ABSTRACT High infection rates of wild olive (Olea europaea sp. sylvestris) feeder roots and soil infestation by a new root-knot nematode were found in sandy soil at Vejer de la Frontera (Cádiz), southern Spain. Morphometric traits and analyses of the nematode esterase electrophoretic pattern as well as of the internal transcribed spacer 1 (ITS1)-5.8S gene and D2-D3 fragment of the 28S gene of rDNA showed that specimens differed clearly from known root-knot nematodes. Studies of host-parasite relationships showed a typical susceptible reaction in naturally infected wild olive plants and in olive planting stocks (cvs. Arbequina and Picual) artificially inoculated with the nematode. However, the nematode did not reproduce in artificially inoculated chickpea, pea, and tomato. Because of the ability of this new nematode to infect wild and cultivated olives only, we suggest the common name, "Mediterranean olive root-knot nematode." The species is herein described and illustrated, and named as Meloidogyne baetica n. sp. The new root-knot nematode can be distinguished from other Meloidogyne spp. by (i) the perineal pattern, which is almost similar to that of M. artiellia, characterized by distinct inner striae forming two distinct longitudinal bands, extending throughout the perineum to just below the vulva; (ii) female excretory pore anterior to the level of stylet knobs, excretory pore distance from anterior end/length of stylet ratio extremely small (0.5 to 0.8); and (iii) second-stage juveniles with elongate-conoid tail. Phylogenetic trees derived from maximum parsimony analyses showed that M. baetica is closely related to M. artiellia, the cereal and legume root-knot nematode.  相似文献

Breeding for resistance was considered one of the possible ways to limit the damages caused by the epidemics of Dutch elm disease (DED) during the last century. The elm breeding program developed by CNR in Italy was based on the idea that the Mediterranean environment would need its own specific selections. A base broadening of the genetic resources was operated. A base of native elms with a set of good characters to act as parents was bred with different Asian elm species that showed the ability of acclimatization to the different climates in which elms have to be planted. For this aim a large collection of elm species was constituted, followed by hybridisation studies. Progenies were tested for DED resistance. The more resistant clones were planted in trial fields characterised by contrasting Mediterranean climates in order to select the best potential genotypes adapted either to coastal or to mountain environmental conditions. More than 60 clones resistant, fast-growing and showing remarkable aesthetic ornamental characters were obtained.  相似文献

An EST-based PCR-RFLP method was employed to gain insight into genome organization in eight allopolyploid Coffea arabica cultivars and seven diploid coffee species. The PCR-amplified products at 19 EST loci were digested with 46 different restriction enzymes and size fractioned in agarose gels. Most often, the sum of the fragments length was double or more than the PCR product. In arabica, this condition could be explained by assuming the presence of duplicated loci in paralogous chromosomes and this was supported by considerable evidence of multiple loci SSR amplification. Based on the RFLP analysis, 12 EST loci were polymorphic. The level of polymorphism was higher in different species compared to the arabica varieties. Sequencing of the amplified products revealed a SNP frequency of 0.021 among diploid species and of 0.007 among arabica varieties. We propose that the involvement of two genomes in C. arabica maintains a residual level of heterozygosity in the form of paralogous chromosomes, while the self-fertilization in this species tends to drive of homozygosity. The heterozygosity of paralogous chromosomes in arabica creates valuable polymorphism essential for species diversity and survival in various ecological niches, while self-fertility tends to preserve in homozygosity many genes of functional significance.  相似文献

The objective of this research was to evaluate the nitrogen, phosphorus and potassium (NPK) fertilizer effect on the development of ‘Valencia’ sweet orange nursery trees budded on Rangpur lime rootstocks in a greenhouse using containers. The experiment was a complete (1/5) 53 factorial randomized blocks design. Treatments comprised five concentrations (g per plant) of N (1.25; 6.25; 11.25; 16.25; 21.25); K (0.42; 3.75; 6.22; 9.34; 12.45); and P (0.19; 0.89; 1.59; 2.29; 2.99). Carbon dioxide assimilation rate (A), transpiration rate (E), stomata conductance (gs), and internal carbon dioxide (CO2) concentration (Ci) and water use efficiency (WUE). Reducing sugar, sucrose and starch were evaluated. Response functions were adjusted and nutrient rates for maximum yield were presented. The results indicated that the high levels of N (>16.25 g per plant) interferes negatively on photosynthesis. Potassium at intermediate fertilization levels (from 3.75 to 6.22 g per plant) had positive effects on photosynthesis. But P had little interference on photosynthesis. Carbohydrates levels were not related to nutrients fertilized.  相似文献

Urea is the most common nitrogen(N)fertilizer used in the tropics but it has the risk of high gaseous nitrogen(N)losses.Use of nitrification inhibitor has been suggested as a potential mitigation measure for gaseous N losses in N fertilizer-applied fields.In a field trial on a tropical Andosol pastureland in Costa Rica,gaseous emissions of ammonia(NH3)and nitrous oxide(N2O)and grass yield were quantified from plots treated with urea(U;41.7 kg N ha-1application-1)and urea plus the nitrification inhibitor nitrapyrin(U+NI;41.7 kg N ha-1application-1and 350 g of nitrapyrin for each 100 kg of N applied)and control plots(without U and NI)over a six-month period(rainy season).Volatilization of NH3(August to November)in U(7.4%±1.3%of N applied)and U+NI(8.1%±0.9%of N applied)were not significantly different(P>0.05).Emissions of N2O in U and U+NI from June to November were significantly different(P<0.05)only in October,when N2O emission in U+NI was higher than that in U.Yield and crude protein production of grass were significantly higher(P<0.05)in U and U+NI than in the control plots,but they were not significantly different between U and U+NI.There was no significant difference in yield-scaled N2O emission between U(0.31±0.10 g N kg-1dry matter)and U+NI(0.47±0.10 g N kg-1dry matter).The results suggest that nitrapyrin is not a viable mitigation option for gaseous N losses under typical N fertilizer application practices of pasturelands at the study site.  相似文献

Nitrification inhibitors are widely used in agriculture to mitigate nitrous oxide(N2O)emission and increase crop yield.However,no concrete information on their mitigation of N2O emission is available under soil and environmental conditions as in Pakistan.A field experiment was established using a silt clay loam soil from Peshawar,Pakistan,to study the effect of urea applied in combination with a nitrification inhibitor,nitrapyrin(2-chloro-6-tri-chloromethyl pyridine),and/or a plant growth regulator,gibberellic acid(GA_3),on N2O emission and the nitrogen(N)uptake efficiency of maize.The experimental design was a randomized complete block with five treatments in four replicates:control with no N(CK),urea(200 kg N ha-1)alone,urea in combination with nitrapyrin(700 g ha-1),urea in combination with GA_3(60 g ha-1),and urea in combination with nitrapyrin and GA_3.The N2O emission,yield,N response efficiency,and total N uptake were measured during the experimental period.The treatment with urea and nitrapyrin reduced total N2O emission by 39%–43%and decreased yield-scaled N2O emission by 47%–52%,relative to the treatment with urea alone.The maize plant biomass,grain yield,and total N uptake increased significantly by 23%,17%,and 15%,respectively,in the treatment with urea and nitrapyrin,relative to the treatment with urea alone,which was possibly due to N saving,lower N loss,and increased N uptake in the form of ammonium;they were further enhanced in the treatment with urea,nitrapyrin,and GA_3 by 27%,36%,and 25%,respectively,probably because of the stimulating effect of GA_3 on plant growth and development and the reduction in biotic and abiotic stresses.These results suggest that applying urea in combination with nitrapyrin and GA_3 has the potential to mitigate N2O emission,improve N response efficiency,and increase maize yield.  相似文献

In tropical soils, the high turnover rate and mineralization of organic matter (OM) associated with intensive agricultural use, generally leads to faster soil degradation than that observed in temperate climatic zones. The application of sewage sludge to the soils is one proposed method of maintaining soil organic matter, and is also an alternative method of disposing of this waste product. As well as containing large quantities of OM, sludge is also a significant source of supplementary nitrogen, phosphorus and other essential nutrients for plant growth. However, it is necessary to understand the qualitative and quantitative changes that take place in the OM in soil treated with sewage sludge. The approach of the present study was intended to identify possible structural changes caused by sewage sludge applications on soil humic acids (HAs). The HAs extracted from a Typic Achrortox under sewage sludge applications were characterized by electron paramagnetic resonance (EPR) spectroscopy. The soil samples were collected from a field experiment designed to evaluate the effects of different doses of sewage sludge on corn growth and development in Brazil. The sewage sludge originated from urban waste treated at the sewage sludge treatment station in the city of Franca, state of Sao Paulo, Brazil. The following soil treatments were studied: control (non-cultivated soil under natural vegetation (NC)), control soil amended with NPK (conventional corn fertilization) and four treatments N1, N2, N4 and N8 with applications of 3.5, 7, 14 and 28 Mg ha−1 of sewage sludge (dry matter), respectively. HAs were extracted from the surface layer using the methodology of the International Humic Substance Society (IHSS). Fe3+ and VO2+ ions complexed with HAs, and also semiquinone-type free radical (SFR) at concentrations of approximately 2.0 × 1018 spins g−1 HA were identified in EPR spectra. The levels of SFR were lower for treatments where the applied sewage sludge doses were equivalent to four and eight times the normal doses of N mineral fertilization, reaching values of 1.7 × 1018 and 1.24 × 1018 spins g−1 HA, respectively. The observed decrease in SFR content as sewage sludge dose was increased, was probably associated with the incorporation of less aromatic components into HAs originating from the sewage sludge.  相似文献

Compounds with a (2H)-1,4-benzoxazin-3(4H)-one skeleton have attracted the attention of phytochemistry researchers since 2,4-dihydroxy-(2H)-1,4-benzoxazin-3(4H)-one (DIBOA) and 2,4-dihydroxy-7-methoxy-(2H)-1,4-benzoxazin-3(4H)-one (DIMBOA) were isolated from plants belonging to the Poaceae family. These compounds exhibit interesting biological properties, such as phytotoxic, antimicrobial, antifeedant, antifungal, and insecticidal properties. These chemicals, in addition to a wide variety of related compounds involved in their metabolism, detoxification mechanisms, and degradation on crop soils and other systems, have high interest and in some cases potential agronomic utility. This paper presents a complete review of the methods employed for their synthetic obtention in addition to some of the authors' own contributions to their chemistry. The degradation and phytotoxicity experiments carried out in ongoing research into the potential agronomic utility of these compounds required large amounts of them, which were obtained from natural sources. This paper presents a modified methodology to access DIMBOA from Zea mays cv. Apache and to obtain 2-O-beta-D-glucopyranosyl-2,4-dihydroxy-(2H)-1,4-benzoxazin-3(4H)-one (DIBOA-Glc) and DIBOA from Secale cereale L. New synthetic methodologies were employed for the obtention of the lactams 2-hydroxy-(2H)-1,4-benzoxazin-3(4H)-one and 2-hydroxy-7-methoxy-(2H)-1,4-benzoxazin-3(4H)-one and the malonamic acids N-(2-hydroxyphenyl)malonamic acid and N-(2-hydroxy-7-methoxyphenyl)malonamic acid. The aminophenoxazines 2-amino-7-methoxyphenoxazin-3-one and 2-acetamido-7-methoxyphenoxazin-3-one have been synthesized in the authors' laboratory by novel procedures. All of the methodologies employed allowed the desired compounds to be obtained in high yield and in an easy-to-scale manner.  相似文献
