In study I, plasma progesterone concentrations were evaluated in anoestrous mares that received an intravaginal progesterone release device (IPRD) for 10 days. Mares were divided into 3 groups based on the dosage of progesterone (0 g, n=3; 1.38 g, n=5; and 1.9 g, n=5). No statistical differences were found in plasma progesterone concentrations between the two doses tested. In study II, the effects of a protocol based on a short program of artificial light combined with an IPRD containing 1.38 g of progesterone on oestrous behaviour and onset of ovulation were evaluated. IPRDs were inserted into 31 late transitional mares (10 days of treatment). The mares were divided into a control group (n=9, IPRD with 0 g of progesterone) and two treatment groups (T1, n=10, IPRD with 0 g of progesterone and artificial light; T2, n=12, IPRD with 1.38 g of progesterone and artificial light). The percentages of mares in heat within the first 14 days after treatment were 100%, 70%, and 100% in the control, T1, and T2 groups, respectively (P=0.097), and their ovulation rates were 44%, 60%, and 100%, respectively (P≤0.01). In conclusion, a protocol based on artificial light and an IPRD containing 1.38 g of progesterone for 10 days could be considered to advance the first ovulation of the year in late transitional mares, as it ensures a higher rate of ovulation within the first 14 days after treatment.  相似文献

In pepper (Capsicum annuum), the major genes (R‐genes) Me1 and Me3 confer resistance against root‐knot nematodes (Meloidogyne spp.). The combination of R‐genes and quantitative resistance factors in the same genotype is considered a good breeding strategy for increasing the durability of R‐genes. To ascertain this hypothesis, five pepper inbred lines, differing in their quantitative resistance level, were combined with Me1 or Me3 genes in F1 hybrids. The resistance of inbred lines and F1 hybrids was evaluated in a greenhouse with soil naturally infected by M. incognita in two successive growing years. In both years, lines carrying Me3 were less infected by the nematode when combined with quantitative resistance. An increase in nematode infection was observed in the second growing year in lines carrying Me1 or Me3, independently of quantitative resistance. The infection level recorded in inbred lines without R‐genes was similar in both years. The effectiveness of quantitative resistance controlling M. incognita is confirmed in greenhouse conditions, although the durability of Me1 and Me3 when combined with quantitative resistance factors was not seen to increase.  相似文献

• Current intensification trends in the Rio de la Plata need urgent re-direction.•Integrated crop-livestock systems reconcile food production with ecosystem services.•Case studies validate recoupling as a sustainable way to ecological intensification.The Rio de la Plata region comprises central Argentina, Uruguay, and southern Brazil. Modern agriculture developed around 1900 with recent decades being characterized by the advance of cropping areas over native grasslands. Highly specialized agriculture has decoupled crop and livestock production but has succeeded in intensifying yields. However, significant losses of ecosystem services have been reported. Thus, questions have been raised on the sustainability of this pathway. A glance at world regions that have experienced similar trends suggests that an urgent course correction is needed. A major concern has been the lack of diversity in regions with highly specialized agriculture, promoting renewed interest in integrated crop-livestock systems (ICLS), not only because ICLS are more diverse than specialized systems, but also because they are rare examples of reconciliation between agroecosystem intensification and environmental quality. Consequently, this paper discusses alternatives to redesign multifunctional landscapes based on ICLS. Recent data provide evidence that recoupling crop and animal production increases the resilience of nutrient cycling functions and economic indicators to external stressors, enabling these systems to face climate-market uncertainty and reconcile food production with the provision of diverse ecosystem services. Finally, these concepts are exemplified in case studies where this perspective has been successfully applied.  相似文献

Galaxias maculatus is an osmeriform native fish of the Southern Hemisphere, in which the crystalline larvae is considered as a luxury delicacy, for this reason, it has been commercially exploited in Chile, Argentina and New Zealand. However, the fisheries have been rapidly decreasing due to the overexploitation and the predation of introduced species. Because of these events, there is a need to determine a carrying capacity for an intensive fish culture. In order to optimize stocking densities for fish culture, this paper proposes objectives to determine oxygen consumption (OC) rates, dissolved oxygen concentrations that produce signs of hypoxia and the average time elapsed between food intake and peak OC in G. maculatus. In the oxygen experiments under routine metabolism conditions, we found that G. maculatus adults and whitebait showed signs of asphyxia at dissolved oxygen concentrations between 1.3 and 2.2 mg L?1 and that adults tolerated dissolved oxygen levels as low as 1.3 mg L?1. The results showed that G. maculatus individuals with an average weight of 0.04 g consumed 0.048 mg O2 h?1, whereas individuals with an average weight of 1.4 g consumed 0.345 mg O2 h?1. Galaxias maculatus increased the OC rate by 31%, from 0.39 to 0.51 mg O2 h?1 g?1, occurring 14 min after food intake. The carrying capacities for industrial cultures of G. maculatus, were estimated using an allometric equation (OC=0.2363 ×W 0.612 ), a water flow rate of 1 m3 h?1 and an input oxygen concentration of 10 mg L?1 at 12 °C. The density culture of whitebait (4 g) can be allowed to reach 8–11 kg m?3; therefore, these stocking densities reduce the risk of hypoxia and mortality, ensuring the appropriate growth and feed conversion rates.  相似文献

A 28-day indoor trial was conducted to evaluate the water quality, phytoplankton composition and growth of Litopenaeus vannamei in an integrated biofloc system with Gracilaria birdiae and Gracilaria domingensis. The experimental design was completely randomized with three treatments: control (shrimp monoculture); SB (shrimp and G. birdiae) and SD (shrimp and G. domingensis), all with three replicates. Random sampling was done (6 % of total population per experimental unit) to confirm white spot syndrome Virus (WSSV) infection using nested-PCR analysis due to suspicion of presence of the virus in the experiment (treatment and control groups). Shrimp L. vannamei (2.63 ± 0.10 g) were stocked in experimental tanks at a density of 425 shrimp m?3, and the Gracilaria was stocked at a biomass of 2.0 kg m?3. Shrimp mortality began in both the experimental and control groups at 10 days of culture. The integrated biofloc system (shrimp and seaweed) increased settleable solids (by 26–52 %); final weight (by 6–21 %); weekly growth (by 17–43 %); weight gain (by 17–43 %); specific growth rate (by 16–36 %); and yield (by 5–7 %) and decreased feed conversion ratio (by 21–28 %) and Cyanobacteria density about 16 % as compared to the control (shrimp monoculture). The use of red seaweed Gracilaria in an integrated biofloc system can enhance shrimp growth and reduce Cyanobacteria density in the presence of WSSV.  相似文献

The effect of three crude dietary lipid levels (4, 8, and 12%) on egg quality of the female redclaw crayfish, Cherax quadricarinatus, was evaluated. Three replicate groups of 40 crayfish (mean initial weight ± SD, 23.0 ± 3.0 g) per diet treatment were stocked in 15,000 ‐L tanks, at 28 C. After 75 d of culture, there were significant differences (P < 0.001) in egg area (3.52–3.90 mm2), volume (34.0–39.3 mm3), weight (4.81–5.36 mg), and diameter (2.15–2.27 mm) in response to dietary lipids in the diet. Using the quadratic equation, the maximum responses of these parameters corresponded to lipid levels from 8.60–8.84%. There were no significant differences (P > 0.05) in survival (82.1–86.9%), final weight (41.0–43.7 g), number of spawning females (18.12–27.38%), and fecundity (9.07–10.31 eggs/g female). Additionally, there were no significant differences (P > 0.05) in egg mean protein (2306.87 ± 440.08 μg/egg), lipid (441.95 ± 83.23 μg/egg) and carbohydrate (74.81 ± 10.12 μg/egg) contents, and energy (13.49 ± 2.25 kcal/egg). The optimum overall response was obtained for dietary lipid content of 8.7%, which we recommend to optimize egg quality of this freshwater crayfish.  相似文献

Background

The invasive gastropod Pomacea canaliculata has received great attention in the last decades as a result of its negative impact on crops agriculture, yet knowledge of their digestive physiology remains incomplete, particularly the enzymatic breakdown of macromolecules such as proteins and lipids.

Results

Discovery proteomics revealed aspartic peptidases, cysteine peptidases, serine peptidases, metallopeptidases and threonine peptidases, as well as acid and neutral lipases and phospholipases along the digestive tract of P. canaliculata. Peptides specific to peptidases (139) and lipases (14) were quantified by targeted mass spectrometry. Digestion begins in the mouth via diverse salivary peptidases (nine serine peptidases; seven cysteine peptidases, one aspartic peptidase and 22 metallopeptidases) and then continues in the oesophagus (crop) via three luminal metallopeptidases (Family M12) and six serine peptidases (Family S1). Downstream, the digestive gland provides a battery of enzymes composed of aspartic peptidase (one), cysteine peptidases (nine), serine peptidases (12) and metallopeptidases (24), including aminopeptidases, carboxypeptidases and dipeptidases). The coiled gut has M1 metallopeptidases that complete the digestion of small peptides. Lipid extracellular digestion is completed by triglyceride lipases.

Conclusion

From an integrative physiological and anatomical perspective, P. canaliculata shows an unexpected abundance and diversity of peptidases, which participate mainly in extracellular digestion. Moreover, the previously unknown occurrence of luminal lipases from the digestive gland is reported for the first time. Salivary and digestive glands were the main tissues involved in the synthesis and secretion of these enzymes, but plausibly the few luminally exclusive peptidases are secreted by ventrolateral pouches or epithelial unicellular glands. © 2022 The Authors. Brucella melitensis is the Brucella species most frequently associated with brucellosis in humans. It is also the causative agent of the disease in goats and other ruminants. Although significant aspects of the pathogenesis of infection by this intracellular pathogen have been clarified, several events during invasion of host cells remain to be elucidated. In this study, infections of human macrophages from the THP-1 monocyte cell line were conducted with B. melitensis Bm133 wild-type strain and a strain of Salmonella serovar Enteritidis as a control. A multiplicity of infection of 100 was used in trials focused on defining the relative expression of syntaxin 4 (STX4), a soluble N-ethylmaleimide-sensitive factor attachment protein receptor, in the early events of phagocytosis (at 15, 30, 45, and 60 min). Immunoblot assays were also done to visualize expression of the protein in cells infected with either bacterial strain. The expression of STX4 was not significantly different in cells infected with B. melitensis strain Bm133 compared to that observed in cells infected with S. Enteritidis. When the expression of STX4 mRNA was inhibited with short or small interfering, or silencing, RNA in the THP-1 cells, the survival of B. melitensis was significantly reduced at time 0, when gentamicin treatment of cultures was begun (after 1 h of phagocytosis), and also at 2 h and 12 h after infection.  相似文献
