Induced polyploidy in inter‐subspecific maize hybrids to reduce heterosis breakdown and restore reproductive fertility 下载免费PDF全文

The aim of this study was to reduce the breakdown of hybrid vigour by crossing maize × teosinte subspecies through induced tetraploidy and to improve the biomass and reproductive fertility of the induced tetraploids. To achieve this, seeds of maize breeding lines selected from an open‐pollinated population ‘Sargodha‐2002’ and a maize × teosinte cross were germinated in colchicine solution (0·25, 0·5 or 1·0%) until they had a thick radical and protruded plumule. Colchicine at 0·5% induced the highest number of tetraploids with the lowest number of chimeric plants. The induced tetraploids of maize and maize × teosinte crosses showed a significant (P ≤ 0·05) increase in leaf area (5 and 14%), total soluble solids (20 and 18%), leaf oil percentage (42 and 12%) and leaf crude protein contents (10 and 14%) in leaves relative to the diploid subspecies. Moreover, induced tetraploidy also arrested hybrid vigour by slowing down the decay of heterosis in progenies (4x) for plant height, leaf biomass and leaf oil percentage. In the dog, creatine kinase (CK) is mostly present in the skeletal muscles, myocardium, brain and intestine. The MM isoenzyme predominates in muscles and myocardium. In plasma, reference values depend on the technique used and CK-MB accounts for about 30–45% of total CK activity. Sex has no influence on plasma CK activity, which is higher in young dogs than in adults. Plasma CK is elevated after physical exercise. After its release from the cells, CK reaches the plasma mostly via the lymphatic route and then remains in the plasma compartment. It is rapidly cleared with a half-life of about 2 hours. Muscle diseases are the main source of plasma CK elevations: inherited myopathies, malignant hyperthermia, hypothyroidism, vitamin E-selenium deficiency, prolonged decubitus, intramuscular injections, surgery, etc. Plasma CK is also increased in experimental myocardial infarction, for which the dog is an interesting model, allowing quantification of the damage by measuring the total CK activity released.Abbreviations ADP

A total of 2388 cattle and 442 shelters, from two provinces (Elazig and Malatya) endemic for tropical theileriosis in the east of Turkey, were studied for Hyalomma tick populations from July 1993 to July 1995 in Elazig and from May 1998 to January 1999 in Malatya. Four thousand five hundred and eighty one of 7455 Hyalomma ticks were collected from cattle, the other ticks (2874) were collected from shelters. All of the ticks collected from shelters were Hyalomma anatolicum anatolicum. Two thousand eight hundred and ninety five (63.1%) of 4581 Hyalomma ticks collected from cattle were H.a. anatolicum. 23.8% (1047/4581), 11.7% (536/4581) and 0.6% (3/4581) of Hyalomma ticks were Hyalomma anatolicum excavatum, Hyalomma detritum and Hyalomma marginatum marginatum, respectively. A total of 5909 Hyalomma adult ticks collected from cattle (3362/5909) and shelters (2447/5909) were dissected and salivary glands were stained with Methylgreen/Pyronin method. Thousand one hundred and fifty (46.9%) of 2447 H.a. anatolicum collected from shelters and 412 (19.1%) of 2147 H.a. anatolicum collected from cattle were positive for Theileria infection. Twenty (2.4%) of 820 H.a. excavatum and 23 (4.6%) of 495 H. detritum collected from cattle were positive. The mean number of infected acini per infected male and female ticks collected from cattle were 11.3 and 22.4 in H.a. anatolicum, 4 and 6.8 in H.a. excavatum, 17.9 and 18.3 in H. detritum, respectively. In H.a. anatolicum collected from shelters, the above rates were 11.8 and 17.6 in male and female ticks, respectively. A tumor-like tissue mass of lateral-limbal site in the right eye of a 12-year-old Cob Pony mare was presented for surgical removal. The mass, covering the lateral limbus, bulbar conjunctiva and part of the cornea, was surgically removed, and the corneal defect repaired with a pedicle bulbar conjunctival graft. This study was carried out to determine the prevalence and distribution of tropical theileriosis in cattle in eastern Turkey by microscopical, serological and molecular methods. A total of 1561 whole blood, 1505 serum and 1483 blood smear samples were collected from cattle of various breeds and ages in 11 towns of Eastern Turkey. Theileria annulata piroplasm DNA extracted from cattle blood was amplified by polymerase chain reaction (PCR) using species-specific primers. Serum antibodies against T. annulata were investigated by indirect fluorescence antibody test (IFAT). Blood smears were examined for Theileria piroplasms by microscopical examination (ME). In the examination of DNA extracted from 1561 blood samples, an amplicon with the size of 721bp was obtained in 37.8% (590/1561) of these samples. Serum antibodies against T. annulata and piroplasm of Theileria spp. were detected in 34.9% (526/1505) and 19.7% (293/1483) of the samples, respectively. The differences between ME and PCR results and between ME and IFAT results were statistically significant (P < 0.05). In contrast, there was no significant difference between the PCR and IFAT results. A total of 179 ticks (136 female; 43 male) belonging to Hyalomma spp. were collected from cattle from three towns. Total Suspended particulate matter (TSP) in urban atmosphere of Islamabad was collected using a high volume sampling technique for a period of one year. The nitric acid–perchloric acid extraction method was used and the metal contents were estimated by atomic absorption spectrophotometer. The highest mean concentration was found for Ca at 4.531 µg/m3, followed by Na (3.905 µg/m3), Fe (2.464 µg/m3), Zn (2.311 µg/m3), K (2.086 µg/m3), Mg (0.962 µg/m3), Cu (0.306 µg/m3), Sb (0.157 µg/m3), Pb (0.144 µg/m3) and Sr (0.101 µg/m3). On an average basis, the decreasing metal concentration trend was: Ca > Na > Fe > Zn > K > Mg > Cu > Sb > Pb > Sr > Mn > Co > Ni > Cr > Li > Cd ≈ Ag. The TSP levels varied from a minimum of 41.8 to a maximum of 977 µg/m3, with a mean value of 164 µg/m3, which was found to be higher than WHO primary and secondary standards. The correlation study revealed very strong correlations (r?>?0.71) between Fe–Mn, Sb–Co, Na–K, Mn–Mg, Pb–Cd and Sb–Sr. Among the meteorological parameters, temperature, wind speed and pan evaporation were found to be positively correlated with TSP, Ca, Fe, K, Mg, Mn and Ag, whereas, they exhibited negative relationships with relative humidity. On the other hand, Pb, Sb, Zn, Co, Cd and Li revealed significant positive correlations with relative humidity and negative with temperature, wind speed and pan evaporation. The major sources of airborne trace metals identified with the help of principle component analysis and cluster analysis were industrial emissions, automobile exhaust, biomass burning, oil combustion, fugitive emissions, resuspended soil dust and earth crust. The TSP and selected metals were also studied for seasonal variations, which showed that Na, K, Zn, Cu, Pb, Sb, Sr, Co and Cd peaked during the winter and remained lowest during the summer, while Ca, Fe, Mg and Mn were recorded highest during the spring. 相似文献

Zein is a group of alcohol-soluble corn proteins, which consists of several individual proteins. A single-step gel filtration chromatography method was developed to fractionate individual zeins from ethanol extracts of whole corn. A Superdex prep 75 column was used with different mobile phases to fractionate the zeins, which were analyzed by SDS-PAGE and UV spectrophotometry. With 70% aqueous ethanol as the mobile phase, fractions containing a mixture of alpha-zein/beta-zein and alpha-zein/delta-zein were obtained. With ammonium bicarbonate added to the 70% ethanol mobile phase, it was possible to obtain beta-zein and delta-zein fractions devoid of other proteins. However, all fractions containing alpha-zein also contained minor amounts of delta-zein and/or beta-zein. Almost all fractions also contained non-protein impurities. 相似文献
