Regulation of bone mass in mice by the lipoxygenase gene Alox15

The development of osteoporosis involves the interaction of multiple environmental and genetic factors. Through combined genetic and genomic approaches, we identified the lipoxygenase gene Alox15 as a negative regulator of peak bone mineral density in mice. Crossbreeding experiments with Alox15 knockout mice confirmed that 12/15-lipoxygenase plays a role in skeletal development. Pharmacologic inhibitors of this enzyme improved bone density and strength in two rodent models of osteoporosis. These results suggest that drugs targeting the 12/15-lipoxygenase pathway merit investigation as a therapy for osteoporosis.     

Beryllium: Effect of Ultra-High Pressure on Resistance

Beryllium, subjected to a pressure of 93 kb at 25 degrees C undergoes a change in structure, as indicated by a 45-percent decrease in resistance. At 55 kb a small change in the resistance-pressure relationship suggests another modification of the usual hexagonal-close-packed structure.     

Little evidence for developmental plasticity of adult hematopoietic stem cells

To rigorously test the in vivo cell fate specificity of bone marrow (BM) hematopoietic stem cells (HSCs), we generated chimeric animals by transplantation of a single green fluorescent protein (GFP)-marked HSC into lethally irradiated nontransgenic recipients. Single HSCs robustly reconstituted peripheral blood leukocytes in these animals, but did not contribute appreciably to nonhematopoietic tissues, including brain, kidney, gut, liver, and muscle. Similarly, in GFP+:GFP- parabiotic mice, we found substantial chimerism of hematopoietic but not nonhematopoietic cells. These data indicate that "transdifferentiation" of circulating HSCs and/or their progeny is an extremely rare event, if it occurs at all.     

Constraints on neon and argon isotopic fractionation in solar wind

To evaluate the isotopic composition of the solar nebula from which the planets formed, the relation between isotopes measured in the solar wind and on the Sun's surface needs to be known. The Genesis Discovery mission returned independent samples of three types of solar wind produced by different solar processes that provide a check on possible isotopic variations, or fractionation, between the solar-wind and solar-surface material. At a high level of precision, we observed no significant inter-regime differences in 20Ne/22Ne or 36Ar/38Ar values. For 20Ne/22Ne, the difference between low- and high-speed wind components is 0.24 +/- 0.37%; for 36Ar/38Ar, it is 0.11 +/- 0.26%. Our measured 36Ar/38Ar ratio in the solar wind of 5.501 +/- 0.005 is 3.42 +/- 0.09% higher than that of the terrestrial atmosphere, which may reflect atmospheric losses early in Earth's history.     

Human-specific gain of function in a developmental enhancer

Changes in gene regulation are thought to have contributed to the evolution of human development. However, in vivo evidence for uniquely human developmental regulatory function has remained elusive. In transgenic mice, a conserved noncoding sequence (HACNS1) that evolved extremely rapidly in humans acted as an enhancer of gene expression that has gained a strong limb expression domain relative to the orthologous elements from chimpanzee and rhesus macaque. This gain of function was consistent across two developmental stages in the mouse and included the presumptive anterior wrist and proximal thumb. In vivo analyses with synthetic enhancers, in which human-specific substitutions were introduced into the chimpanzee enhancer sequence or reverted in the human enhancer to the ancestral state, indicated that 13 substitutions clustered in an 81-base pair module otherwise highly constrained among terrestrial vertebrates were sufficient to confer the human-specific limb expression domain.     

Polar lightning and decadal-scale cloud variability on Jupiter

Although lightning has been seen on other planets, including Jupiter, polar lightning has been known only on Earth. Optical observations from the New Horizons spacecraft have identified lightning at high latitudes above Jupiter up to 80 degrees N and 74 degrees S. Lightning rates and optical powers were similar at each pole, and the mean optical flux is comparable to that at nonpolar latitudes, which is consistent with the notion that internal heat is the main driver of convection. Both near-infrared and ground-based 5-micrometer thermal imagery reveal that cloud cover has thinned substantially since the 2000 Cassini flyby, particularly in the turbulent wake of the Great Red Spot and in the southern half of the equatorial region, demonstrating that vertical dynamical processes are time-varying on seasonal scales at mid- and low latitudes on Jupiter.     

Effectiveness of conservation easements for reducing development and maintaining biodiversity in sagebrush ecosystems

Conservation easements are the primary tool used globally by land trusts and governmental agencies to achieve conservation goals on private lands, but empirical evaluations of their effectiveness are lacking. Here we compared biodiversity in sagebrush ecosystems and recent rates of change in road and structure densities on properties with and without easements held by The Nature Conservancy in Wyoming, USA. To determine whether any differences between properties with and without easements were better explained by development pressure or by management practices, properties were stratified by low versus high residential development pressure and property managers were surveyed. In areas with high development pressure, easement properties had fewer structures and tended to have fewer, smaller roads than properties without easements. In the high-pressure areas, properties with easements also had greater use by some wildlife species than properties without easements. Regardless of easement presence, there was higher cover of exotic plant species and fewer mammal burrows in high than low-pressure areas. There were no significant differences in land management practices between properties with and without easements, but managers of properties with easements tended to seek management support more often than other managers. This may present an opportunity to provide support for specific management activities on easements to ensure that they continue to meet intended goals. Given the importance of easements as an alternative to nature reserves and the significant investment being made to acquire additional easements, it is essential to continue to evaluate whether easements are an effective tool in other locations.
