Mycobacterium avium subspecies paratuberculosis (M. a. paratuberculosis) is a pathogen of ruminants, causing paratuberculosis (characterized by severe emaciation). The disease is endemic in many countries including the UK and places a severe economic burden on the global livestock industry. Two types of M. a. paratuberculosis can be classified by pulsed-field electrophoresis (I/III and II), which are phenotypically distinct and appear to have different host preferences. Proteomes of Type I and Type II M. a. paratuberculosis were analyzed by 2-D gel electrophoresis to determine if any significant differences existed between the subtypes. Seven different strains of Type I and 18 strains of Type II were analyzed and compared to detect type-specific differences. These 'type-specific' differences existed regardless of growth phase and were also exhibited in cells isolated directly from pathogenic lesions. Twenty-three spots predominated on the Type I profile, from which 17 proteins were identified. Twenty-one spots predominated on the Type II profile, from which 16 proteins were identified. None of the proteins identified as differentially represented on the profiles of Type I or Type II corresponded to open reading frames of the defining genomic regions as previously described for the Type I (sheep) and Type II (cattle). Sequence polymorphisms existing in Type I and II strains were identified in some open reading frames or regulatory regions of genes that correspond to proteins expressed in a type-specific fashion. The consequence of these is discussed in relation to protein expression and their impact on the type phenotype is discussed.  相似文献

Context

Modulus of elasticity (MOE) is an important mechanical property determining the end-use and value of loblolly pine (Pinus taeda L.) lumber.

Aim

In this study, a model was developed to predict the within tree variation of MOE, from pith-to-bark and stump-to-tip, using data collected from a 21-year-old unthinned stand where trees were planted under seven initial stand density levels (746?C2,243 trees/hectare).

Methods

The study was laid out in a randomized complete block design, with seven levels of initial planting density, replicated three times. Seven trees were destructively sampled from each plot, and bolts were cut from each tree at heights of 2.4, 7.3, and 12.2?m. Static bending samples (of dimension 2.5?×?2.5?×?40.6?cm) representing pith-to-bark variation were cut from each bolt and MOE measured. A three-parameter logistic function was used to model the pith-to-bark variation in stiffness with distance from pith as an explanatory variable.

Results

Based on the final fitted model, it was found that the asymptotic parameter (maximum outerwood MOE?=?13.48?GPa) was not influenced by sampling height, initial planting density, and stem slenderness. However, the inflection and scale parameters were significantly influenced by these variables.

Conclusions

In summary, we found that initial planting density had a significant influence on the amount of corewood produced with higher initial planting densities producing a lower proportion of corewood as indicated by a linear decrease in inflection point with an increase in planting density.  相似文献

The wild lettuce, Lactuca aculeata Boiss. et Ky., is closely related and fully interfertile with cultivated lettuce, Lactuca sativa L. Altogether, 155 accessions of this wild species, which is rare conserved within the world’s germplasm collections, were collected from 12 localities throughout northeastern Israel. Ten of these localities represent different sites across the Golan Heights, while two, high-density populations were suprisingly found at the Hula Plain (first report of this species in this region). Lactuca aculeata was recorded at various elevations (222–968 m a.s.l.) and habitats. The taxonomic status of 30 L. aculeata accessions was morphologically validated during ex situ seed regeneration of 31 random accessions representing all 12 localities. Characterization of 12 traits showed that the Israeli populations of L. aculeata do not exhibit broad morphological variability, but more great levels of variation were obtained for developmental traits. Relatively little variability was observed in the morphology of cauline leaves or in the distribution of anthocyanin pigmentation in bracteae. In two cases, L. aculeata plants lacked trichomes in the inflorescence, a typical feature in the majority of plants. These unique collections of wild lettuce may carry novel sources of genetic variation for a wide range of traits and, thus, should be of interest for careful evaluation and exploitation in lettuce breeding.  相似文献

Variations in intracranial dural venous sinus anatomy have been widely reported in humans, but there have been no studies reporting this in dogs. The purpose of this retrospective study was to describe variations in magnetic resonance (MR) venographic anatomy of the dorsal dural venous sinus system in a sample population of dogs with structurally normal brains. Medical records were searched for dogs with complete phase contrast, intracranial MR venograms and a diagnosis of idiopathic epilepsy. Magnetic resonance venograms were retrieved for each dog and characteristics of the dorsal dural sinuses, symmetry of the transverse sinuses and other anatomic variations were recorded. A total of 51 dogs were included. Transverse sinus asymmetry was present in 58.8% of the dogs, with transverse sinus hypoplasia seen in 39.2%, and aplasia in 23.5% of dogs. For 70.6% of dogs, at least one anatomic variation in the dorsal sagittal sinus was observed, including deviation from the midline (33.3%) and collateral branches from either the dorsal sagittal sinus or dorsal cerebral veins (54.9%). In 5 dogs (9.8%) a vessel was also identified running from the proximal transverse sinus to the distal sigmoid sinus, in a similar location to the occipital sinus previously reported in children. Findings from this study indicated that, as in humans, anatomic variations are common in the intracranial dural venous sinus system of dogs. These anatomic variations should be taken into consideration for surgical planning or diagnosis of cerebrovascular disease.  相似文献

The art and science of developing effective policies and practices to enhance sustainability and adapt to new climate conditions on rangelands and savannas are typically founded on addressing the “average” or “typical” resource user. However, this assumption is flawed since it does not appreciate the extent of diversity among resource users; it risks that strategies will be irrelevant for many people and ignored, and that the grazing resource itself will remain unprotected. Understanding social heterogeneity is vital for effective natural resource management. Our aim was to understand the extent to which graziers in the northern Australian rangelands varied in their capacity to adapt to climate variability and recommended practices. Adaptive capacity was assessed according to four dimensions: 1) the perception of risk, 2) skills in planning, learning and reorganising, 3) financial and emotional flexibility, and 4) interest in adapting. We conducted 100 face-to-face interviews with graziers in their homes obtaining a 97% response rate. Of the 16 possible combinations that the four dimensions represent, we observed that all combinations were present in the Burdekin. Any single initiative to address grazing land management practices in the region is unlikely to address the needs of all graziers. Rather, policies could be tailored to type-specific needs based on adaptive capacity. Efforts to shift graziers from very low, low, or moderate levels of adaptive capacity are urgently needed. Transmissible spongiform encephalopathies (TSEs) or prion diseases are unique disorders that are not caused by infectious micro-organisms (bacteria or fungi), viruses or parasites, but rather seem to be the result of an infectious protein. TSEs are comprised of fatal neurodegenerative disorders affecting both human and animals. Prion diseases cause sponge-like degeneration of neuronal tissue and include (among others) Creutzfeldt–Jacob disease in humans, bovine spongiform encephalopathy (BSE) in cattle and scrapie in sheep. TSEs are characterized by the formation and accumulation of transmissible (infectious) disease-associated protease-resistant prion protein (PrPSc), mainly in tissues of the central nervous system. The exact molecular processes behind the conversion of PrPC into PrPSc are not clearly understood. Correlations between prion protein polymorphisms and disease have been found, however in what way these polymorphisms influence the conversion processes remains an enigma; is stabilization or destabilization of the prion protein the basis for a higher conversion propensity? Apart from the disease-associated polymorphisms of the prion protein, the molecular processes underlying conversion are not understood. There are some notions as to which regions of the prion protein are involved in refolding of PrPC into PrPSc and where the most drastic structural changes take place. Direct interactions between PrPC molecules and/or PrPSc are likely at the basis of conversion, however which specific amino acid domains are involved and to what extent these domains contribute to conversion resistance/sensitivity of the prion protein or the species barrier is still unknown.  相似文献

Abrupt dietary transitions and feeding of rapidly fermentable diets are common practices in the horse industry and have been associated with digestive and metabolic disorders that can impair the performance of horses. The present study investigated the effect of dietary transition from pasture grazing to confinement with concentrate feeding, and back, on fecal pH and bacterial populations of Streptococcus spp and Lactobacillus spp. Six Thoroughbred fillies, previously grazing perennial ryegrass and white clover-based pasture, were housed in individual stalls and fed an increasing ratio of concentrate to conserved forages for 13 days (days 1-13), followed by an abrupt transition back to only pasture-grazing for 3 days (days 14-16). The concentrate was initially offered at 0.83 kg dry matter (DM)/d and increased to 5 kg DM/d, whereas ensiled alfalfa was initially offered at 0.61 kg DM/d, increasing to 1.22 kg DM/d. Meadow hay was initially offered at 6.73 kg DM/d, decreasing to 1.6 kg DM/d. Fecal specimens were collected daily for determination of pH, and every 2 days for quantitative analysis of Streptococcus spp and Lactobacillus spp. Mean fecal pH increased significantly from pasture baseline values (pH 6.18) during the initial confinement and supplementation on day 1 (6.37), day 2 (6.52), day 3 (6.58), and day 4 (6.43) (standard error of mean [SEM]: 0.056; P < .001). By day 5, mean fecal pH values had decreased to, and remained at, baseline values until the horses returned to pasture, when another increase occurred at day 15 (6.45). Fecal colony forming units (cfu) of Streptococcus spp and Lactobacillus spp increased linearly (r = 0.94; P < .001) from 6.0 and 6.1 log10 cfu/g on day -4, to 7.8 log10 cfu/g on day 14 (SEM: 0.2 P < .001), respectively. Fecal cfu decreased on return to a pasture-only diet (P < .001). In this study, the increment of bacterial populations was associated with a relatively stable fecal pH and highlights the difficulty in identifying the effects of dietary transition on the equine hindgut health, without microbial culture.  相似文献

There are few reliable data sets to inspire confidence in policymakers that soil organic carbon (SOC) can be measured on farms. We worked with farmers in the Tamar Valley region of southwest England to select sampling sites under similar conditions (soil type, aspect and slope) and management types. Topsoils (2–15 cm) were sampled in autumn 2015, and percentage soil organic matter (%SOM) was determined by loss on ignition and used to calculate %SOC. We also used the stability of macroaggregates in cold water (WSA) (‘soil slaking’) as a measure of ‘soil health’ and investigated its relationship with SOC in the clay‐rich soils. %SOM was significantly different between management types in the order woodland (11.1%) = permanent pasture (9.5%) > ley‐arable rotation (7.7%) = arable (7.3%). This related directly to SOC stocks that were larger in fields under permanent pasture and woodland compared with those under arable or ley‐arable rotation whether corrected for clay content (F = 8.500, p < .0001) or not (F = 8.516, p < .0001). WSA scores were strongly correlated with SOC content whether corrected for clay content (SOCadj R2 = .571, p < .0001) or not (SOCunadj R2 = 0.490, p = .002). Time since tillage controlled SOC stocks and WSA scores, accounting for 75.5% and 51.3% of the total variation, respectively. We conclude that (1) SOC can be reliably measured in farmed soils using accepted protocols and related to land management and (2) WSA scores can be rapidly measured in clay soils and related to SOC stocks and soil management.  相似文献

Many studies have been undertaken in rodents to study the pathogenesis of transmissible spongiform encephalopathies (TSE). Only a few studies have focused on the pathogenesis of bovine spongiform encephalopathy (BSE) and scrapie in their natural hosts. In this review, we summarize the most recent insights into the pathogenesis of BSE and scrapie starting from the initial uptake of TSE agents and crossing of the gut epithelium. Following replication in the gut-associated lymphoid tissues (GALT), TSE agents spread to the enteric nervous system (ENS) of the gut. Infection is then carried through the efferent fibers of the post-ganglionic neurons of the parasympathetic and sympathetic nervous system to the pre-ganglionic neurons in the medulla oblongata of the brain and the thoracic segments of the spinal cord. The differences between the pathogenesis of BSE in cattle and scrapie in sheep are discussed as well as the possible existence of additional pathogenetic routes.  相似文献
