Supercritical fluid extraction (SFE) from aniseed using carbon dioxide was performed at 30 degrees C and pressures of 80-180 bar. The chemical composition of the SFE extract was determined by GC-MS; the quantitative analysis was done by GC-FID and TLC. The total amount of extractable substances or global yield (mass of extract/mass of feed) for the SFE process varied from 3.13 to 10.67% (mass). The solubilities of the anise essential oil in CO(2) were 0.0110, 0.0277, 0.0143, and 0.0182 kg of solute/kg of CO(2) at 80, 100, 140, and 180 bar, respectively. The major compounds identified and quantified in the extracts were anethole ( approximately 90%), gamma-himachalene (2-4%), p-anisaldehyde (<1%), methylchavicol (0.9-1.5%), cis-pseudoisoeugenyl 2-methylbutyrate ( approximately 3%), and trans-pseudoisoeugenyl 2-methylbutyrate ( approximately 1.3%). Four hundred twenty-nine isolates of Escherichia coli from calves were tested for the production of HeLa cell cytotoxin(s). Isolates that produced enough cytotoxin to be detected in culture supernatants of iron-depleted broth were considered to produce increased amounts of cytotoxins. Isolates also were tested for homology with a DNA probe for a gene that encodes localized adherence of human enteropathogenic E coli. Four isolates produced increased amounts of cytotoxin that was neutralized by Shiga antitoxin (toxin designated as Shiga-like toxin-I [SLT-I]). A 5th isolate produced increased amounts of cytotoxin (SLT+) that was not neutralized by the Shiga antitoxin, but was neutralized by antitoxin against a variant of SLT (toxin designated as SLT-II). None of the isolates hybridized with the probe for the localized adherence gene. Three of the SLT+ isolates belonged to human enteropathogenic E coli serogroups O26 and O111. All 5 of the SLT+ isolates were from calves with diarrhea, but none of the 5 SLT+ isolates contained genes for classic heat-labile or heat-stable enterotoxins, for K99 fimbriae, or for invasiveness; neither did any of them adhere to HeLa cells in culture. Three of the 5 SLT+ isolates had attaching and effacing activities when inoculated into ligated intestinal loops of rabbits. One of the isolates with attaching and effacing activity in rabbits was originally isolated from a calf with lesions characteristic of those produced by attaching effacing E coli (AEEC). Toxoplasma gondii, an intracellular protozoan parasite, is one of the major causes of infectious abortion in sheep. To further understand the pathogenesis of toxoplasmosis, serum, amniotic and allantoic fluids and foetal stomach contents were collected from experimentally infected pregnant ewes to determine pathogen numbers and other markers of infection. Fifteen pregnant ewes (90 days of gestation) were each orally inoculated with 3000 sporulated oocysts of T. gondii. Serum samples were collected weekly following challenge. Amniotic and allantoic fluids and foetal stomach contents were collected at 21, 25, 28, 33 and 35 days post-infection. Characteristic placental lesions were detected in 1 of 4 challenged ewes at day 25, 3 of 4 challenged ewes at day 28 and in all challenged ewes at days 33 and 35 post-infection. T. gondii was detected only sporadically in amniotic and allantoic fluids before 35 days of infection, by real-time PCR, and only in ewes with placental lesions. At 35 days post-infection, high numbers of parasite were detected in both amniotic and allantoic fluids. An increase in the number of fluids from challenged animals with IgM and IgG was detected over time, except for IgG in allantoic fluid, which was detected in all samples from day 21 post-infection. IgG in amniotic and allantoic fluids was shown to be specific for T. gondii, and reacted with antigens with an apparent molecular mass of approximately 22 kDa and 30 kDa. Results suggest a maternal source of immunoglobulin in the allantoic fluid and a foetal source of immunoglobulin in the amniotic fluid early in infection but that both sources may contribute immunoglobulin to both fluids at a later stage. 相似文献

We determined the seasonal nutrient and secondary compound intake, in vivo diet digestibility (DD) and diet species composition of goats in different physiological states in a desert rangeland of Argentina. During the wet and the dry seasons, 24 mature female Creole goats (six non‐lactating, non‐pregnant and six lactating in each season) were used in a 2 × 2 factorial design. Diet botanical composition, DD and dry‐matter intake (DMI) were estimated by the combined use of visual observation, microhistological analyses of faecal samples and n‐alkane technique. Browse species dominated the goats’ diets (>80%) in both seasons and physiological states, with a twofold increase in the proportion of forbs during the wet season (p < 0.10). Lactating goats showed greater DMI than non‐lactating animals, and they also displayed greater intakes of some tanniferous (Mimosa ephedroides) and high‐protein (Atriplex lampa) shrubs (p < 0.10). Lactating and non‐lactating goats selected diets with adequate crude protein (CP) and fibre content but insufficient energy, although lactating goats tended to increase energy and tannin content in their diets (p < 0.20). Physiological state did not influence DD, but digestibility was greater (p < 0.10) in the wet season, attributed to the greater protein content and lower tannin content of the diet. These results have important implications for managerial interventions on traditional goat production systems in the study area aiming at enhancing goat nutrition and welfare, as well as the sustainability of rangeland resources in the region. 相似文献

Rice fields represent important production rates in Portugal. However, the intensive soil management and the exploitation of agrochemicals may pose a threat to nontarget organisms. Hence, the present work regards the toxicity screening of surface waters and sediment elutriates collected during the drainage of fields in the vicinity of a rice paddy (Quinta do Seminário, Soure, Portugal): 1. in River Pranto (RP), the river from which the field irrigation water is canalized; 2. inside the rice paddy, from the main drainage channel – Vala de Enxugo (VE). For that purpose, it was used a combination of physico-chemical analyses and bioassays with two green algae species – Pseudokirchneriella subcapitata and Chlorella vulgaris. The chemical screening showed an apparent absence of xenobiotics in sediment samples, while no pesticides were found within the chemical contaminants detected in water samples. The nutrient load reflected low levels of inorganic contamination. Bioassays revealed that P. subcapitata was more sensitive to the overall physico-chemical conditions in natural samples than C. vulgaris, being its growth inhibited under water samples from both sites. On a whole, water samples, mainly those from the main irrigation/drainage channel of the rice fields (VE), were more deleterious to microalgae than those from RP or any of the elutriates. 相似文献

Because of the importance of reproduction in stock breeding systems, it is necessary to find selection criteria that increase reproductive efficiency. The aim of this study was to estimate genetic parameters for the probability of conception on first service (PROB) in Murrah heifers, and its association with other traits of economic interest [age at first calving (AFC), service period, calving interval and milk yield at 270 days], with the purpose of evaluating their use as selection criteria. Reproductive information and first lactation records of 1200 Murrah heifers were used to perform two‐trait analyses between PROB and the other characteristics. Bayesian inference was used to estimate the variance components, considering PROB as threshold and the other as linear factors. The results demonstrate that this trait has heritability of 0.15, indicating the possibility of a genetic gain by using it for selection. With respect to the genetic correlation estimates, the only high‐magnitude association was with AFC (?0.899), which is the current criterion indicating sexual precocity of females. In the light of the parameters estimated, the first‐service pregnancy rate is an alternative for indication of sexual precocity, although presenting a smaller genetic gain than the current standard AFC. Nevertheless, additional research should be conducted regarding this trait to assess the economic importance of its use in dairy buffalo production systems. 相似文献

Effects on erosion of two post-fire management practices: clear-cutting versus non-intervention 总被引:1,自引:0,他引:1
The different erosion responses derived from two contrasting post-fire management approaches were evaluated: the clear-cutting and immediate removal of the burnt trees and; the non-intervention, that is, leaving the vegetation untouched and the burnt trees standing. Erosion rates were obtained from 200 m2 experimental plots, large enough to include the complete processes of rill and inter-rill erosion. Erosion rates of the clear-cut plot were very similar to those of the non-modified plot. Part of the burnt trees was still economically usable. The trees left in situ gradually decayed and were tumbled by the wind; the fire risk incremented. The opening of timber track networks without conservation measures was one of the most important erosion problems associated with clear-cutting. The complementary practice of brushwood dike-construction would need a preliminary study before being applied. 相似文献
