Edible coatings based on high molecular weight chitosan, pure or combined with methylcellulose or oleic acid, were applied to fresh-cut carrots cv. Nantesa by simple immersion and by applying a vacuum pulse (5 kPa for 4 min). Water vapour resistance, colour, mechanical properties and respiration rates of non-coated and coated samples were determined. Coatings improved sample appearance, since they diminished the occurrence of the white blush during storage. When applied by simple immersion, they neither conferred significant barrier properties nor the preservation of the mechanical properties of fresh-cut carrot samples. In contrast, coating application with a vacuum pulse enhanced all the positive effects, since the resistance of water vapour transmission of the samples was significantly improved, and better preservation of the sample colour and mechanical response during cold storage was obtained. Differences in film composition did not significantly affect the coating behaviour, probably due to the variability induced by different factors when coatings were applied to the carrot surface. 相似文献
Crude extracts and fractions of Bertholletia excelsa stem barks were tested for trypanocidal activity. ent-kaurenoic acid (KA) isolated from Viguiera robusta was tested for activity on vascular smooth muscle contactility. Cumulative concentration-response curves were obtained for a stepwise increase (10(-10)-10(-05) mol/l) in the concentration of phenylephrine (Phe), a selective alpha(1)-adrenergic agonist. The effects in the presence of KA (0.2, 2.0 and 20.0 microg/ml), and 90 min after the removal of KA from the medium bath were compared to controls. At 20.0 microg/ml, KA inhibited the in vitro contractility of rat carotid artery elicited by Phe, but had no effect at lower concentrations (0.2 and 2.0 microg/ml). The amount of photosynthetic radiation intercepted by a crop is a function of the incident solar radiation on the plants, the leaf area index (LAI), and the light extinction coefficient (k). We quantified LAI and k in stands of black wattle (Acacia mearnsii De Wild.) over a 7-year growth cycle at two locations in the state of Rio Grande do Sul, Brazil. Our study was conducted in commercial stands in agroecological regions with high densities of black wattle plantations. LAI was calculated as the ratio between the leaf area of a tree and its planting space, and k was derived from Beer’s law. LAI depends on the planting site and stand age. Between the two sites, the LAI was similar over time, the amount of variation differed. Values of k depended only on stand age, with the highest average observed for stands up to 5 years old. The trend of k during the plantation cycle was inversely proportional to LAI and was correlated with LAI, leaf area, leaf dry mass, canopy volume, height, branches dry mass, total dry mass, and crown diameter. 相似文献
To prevent, detect, and protect against forest fires, forest personnel need to define rules for determining forest fire risk. In Portugal, all municipalities must annually produce forest fire risk(FFR) maps. To produce more reliable FFR maps more easily, we developed an open source model using the Modeler plugin of SEXTANTE in the program QGIS version 2.0 Dufour. The model provides all the maps involved in the FFR model(susceptibility map, hazard map, vulnerability map, economic value map,and potential loss map) and was produced according to Portuguese Forest Authority's(AFN, Autoridade Florestal Nacional) rules for determining the FFR. This model was tested for the Portuguese municipality Santa Maria da Feira, where 40 % of the total municipality area falls in the category ‘‘very high' or ‘‘high' fire risk. The ‘‘very high'fire risk area is mainly classified as broad-leaved forest and has the steepest slopes(15 %). The distance of burned areas to roads was also analyzed; the proportion of burned areas increased with increasing distance to the main roads.In addition, 92.6 % of the ‘‘high' and ‘‘very high' risk zones were located in areas with lower elevation. These results confirmed that forest fire is strongly influenced not only by environmental factors but also by anthropogenic factors. The procedure implemented here was compared with our open source application already available in QGIS and also to the same procedure implemented in GIS proprietary software. Although the results were obviously the same, the model developed here presents several advantages over the other two approaches. Besides being faster,it is easy to change the model parameters according to user needs(i.e., to the rules of different countries), and can be modified and adapted to other variables and other areas to create risk maps for different natural phenomena(e.g.,floods, earthquakes, landslides). The model is easy to use and to create risk and hazard maps rapidly in a free, open source environment that does not require any programming knowledge. 相似文献
It has been postulated that osmotic effects on plant tissue are mediated by abscisic acid (ABA). Hybrid larch (Larix kaempferi (Lambert) Carr. x L. decidua Mill.) plantlets, transformed with the ABA-inducible wheat Em promoter associated with the Gus reporter gene, were axenically inoculated with two ectomycorrhizal fungi: Cenococcum geophilum Fr., considered tolerant to water stress, and Laccaria bicolor (Marie) Orton, considered less tolerant to drought. The mycorrhizal and non-mycorrhizal transgenic plantlets were subjected to osmotic stress by adding polyethylene glycol (PEG) to the culture medium. In the presence of PEG, L. bicolor and C. geophilum reduced shoot water potential and turgor potential, but increased host osmotic potential. Treatment of plantlets with PEG induced a significant increase in endogenous ABA concentrations. Laccaria bicolor and C. geophilum behaved similarly and significantly decreased the ABA response of plantlets to PEG treatment. Moreover, inoculation with either fungus regulated the ABA response of the plantlets even when the fungus was separated from the host by a cellophane sheet that prevented mycorrhiza formation. Although the wheat Em promoter was inducible in larch plantlets, it was not regulated by endogenous ABA. Induction of the wheat Em promoter in larch plantlets depended on organ type, with maximum induction in the root apex. In the present study a technique was developed with the aim of guaranteeing the composition and security of fish meals, since it allows verification of whether these meals contain land animal remains. The method is based on polymerase chain reaction (PCR) and length polymorphism, followed by a restriction fragment length polymorphism (RFLP). Specific primers for every species were designed and calibrated, generating exclusively a PCR product with a specific size when DNA for each species was present in the sample. This technique allows the detection of land animal remains in fish meals, specifically cow, chicken, pig, horse, sheep, and goat. The identity of the PCR products can be confirmed by RFLP analysis using only one restriction enzyme. The selected restrictase generated one characteristic restriction profile for every species included in this study. The detection limit of this method was calculated by using mixtures of fish meals in different proportions and meal that exclusively contained remains of one of these land species studied. The analytical strategy herein proposed was applied to fish and meat meals, giving good results, both in the analyzed standards and in commercial samples. 相似文献
The commercialization of animal feeds infected by prions proved to be the main cause of transmission of bovine spongiform encephalopathy (BSE). Therefore, feed bans were enforced, initially for ruminant feeds, and later for all feeds for farmed animals. The development and validation of analytical methods for the species-specific detection of animal proteins in animal feed has been indicated in the TSE (Transmissible Spongiform Encephalopathies) Roadmap (European Commission. Methods based on polymerase chain reaction (PCR) seem to be a promising solution for this aim. The main objective of this study was to determine the applicability of four different real-time PCR methods, developed by three National expert laboratories from the European Union (EU), for the detection and identification of cattle or ruminant species in typical compound feeds, fortified with meat and bone meals (MBM) from different animal species at different concentration levels. The MBM samples utilized in this study have been treated using the sterilization condition mandatory within the European Union (steam pressure sterilization at 133 degrees C, 3 bar, and 20 min), which is an additional challenge to the PCR methods evaluated in this study. The results indicate that the three labs applying their PCR methods were able to detect 0.1% of cattle MBM, either alone or in mixtures with different materials such as fishmeal, which demonstrates the improvement made by this technique, especially when compared with results from former interlaboratory studies. 相似文献
A feeding experiment was carried out to determine the efficiency of different commercial sources, chemical forms and levels, of dietary astaxanthin, to appropriately pigment the red porgy (Pagrus pagrus) skin. According to this, total carotenoid content, profiles and chemical forms present in the skin were determined. In order to establish the potential for antioxidant protecting role of astaxanthin supplemented diets, peroxide levels and lipid composition of skin were also determined.
Red porgy alevins were fed six dietary treatments in triplicate; a basal diet (B) without carotenoids; two diets (N25 and N50) formulated to supply either 25 or 50 mg kg− 1 of an esterified source of astaxanthin (Haematococcus pluvialis, NatuRose™); two diets (CP25 and CP50) with either 25 or 50 mg kg− 1 of unesterified astaxanthin (Carophyll® Pink); and a positive control diet (B + S) proved as a successful pigmenting-diet in previous experiences (B + S, 88% basal diet:12% frozen shrimp) [Cejas, J., Almansa, E., Tejera, N., Jerez, S., Bolaños, A., Lorenzo, A., 2003. All fish fed carotenoid supplemented diets displayed a pink-coloured skin after 4 months of feeding in contrast to the greyish appearance displayed by fish fed the basal diet not supplemented with carotenoids (B). Furthermore, astaxanthin diesters were the major carotenoid in the skin of pink fish. A second carotenoid, tentatively identified as tunaxanthin diester, was also detected. The best results in terms of skin natural reddish hue, total carotenoid and astaxanthin contents were found by using the esterified forms of dietary astaxanthin (N25, N50 and B + S). Interestingly, the lowest levels of lipid peroxides were found in the fish fed these three treatments. However, no effect of treatment on lipid composition was found. In conclusion, red porgy alevins are able to efficiently utilise dietary natural or synthetic astaxanthin, and deposit this pigment in its esterified form to acquire an acceptable pink-coloured skin compared to that of the wild fish. 相似文献
