The usefulness of molecular absorption was investigated for the determination of total sulfur (S) in agricultural samples by high-resolution continuum source flame molecular absorption spectrometry. The lines for CS at 257.595, 257.958, and 258.056 nm and for SH at 323.658, 324.064, and 327.990 nm were evaluated. Figures of merit, such as linear dynamic range, sensitivity, linear correlation, characteristic concentration, limit of detection, and precision, were established. For selected CS lines, wavelength-integrated absorbance equivalent to 3 pixels, analytical curves in the 100-2500 mg L(-1) (257.595 nm), 250-2000 mg L(-1) (257.958 nm), and 250-5000 mg L(-1) (258.056 nm) ranges with a linear correlation coefficient better than 0.9980 were obtained. Results were in agreement at a 95% confidence level (paired t test) with those obtained by gravimetry. Recoveries of S in fungicide and fertilizer samples were within the 84-109% range, and the relative standard deviation (n=12) was typically <5%.  相似文献

ABSTRACT: The application of genetic breeding programmes to eradicate transmissible spongiform encephalopathies in goats is an important aim for reasons of animal welfare as well as human food safety and food security. Based on the positive impact of Prnp genetics on sheep scrapie in Europe in the past decade, we have established caprine Prnp gene variation in more than 1100 goats from the United Kingdom and studied the association of Prnp alleles with disease phenotypes in 150 scrapie-positive goats. This investigation confirms the association of the Met142 encoding Prnp allele with increased resistance to preclinical and clinical scrapie. It reveals a novel association of the Ser127 encoding allele with a reduced probability to develop clinical signs of scrapie in goats that are already positive for the accumulation of disease-specific prion protein in brain or periphery. A United Kingdom survey of Prnp genotypes in eight common breeds revealed eleven alleles in over thirty genotypes. The Met142 encoding allele had a high overall mean allele frequency of 22.6%, whereas the Ser127 encoding allele frequency was considerably lower with 6.4%. In contrast, a well known resistance associated allele encoding Lys222 was found to be rare (0.9%) in this survey. The analysis of Prnp genotypes in Mexican Criollas goats revealed nine alleles, including a novel Phe to Leu substitution in codon 201, confirming that high genetic variability of Prnp can be found in scrapie-free populations. Our study implies that it should be feasible to lower scrapie prevalence in goat herds in the United Kingdom by genetic selection.  相似文献

Two strains of the same virus (isolates AR 168 and 7856), were isolated in 1981 from an apparently healthy cow and a sick sheep in TX, U.S.A. These isolates were shown to be members of the Bunyamwera serogroup (family Bunyaviridae, genus Bunyavirus) by complement-fixation tests. Serum dilution-plaque reduction neutralization test results indicated that the isolates are closely related to Cache Valley virus. The virus isolates were characterized by sensitivity to lipid solvent, size (50-100 nm by filtration and 70 nm by electron microscopy), heat (56 degrees C) and pH 3 lability, cytopathic effects or plaques in cultures of Vero, LLC-MK2, embryonic bovine testicle and PS cells, and pathogenicity for suckling and weaned mice by the intracranial but not the intraperitoneal route. Gnotobiotic and conventional sheep and goats were experimentally infected by inoculation with one of the isolates given either intravenously or intraperitoneally. Elevation of body temperature, depression, tremors, muscle spasms, disorientation, feeding anomalies, convulsions, or other signs of central nervous system disturbances were observed.  相似文献

Hong Kong continues to struggle over which environmental governance approach is in its best interest while fulfilling its environmental obligations. With regard to climate change, Hong Kong's approach is characterised by a passive form of governance that is highly dependent on China's national policy directions. This is reflected, for example, in Hong Kong having not set its own mitigation targets. Market mechanisms have received little attention in developing a city‐wide climate change strategy. A transformative impulse, China's national emissions trading scheme, may provide momentum to a market‐based approach. However, the necessary conditions for such a market mechanism to be successfully implemented in China remain relatively undeveloped. This raises question about early participation by Hong Kong. Direct benefits are likely to be limited, due to Hong Kong's economic structure and weak demand for emissions reduction. Besides, there are regulatory barriers to enforcing emissions targets and/or recognising emissions allowances and credits from China. We therefore argue that nationwide emissions trading may, at this time, present more challenges than opportunities for Hong Kong to leverage its efforts on climate change mitigation. An alternative is to promote voluntary emissions trading that will require active involvement and leadership by businesses.  相似文献

Context

Increased edge density is among the main negative effects of habitat loss and fragmentation. Roads are linear infrastructures that may promote barrier effects due to disturbance and mortality effects. We hypothesized that edges of habitat patches bordered by roads are less permeable than roadless edges.

Objectives

We tested whether edge permeability and avoidance are influenced by the presence of paved and dirt roads bordering habitat patches, relatively to roadless edges.

Methods

We translocated 55 montane akodonts (Akodon montensis) from the interior of vegetation remnants to their edges, and tracked fine-scale movements using spool-and-line devices. Edges were bordered by dirt roads (n = 12 mice), paved roads (n = 21) or were not bordered by roads (n = 22). We assessed edge permeability by comparing the number of tracks with crossings, and by comparing the empirical data to simulated correlated random walks. We also assessed edge avoidance by comparing the net direction travelled and net displacement from edge.

Results

No edge crossings were recorded in roaded edges, whereas 36% of tracks in roadless edges crossed the edge at least once. Simulations indicated a significantly lower permeability of roaded edges, while the observed number of crossings in roadless edges was within the expected range. We found no evidence of higher avoidance of roaded edges, as both net direction travelled and displacement were similar across edge types.

Conclusions

Roads decreased edge permeability for the montane akodont. This is likely to increase population isolation among vegetation remnants by reducing the structural connectivity in the already fragmented landscape.

Context

Land use changes have modified the extent and structure of native vegetation, resulting in fragmentation of native species habitat. Connectivity is increasingly seen as a requirement for effective conservation in these landscapes, but the question remains: ‘connectivity for which species?’.

Objective

The aim of this study was to develop and then apply a rapid, expert-based, dispersal guild approach where species are grouped on similar fine-scale dispersal behaviour (such as between scattered trees) and habitat characteristics.

Methods

Dispersal guilds were identified using clustering techniques to compare dispersal and habitat parameters elicited from experts. We modelled least-cost paths and corridors between patches and individual movement probabilities within these corridors for each of the dispersal guilds using Circuitscape. We demonstrate our approach with a case study in the Tasmanian Northern Midlands, Australia.

Results

The dispersal guild approach grouped the 12 species into five dispersal guilds. The connectivity modelling of those five guilds found that broadly dispersing species in this landscape, such as medium-sized carnivorous mammals, were unaffected by fragmentation while from the perspective of the three dispersal guilds made up of smaller mammals, the landscape appeared highly fragmented.

Conclusions

Our approach yields biologically defensible outputs that are broadly applicable, particularly for conservation planning where data and resources are limited. It is a useful first step in multi-species conservation planning which aims to identify those species most in need of conservation efforts.

A device capable of testing the nociceptive threshold to mechanical stimuli has been developed for use in horses. It was operated by compressed gas and was designed so that the part attached to the horse was semi disposable. The stimulus was applied via a 2mm diameter blunt ended pin and had a number of safety features incorporated in the design to ensure that the maximum force applied was 27 newtons. The device has been used to assess the response to detomidine in a group of thoroughbreds and has proved reliable and robust in use.  相似文献

The analgesic activity of fentanyl was measured in sheep using both thermal and mechanical test systems. Fentanyl at a dose rate of 5 μg/kg given intravenously (iv) produced significant analgesia to thermal pain for some 30 mins but no detectable mechanical antinociceptive activity. However, at a dose rate of 10 μg/kg the drug produced both thermal (for 60 mins) and mechanical (40 mins) antinociceptive effects. In one sheep a dose of 20 μg/kg produced thermal analgesia for 110 mins and mechanical antinociception for 60 mins. Following iv injection at 10 μg/kg in five of the sheep, there was a brief period of respiratory depression evidenced by a significant fall in PaCO2 of the order of 25 per cent and an increase in PCO2 levels, but these changes were short lived and levels were back to normal at 15 mins after injection.  相似文献
