Intense selection for speed, endurance or pulling power in the domestic horse (Equus caballus) has resulted in a number of adaptive changes in the phenotype required for elite athletic performance. To date, studies in humans have revealed a large number of genes involved in elite athletic performance, but studies in horses are rare. The horse genome assembly and bioinformation tools for genome analyses have been used to compare human performance genes with their equine orthologues, both to retrieve pathways for these genes and to investigate their chromosomal distribution. In this review, 28 candidate genes for equine performance are presented that have polymorphisms associated with human elite athletic performance and may have impact on athletic performance in horses. A significant accumulation of candidate genes was found on horse chromosomes 4 and 12. Genes involved in pathways for focal adhesion, regulation of actin cytoskeleton, neuroactive ligand-receptor interaction, and calcium signalling were over-represented. Genome-wide association studies for athletic performance in horses may benefit from the strong conserved synteny of the chromosomal arrangement of genes in humans and horses. 相似文献

Seizures or convulsions that occur during anaesthesia in veterinary patients are infrequently reported in the literature. Consequently, the incidence of such events is unknown. Several drugs commonly used in clinical veterinary anaesthesia have been shown to induce epileptiform activity in both human clinical patients and experimental candidates. The present case report describes convulsions in a four-year old male Bernese mountain dog during maintenance of anaesthesia with isoflurane after premedication with acepromazine and methadone followed by co-induction with propofol and ketamine. The dog had no history of previous convulsions. The use of several sedative and anaesthetic drugs makes it difficult to find one single causative pharmaceutical. 相似文献

Microglia are the principal immune effector elements of the brain sharing immunophenotypic and functional characteristics of macrophages as well as of antigen presenting cells (APCs). The purpose of this study was to isolate canine microglial cells and make them available for ex vivo characterizations of their functions and immunophenotype. After isolation, carried out by density gradient centrifugation, microglial cells accumulated on distinct interfaces of 1.077 and 1.066 g/ml of a Percoll gradient. Identification of microglial cells in other species is realized by their specific immunophenotype of CD11b/c+ and CD45low. Our results indicate, that expression of CD45 is very low or even absent in canine microglial cells. In addition, they expressed CD18 and CD11b/c+, as determined by flow cytometry and immunohistochemistry. Fourteen additional monoclonal antibodies (mAbs) were used to characterize and compare canine microglial cells with monocytes. Microglia and monocytes can be clearly distinguished by their differential expression intensity of surface antigens (CD45, CD44, CD14). Functional characterization was assessed by a reactive oxygen species (ROS)-generation test and phagocytosis assay using flow cytometry. In conclusion, ex vivo examination of microglia is possible in dogs and most probably reflects the conditions in vivo. The measurement of tissue culture artifacts can be largely avoided using this method. 相似文献

This study reports and analyzes nutrient balances in experimental vegetable production systems of the two West African cities of Tamale (Ghana) and Ouagadougou (Burkina Faso) over a two‐year period comprising thirteen and eleven crops, respectively. Nutrient‐use efficiency was also calculated. In Tamale and Ouagadougou, up to 2% (8 and 80 kg N ha?1) of annually applied fertilizer nitrogen were leached. While biochar application or wastewater irrigation on fertilized plots did not influence N leaching in both cities, P and K leaching, as determined with ion‐absorbing resin cartridges, were reduced on biochar‐amended plots in Tamale. Annual nutrient balances amounted to +362 kg N ha?1, +217 kg P ha?1, and –125 kg K ha?1 in Tamale, while Ouagadougou had balances of up to +692 kg N ha?1, +166 kg P ha?1, and –175 kg K ha?1 y?1. Under farmers' practice of fertilization, agronomic nutrient‐use efficiencies were generally higher in Tamale than in Ouagadougou, but declined in both cities during the last season. This was the result of the higher nutrient inputs in Ouagadougou compared to Tamale and relatively lower outputs. The high N and P surpluses and K deficits call for adjustments in local fertilization practices to enhance nutrient‐use efficiency and prevent risks of eutrophication. 相似文献

Guanabenz (2,6-dichlorobenzylidene-amino-guanidine) is a centrally acting antihypertensive drug whose mechanism of action is via alpha2 adrenoceptors or, more likely, imidazoline receptors. Guanabenz is marketed as an antihypertensive agent in human medicine (Wytensin tablets, Wyeth Pharmaceuticals). Guanabenz has reportedly been administered to racing horses and is classified by the Association of Racing Commissioners International as a class 3 foreign substance. As such, its identification in a postrace sample may result in significant sanctions against the trainer of the horse. The present study examined liquid chromatographic/tandem quadrupole mass spectrometric (LC-MS/MS) detection of guanabenz in serum samples from horses treated with guanabenz by rapid i.v. injection at 0.04 and 0.2 mg/kg. Using a method adapted from previous work with clenbuterol, the parent compound was detected in serum with an apparent limit of detection of approximately 0.03 ng/ml and the limit of quantitation was 0.2 ng/ml. Serum concentrations of guanabenz peaked at approximately 100 ng/ml after the 0.2 mg/kg dose, and the parent compound was detected for up to 8 hours after the 0.04 mg/kg dose. Urine samples tested after administration of guanabenz at these dosages yielded evidence of at least one glucuronide metabolite, with the glucuronide ring apparently linked to a ring hydroxyl group or a guanidinium hydroxylamine. The LC-MS/MS results presented here form the basis of a confirmatory test for guanabenz in racing horses. 相似文献

Zusammenfassung Pollen vonArabidopsis thaliana wurde Röntgenbestrahlungen von 2 bis 32 kr ausgesetzt. Für die Frequenz der dominanten Letalmutationen ergab sich eine exponentielle Dosis-Effekt-Beziehung mit einem Maximalwert von 60,2%. Nach Bestrahlung mit 16 kr wurden gefunden: dominante Letalmutationen in 23%, rezessive Letalmutationen in rund 28% und haplophasische Letalmutationen in rund 51% der geprüften Genome. Die rezessiven Letalmutationen glichen in ihren Manifestierungsmustern den nach Röntgenbestrahlung von Samen auftretenden rezessiven Letalmutationen.
Summary Pollen grains ofArabidopsis thaliana were X-rayed with doses of 2 kr up to 32 kr. The frequency of dominant lethals showed an exponential dose-response-relationship with a maximum value of 60.2 per cent. After irradiation with 16 kr dominant lethals were found in 23 per cent, recessive lethals in 28 per cent, and haplophasic lethals in 51 per cent of the genomes tested. The patterns of manifestation of the receissve lethals were identical with those found after X-raying of seeds.

The insulin-like growth factor-II (IGF-II) and the IGF-II/mannose-6-phosphate (M6P) receptor are thought to play an important role in fetal growth and development. We have studied the expression of the IGF-II/M6P receptor in fetal bovine tissues from 5 through 36 weeks' gestation. Tissues from bovine fetuses were extracted in buffer containing 2% Triton-X-100 and 2% sodium dodecyl sulfate (SDS). Aliquots of the protein extracts were analyzed by SDS polyacrylamide gel electrophoresis and the protein bands were transferred onto nitrocellulose. Immunoblotting was performed with anti-bovine IGF-II/M6P receptor antiserum. In a subset of experiments, ligand blotting was carried out with radiolabeled IGF-II and subsequent autoradiography. IGF-II/M6P receptors were expressed in all tissues examined, with the highest amount of receptor being present in fetal lung and liver. Low amounts of receptors were measured in fetal brain. The amount of receptor was developmentally regulated throughout fetal life. The developmental regulation of receptor expression varied among the different tissues. In conclusion, the IGF-II/M6P receptor is present in all fetal bovine tissues examined. The presence of the IGF-II/M6P receptor seems to be developmentally regulated during bovine fetal life. We hypothesize that this receptor exerts important biologic effects during fetal growth and tissue and organ development. 相似文献
