The objective of this study was to investigate sorption, desorption, and immobilization of Pb in the clay and calcareous loamy sand soils treated with inorganic ligands (NO3?, Cl? and H2PO4?). Pb sorption was also determined in the presence of oxalate and citrate. The maximum Pb sorption capacities (q) ranged from 42.2 to 47.1 mmol kg?1 for the clay soil, and from 45.2 to 47.0 mmol kg?1 for loamy sand soil. It was observed that the binding energy constant (k) for Pb sorbed onto loamy sand soil (528–1061) is higher than that for clay soil (24.38–55.29). The loamy sand soil-sorbed greater quantities of Pb compared to the clay soil when initial pH was ≥ 3. However, it had lower sorption capacity at the lowest initial pH of 2. Additionally, the greatest Pb sorption and immobilization occurred in the soil treated with H2PO4. In the clay soil, the sorption of Pb was depressed at 0.1 mol kg?1 of Cl?, as compared with other ligands. Concerning organic acids, citrate ligand showed the highest decrease in Pb sorption. It could be concluded that the nature of Pb sorption can depend on the type and quantity of ligands present, as well as the soil type.  相似文献

The aim of this study was to estimate the (co)variance components and breeding values for birthweight (BW) in Nellore cattle by considering or not identical weights that exhibit a high frequency within the contemporary group (CG). A total of 175,258 BW records of Nellore cattle born between 2002 and 2018 were used. The CG was formed by farm, year of birth, sex and feeding regime at birth. CGs with more than 16% of identical BW values were eliminated, generating a data file called BWd. Another file was created without removing these animals (BWt). A mixed linear model was used for statistical analysis, which included fixed and random effects. In both data files analysed, single-trait analysis was performed by Bayesian inference. The mean direct and maternal heritability for BW and the correlation between direct and maternal effects were 0.27, 0.07 and −0.07 for BWt, respectively, and 0.30, 0.093 and −0.07 for BWd. This method should affect the estimation of genetic merits of animals for BW, providing greater safety in the choice of sires.  相似文献

The reactivation of the rat brain muscarinic acetylcholine receptor (mACh-R) binding with dimercaptosuccinic acid (DMSA) after in vitro and in vivo inhibition by mercuric chloride (HgCl2) and methylmercuric chloride (MeHg) was investigated. Receptor binding was estimated by the potent and specific antagonist l-[3H]quinuclidinyl benzilate ([3H]QNB). Rat brain synaptosomal membranes were exposed to HgCl2 and MeHg. At 1 × 10?4M. HgCl2 caused complete inhibition of the [3H]QNB binding. The inhibition of [3H]QNB binding by HgCl2 was still higher than 50% at 1 × 10?8M. MeHg caused less inhibition of [3H]QNB binding than HgCl2. The inhibited receptors showed a significant degree of reactivation when treated with DMSA. The recovery was almost complete after MeHg inhibition or with the lower HgCl2 concentrations. Generally, the reactivation was dependent on the concentration of DMSA. When rats injected with either early or delayed doses of DMSA following administration with five consecutive daily doses (8 mg/kg body wt, Gavage method) of MeHg or HgCl2, the inhibition of [3H]QNB binding was less than untreated ones. The early treatment with DMSA decreased the inhibition of [3H]QNB binding due to MeHg or HgCl2 intoxication. However, DMSA was more effective in reducing HgCl2 inhibition than MeHg either in vitro or in vivo treatment. The ability of DMSA to reactivate the mACh-R after inhibition with the mercurials emphasizes the involvement of essential sulfhydryl groups in [3H]QNB binding sites, and also shows that these sulfhydryl groups are the primary target for the MeHg and HgCl2 inhibition of the rat brain muscarinic receptors.  相似文献

The aim of this study was to investigate the incidence of different teat lesions in cows that cause obstruction of milk flow. 18,400 teats were palpated in Konya slaughterhouse. From these 1000 hard milking or obstructed teats were found (5.4%). Based on the radiographical results, distal 1/3, proximal 2/3, complete teat lesions and congenital obstructions were found in 70.1, 16.6 and 0.7% respectively.  相似文献

A Tritrichomonas foetus-specific 5' Taq nuclease assay using a 3' minor groove binder-DNA probe (TaqMan MGB) targeting conserved regions of the internal transcribed spacer-1 (ITS-1) was developed and compared to established diagnostic procedures. Specificity of the assay was evaluated using bovine venereal microflora and a range of related trichomonad species. Assay sensitivity was evaluated with log(10) dilutions of known numbers of cells, and compared to that for microscopy following culture (InPouch TF test kit) and the conventional TFR3-TFR4 PCR assay. The 5' Taq nuclease assay detected a single cell per assay from smegma or mucus which was 2500-fold or 250-fold more sensitive than microscopy following selective culture from smegma or mucus respectively, and 500-fold more sensitive than culture followed by conventional PCR assay. The sensitivity of the conventional PCR assay was comparable to the 5' Taq nuclease assay when testing purified DNA extracted from clinical specimens, whereas the 5' Taq nuclease assay sensitivity improved using crude cell lysates, which were not suitable as template for the conventional PCR assay. Urine was evaluated as a diagnostic specimen providing improved and equivalent levels of T. foetus detection in spiked urine by both microscopy following culture and direct 5' Taq nuclease detection, respectively, compared with smegma and mucus, however inconclusive results were obtained with urine samples from the field study. Diagnostic specimens (n=159) were collected from herds with culture positive animals and of the 14 animals positive by 5' Taq nuclease assay, 3 were confirmed by selective culture/microscopy detection (Fisher's exact test P<0.001). The 5' Taq nuclease assay described here demonstrated superior sensitivity to traditional culture/microscopy and offers advantages over the application of conventional PCR for the detection of T. foetus in clinical samples.  相似文献

Two transgenic strawberry lines (Pel 1 and Pel 3) containing the open reading frame of a fruit specific strawberry pectate lyase gene (FaplC) under the control of the CaMV35S promoter have been obtained to evaluate the role of this gene on fruit softening. Ripen fruits from both lines showed a significant down-regulation of FaplC, being the percentage of silencing of 84 and 71% on Pel 1 and Pel 3, respectively. The agronomic behaviour of transgenic plants was evaluated during two consecutive years. Fruit set increased in the two transgenic lines when compared with control plants, although Pel 1 showed a significant reduction on fruit weight. Firmness of full ripen fruits from Pel lines was significantly higher than control fruits, while color and soluble solids were not affected. The increase of firmness in Pel lines was maintained when ripe fruits were stored for 3 days at 25 °C. Histological analysis of ripe fruits showed lower intercellular spaces and a higher degree of cell to cell contact area in transgenic fruits when compared with controls. Altogether, these results suggest a direct relationship between pectate lyase gene expression and strawberry fruit softening.  相似文献

Lernaea cyprinacea infestation is a major problem for fishing culture in Brazil, which was introduced in the mid-eighties. To attempt controlling this parasite, an evaluation of the activity of the Pinus elliottii resin constituents against it was undertaken. To run the bioassay, fish infested with L. cyprinaceae were transferred to aquaria and kept at room temperature and ventilation for 15 days for adaptation. Afterwards, fish were sacrificed and the parasites were kept under water to run the experiments by evaluating the activities of the steamed oil and the chloroform fraction of the resin at concentrations of 0.5, 1.0, 2.0 and 5.0 ppm. The crude resin was also evaluated at concentrations of 1.0, 5.0 and 10 ppm, as well as the major components of its steamed oil, alpha and beta-pinenes, both at concentrations of 0.5 and 1.0 ppm. The results showed that both fractions from the resin were effective at 0.5 ppm concentration, while the pure compounds were less active. Moreover, the acute toxicity (DL(50)) of the crude resin for the fish Leptorinus piau, aged 1 month, was established at 200.0 ppm, which ensures its safe use.  相似文献
