Microbial communities in soils provide numerous functions vital to agricultural productivity, and there is growing interest in understanding and manipulating these communities to achieve more sustainable production systems. We investigated microbial communities in potato fields in Wisconsin to determine the extent to which microbial communities were structured by inherent soil properties versus factors under grower control. We found that soil properties, including texture and chemistry, shaped microbial communities at broad levels, determining the diversity and abundance of phyla. Organic versus conventional management, however, correlated with finer scale differences in microbial communities, while crop variety was not associated with microbial community composition in this study. These results suggest that grower practices have the potential to alter microbial communities, but these effects will take place within an existing soil context. Future research is necessary to determine how these patterns in microbial community structure relate to functional outcomes for plant health.  相似文献

Metabolic reprogramming has been proposed to be a hallmark of cancer, yet a systematic characterization of the metabolic pathways active in transformed cells is currently lacking. Using mass spectrometry, we measured the consumption and release (CORE) profiles of 219 metabolites from media across the NCI-60 cancer cell lines, and integrated these data with a preexisting atlas of gene expression. This analysis identified glycine consumption and expression of the mitochondrial glycine biosynthetic pathway as strongly correlated with rates of proliferation across cancer cells. Antagonizing glycine uptake and its mitochondrial biosynthesis preferentially impaired rapidly proliferating cells. Moreover, higher expression of this pathway was associated with greater mortality in breast cancer patients. Increased reliance on glycine may represent a metabolic vulnerability for selectively targeting rapid cancer cell proliferation.  相似文献

Abscisic acid (ABA) is an essential hormone for plants to survive environmental stresses. At the center of the ABA signaling network is a subfamily of type 2C protein phosphatases (PP2Cs), which form exclusive interactions with ABA receptors and subfamily 2 Snfl-related kinase (SnRK2s). Here, we report a SnRK2-PP2C complex structure, which reveals marked similarity in PP2C recognition by SnRK2 and ABA receptors. In the complex, the kinase activation loop docks into the active site of PP2C, while the conserved ABA-sensing tryptophan of PP2C inserts into the kinase catalytic cleft, thus mimicking receptor-PP2C interactions. These structural results provide a simple mechanism that directly couples ABA binding to SnRK2 kinase activation and highlight a new paradigm of kinase-phosphatase regulation through mutual packing of their catalytic sites.  相似文献

All possible tools need to be marshalled for marine fish conservation. Yet controversy has swirled around what role, if any, the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES) should play for marine fishes. This paper analyses the relevance and applicability of CITES as a complementary tool for fisheries management. CITES currently regulates the international trade of very few marine fish species, by listing them in its Appendices. After the first meeting of the Parties (member countries) in 1976, no new marine fish taxa were added to the CITES Appendices until 2002, when Parties agreed to act to ensure sustainable and legal international trade in seahorses (Hippocampus spp.) and two species of sharks. Progress has continued haltingly, adding only one more shark, humphead wrasse (Cheilinus undulatus) and sawfishes by 2012. Parties voice concerns that may include inadequate data, applicability of CITES listing criteria, roles of national fisheries agencies, enforcement challenges, CITES' lack of experience with marine fishes, and/or identification and by‐catch problems. A common query is the relationship between CITES and other international agreements. Yet all these arguments can be countered, revealing CITES to be a relevant and appropriate instrument for promoting sound marine fisheries management. In reality, Parties that cannot implement CITES effectively for marine fishes will also need help to manage their fisheries sustainably. CITES action complements and supports other international fisheries management measures. As CITES engages with more marine fish listings, there will be greater scope to analyse its effectiveness in supporting different taxa in different contexts.  相似文献

Posttranslational modifications of the histone octamer play important roles in regulating responses to DNA damage. Here, we reveal that Saccharomyces cerevisiae Rtt109p promotes genome stability and resistance to DNA-damaging agents, and that it does this by functionally cooperating with the histone chaperone Asf1p to maintain normal chromatin structure. Furthermore, we show that, as for Asf1p, Rtt109p is required for histone H3 acetylation on lysine 56 (K56) in vivo. Moreover, we show that Rtt109p directly catalyzes this modification in vitro in a manner that is stimulated by Asf1p. These data establish Rtt109p as a member of a new class of histone acetyltransferases and show that its actions are critical for cell survival in the presence of DNA damage during S phase.  相似文献

This paper focuses on examining the dynamic nature of community supported agriculture (CSA) and the real-world experiences which mark its contours, often making it distinct from the early idealized CSA “model.” Specifically, our study examines the narratives of the farmers of Devon Acres CSA over its duration, in tandem with a survey of recent shareholders in order to understand and explain its evolution. The framework we develop here shows that this CSA is largely characterized by instrumental and functional beliefs and practices, with some elements in the collaborative mode. A key contribution of this research is the development of a framework which helps to highlight the relative fluidity and patchwork quality of CSA participant positions over time. At Devon Acres, the real-world factors and issues influencing CSA evolution are seen to be products of both the local and larger contexts, evident in such areas as shifts in farmer learning and adaptation, differences between beliefs and practices in member volunteer efforts, and changes in farm and resource conditions. With respect to CSA more broadly, we argue that the reality of dominant food system context and site-specific influences on CSA development compels us to rework our attachment to early idealized “model” traits. Expansion in CSA numbers, evidence of adaptation and situated learning, and retention of the local and organic as core traits, speak to the pragmatic yet transformative potential of CSA contribution to food system change.
