato crop is not available, we monitored a subset of these sites over the entire year. Six sites were monitored at 1- to 3-week intervals from June 2014 to June 2016. Psyllids were present on Lycium throughout the year at all sites, including during winter, indicating that Lycium is also a host when the potato crop is seasonally not available. Psyllid populations included a mixture of Northwestern and Western haplotypes. We observed well-defined spring and fall peaks in adult numbers, with peaks separated by long intervals in which psyllid numbers were very low. Seasonal patterns in psyllid numbers on these non-native Lycium hosts were very similar to what has been observed on native Lycium in the desert southwest region of the U.S. Our findings demonstrate that potato psyllid associates with Lycium across a broad geographic region within the Pacific Northwest. These results will assist in predicting sources of potato psyllid colonizing potatoes in this important growing region. 相似文献

Lack of quantitative observations of extent, frequency, and severity of large historical fires constrains awareness of departure of contemporary conditions from those that demonstrated resistance and resilience to frequent fire and recurring drought.
Objectives
Compare historical and contemporary fire and forest conditions for a dry forest landscape with few barriers to fire spread.
Methods
Quantify differences in (1) historical (1700–1918) and contemporary (1985–2015) fire extent, fire rotation, and stand-replacing fire and (2) historical (1914–1924) and contemporary (2012) forest structure and composition. Data include 85,750-ha tree-ring reconstruction of fire frequency and extent; >?375,000-ha timber inventory following >?78,900-ha fires in 1918; and remotely-sensed maps of contemporary fire effects and forest conditions.
Results
Historically, fires?>?20,000 ha occurred every 9.5 years; fire rotation was 14.9 years; seven fires?>?40,469 ha occurred during extreme drought (PDSI <?? 4.0); and stand-replacing fire occurred primarily in lodgepole (Pinus contorta var. murrayana). In contemporary fires, only 5% of the ecoregion burned in 30 years, and stand-replacing fire occurred primarily in ponderosa (Pinus ponderosa) and mixed-conifer. Historically, density of conifers?>?15 cm dbh exceeded 120 trees/ha on?<?5% of the area compared to 95% currently.
Conclusions
Frequent, large, low-severity fires historically maintained open-canopy ponderosa and mixed-conifer forests in which large fire- and drought-tolerant trees were prevalent. Stand-replacing patches in ponderosa and mixed-conifer were rare, even in fires >?40,469 ha (minimum size of contemporary “megafires”) during extreme drought. In this frequent-fire landscape, mixed-severity fire historically influenced lodgepole and adjacent forests. Lack of large, frequent, low-severity fires degrades contemporary forest ecosystems.
The characterization of herbal materials is a significant challenge to analytical chemists. Goldenseal (Hydrastis canadensis L.), which has been chosen for toxicity evaluation by NIEHS, is among the top 15 herbal supplements currently on the market and contains a complex mixture of indigenous components ranging from carbohydrates and amino acids to isoquinoline alkaloids. One key component of herbal supplement production is botanical authentication, which is also recommended prior to initiation of efficacy or toxicological studies. To evaluate material available to consumers, goldenseal root powder was obtained from three commercial suppliers and a strategy was developed for characterization and comparison that included Soxhlet extraction, HPLC, GC-MS, and LC-MS analyses. HPLC was used to determine the weight percentages of the goldenseal alkaloids berberine, hydrastine, and canadine in the various extract residues. Palmatine, an isoquinoline alkaloid native to Coptis spp. and other common goldenseal adulterants, was also quantitated using HPLC. GC-MS was used to identify non-alkaloid constituents in goldenseal root powder, whereas LC-MS was used to identify alkaloid components. After review of the characterization data, it was determined that alkaloid content was the best biomarker for goldenseal. A 20-min ambient extraction method for the determination of alkaloid content was also developed and used to analyze the commercial material. All three lots of purchased material contained goldenseal alkaloids hydrastinine, berberastine, tetrahydroberberastine, canadaline, berberine, hydrastine, and canadine. Material from a single supplier also contained palmatine, coptisine, and jatrorrhizine, thus indicating that the material was not pure goldenseal. Comparative data for three commercial sources of goldenseal root powder are presented. 相似文献
