With this study, we aimed to assess the safety of using Curcuma longa in fish feed. Juvenile Astyanax aff. bimaculatus (0.83 ± 0.04 g) were kept in 24 80‐L aquaria, at a density of 0.5 fish L?1 for 60 days. Six diets supplemented with 0.0, 20.0, 40.0, 60.0, 80.0 and 100.0 g kg?1 were evaluated. Histopathological analysis of the gills showed that there was an increasing linear effect of turmeric on the frequency of lamellar aneurysm, hypertrophy of mucous cells, oedema and necrosis. There was a quadratic effect of turmeric supplementation on the frequency of fusion and disruption of secondary lamellae, vacuolation and hypertrophy of the epithelial cells, and the estimated values to minimize these variables were 4.63, 25.93 and 24.46 g kg?1 respectively. There was a quadratic effect of turmeric on the gills index (IBRA), and the estimated value to minimize this index was 30.23 g kg?1. No effect of turmeric on the frequency of histopathological changes and organ index in fish liver was observed. Thus, we conclude that turmeric is not hepatotoxic for A. aff. bimaculatus; however, its use is safe at concentrations up to 30.23 g kg?1 diet as a function of its gill toxic effect at higher doses. 相似文献

A feeding trial was conducted to evaluate the potential of dietary supplementation of autolysed brewer's yeast (AY) on African catfish. The catfish (22.5 ± 1.15 g/fish, 20 fish 33 L/tank) were fed with either of diets (390 g/kg crude protein, 140 g/kg lipid) supplemented with 0, 3, 6 or 10 g/kg AY (n = 3). After 49 days of feeding, the final body weight and metabolic growth rate of the catfish fed 3 g/kg AY (3‐AY) diet were higher than those fed the control diet (p < .05). The lowest level (p < .05) of alanine transaminase was detected in the blood of the catfish fed 3‐AY diet. The mid‐intestinal histology of the catfish revealed no significant difference (p > .05) in intestinal perimeter ratio. However, an elevated (p < .05) abundance of goblet cells and intraepithelial leucocytes were found in the intestine of catfish fed 3, 6 and 10 g/kg AY diets, with the highest level of abundance recorded in the mid‐intestine of the catfish fed 3‐AY diet. The results suggest that dietary 3 g/kg autolysed brewer's yeast supplementation improves growth performance of African catfish without deleterious effect on liver functionality and gut morphology. 相似文献

Underlying many payments for watershed services (PWS) schemes in tropical montane forest contexts is the assumption of a direct positive relationship between forest cover and dry season stream flow. We developed a low cost research program to assess the forest cover-stream flow relationship in the Los Negros watershed in eastern Bolivia. We asked three questions: (1) can watersheds that are similar enough to undertake paired catchment studies be identified using only simple parameters such as size, aspect and geographic proximity; (2) can a functioning locally based hydrological monitoring system be set up for less than $10 000 by training local farmers to collect hydrological data, and (3) can such data be used to improve the functioning of a PWS initiative? A land use map of the upper Los Negros valley was created from a 2005 Landsat image and a digital elevation model used to calculate physical and hydrological properties of 10 sub-watersheds. Farmers measured stream flow rates in these sub-watersheds from 2005 to 2008 and maintained 10 automatic rain gauges. We found no relationship between forest cover and stream flow. This may indicate that no such relationship exists, but could also reflect the short period of the study, the low quality of the data, and the fact that the sub-watersheds had relatively similar forest coverage (54–76%). We conclude that (1) watersheds can be identified as “similar-enough-for-analysis” using the criteria of size, aspect and proximity without undertaking further research, (2) a useful hydrological monitoring system can be developed for <$10,000 and (3) although our local farmers did not collect sufficiently high quality data to fully explore the forest/water relationship in Los Negros, with improvements in methodologies, low cost, locally based monitoring has the potential to be an important component of future PWS initiatives. We recommend that stream discharge should be calculated directly; only the most locally relevant hydrological criteria, rather than scientifically complete criteria, should be monitored; locally based monitoring must be institutionalized to reduce staff turnover and hydrological monitoring must be embedded within a context that makes it socially acceptable. 相似文献

Agricultural peat soils in the Sacramento-San Joaquin Delta, California have been identified as an important source of dissolved organic carbon (DOC) and trihalomethane precursors in waters exported for drinking. The objectives of this study were to examine the primary sources of DOC from soil profiles (surface vs. subsurface), factors (temperature, soil water content and wet-dry cycles) controlling DOC production, and the relationship between C mineralization and DOC concentration in cultivated peat soils. Surface and subsurface peat soils were incubated for 60 d under a range of temperature (10, 20, and 30 °C) and soil water contents (0.3-10.0 g-water g-soil−1). Both CO2-C and DOC were monitored during the incubation period. Results showed that significant amount of DOC was produced only in the surface soil under constantly flooded conditions or flooding/non-flooding cycles. The DOC production was independent of temperature and soil water content under non-flooded condition, although CO2 evolution was highly correlated with these parameters. Aromatic carbon and hydrophobic acid contents in surface DOC were increased with wetter incubation treatments. In addition, positive linear correlations (r2=0.87) between CO2-C mineralization rate and DOC concentration were observed in the surface soil, but negative linear correlations (r2=0.70) were observed in the subsurface soil. Results imply that mineralization of soil organic carbon by microbes prevailed in the subsurface soil. A conceptual model using a kinetic approach is proposed to describe the relationships between CO2-C mineralization rate and DOC concentration in these soils. 相似文献

OBJECTIVE: To describe shiga-toxigenic Escherichia coil O157:H7 (STEC O157:H7) fecal shedding prevalence, seasonal fecal shedding patterns, and site-specific prevalence from the oral cavity, skin, and feces of dairy cattle. DESIGN: Cross-sectional study. ANIMALS: Adult dairy cattle from 13 herds in Louisiana. PROCEDURE: Samples were cultured for STEC O157 by use of sensitive and specific techniques, including selective broth enrichment, immunomagnetic separation, monoclonal antibody-based O:H enzyme immunoassay serotyping, and polymerase chain reaction virulence gene characterization. Point estimates and 95% confidence intervals were calculated for fecal shedding prevalence as well as site-specific prevalence from the oral cavity, skin, and feces. Logistic regression was used to assess seasonal variation and differences at various stages of lactation with respect to fecal shedding of STEC O157 in cattle sampled longitudinally. RESULTS: Summer prevalence in herds in = 13) was 38.5%, with a cow-level prevalence of 6.5%. Among positive herds, prevalence ranged from 3% to 34.6%. Samples from 3 of 5 herds sampled quarterly over 1 year yielded positive results for STEC O157. In herds with STEC O157, an increase in cow-level prevalence was detected during spring (13.3%) and summer (10.5%), compared with values for fall and winter. Site-specific prevalences of STEC O157:H7 from oral cavity, skin, and fecal samples were 0%, 0.7%, and 25.2%, respectively. CONCLUSIONS AND CLINICAL RELEVANCE: Our data indicated that STEC O157:H7 was commonly isolated from dairy cows in Louisiana, seasonally shed, and isolated from the skin surface but not the oral cavity of cows. 相似文献

BACKGROUND: The expected number of thrombopoietic cells in normal canine bone marrow is poorly defined and there is no consensus on the most appropriate way to prepare cytologic smears to evaluate these cells nor on the optimum method for their quantification. OBJECTIVE: The purpose of this study was to determine total and differential counts of thrombopoietic cells in the bone marrow of clinically healthy Beagle dogs by comparing 4 different smear types and bone marrow core biopsies. METHODS: Twenty-two clinically healthy, male Beagle dogs, 10 to 12 months old, were used in the study. Following bone marrow aspiration and core biopsy from the iliac crest, Giemsa-stained smears were prepared by 4 techniques: drop-squash, particle-squash, buffy coat, and fat-layer smears. Thrombopoietic cells were counted in up to 100 low-power fields (LPF, X10 objective) in the aspiration smears and in all possible high-power fields (HPF, X40 objective) in H&E-stained biopsy sections. RESULTS: Mean total thrombopoietic cell counts were 2.76 cells/LPF (drop-squash), 1.55 cells/LPF (particle-squash), 8.05 cells/LPF (buffy coat), and 3.08 cells/LPF (fat-layer). Core biopsies yielded 5.31 cells/HPF but frequently failed to provide interpretable specimens. There was a significant difference in cell counts among the 4 smear types (P <.001). Based on evaluation of buffy coat smears, thrombopoietic cells included 1.23% megakaryoblasts, 8.77% promegakaryocytes, and 90% megakaryocytes, with a mean maturation index of 0.11. CONCLUSION: Thrombopoietic cell counts in canine bone marrow are influenced by the smear technique. Buffy coat and fat-layer smears may be useful to obtain cellular smears in hemodiluted or small aspirate samples. 相似文献

The efficient storage and germination of seeds underpin the effective use of plants for livelihoods and sustainable development. A total of 204 wild species useful for local communities of the Tehuacán–Cuicatlán Valley were collected and stored in seed banks in country for long term conservation, and 66 % (i.e., 134) duplicated in the U.K., as an effective means of ex situ conservation. Of the 204 species, 147 (122 of which also duplicated in the U.K.) were previously listed as useful plants in the ethnofloristic inventory of the Valley. Based on literature surveys, we found that one of the major impediments to the use of stored seeds of wild species is the lack of knowledge of how to germinate the seed. In detailed studies, we found that seeds of 18 useful plant species from 10 different families germinated readily and could be propagated. In contrast, four species (Actinocheita filicina, Bursera submoniliformis, Karwinskia mollis and Lippia graveolens) produced dormant seeds and therefore further studies are needed before their use can be maximised in large scale propagation programmes in support of conservation and livelihoods. Overall, this large-scale study on useful wild plant species in Mexico confirms that conventional seed banking can effectively support sustainable development and livelihood programmes. 相似文献
