Citrus sudden death (CSD) is a disease of unknown etiology that greatly affects sweet oranges grafted on Rangpur lime rootstock, the most important rootstock in Brazilian citriculture. We performed a proteomic analysis to generate information related to this plant pathogen interaction. Protein profiles from healthy, CSD-affected and CSD-tolerant stem barks, were generated using two-dimensional gel electrophoresis. The protein spots were well distributed over a pI range of 3.26 to 9.97 and a molecular weight (MW) range from 7.1 to 120 kDa. The patterns of expressed proteins on 2-DE gels made it possible to distinguish healthy barks from CSD-affected barks. Protein spots with MW around 30 kDa and pI values ranging from 4.5 to 5.2 were down-regulated in the CSD-affected root-stock bark. This set of protein spots was identified as chitinases. Another set of proteins, ranging in pI from 6.1 to 9.6 with an MW of about 20 kDa, were also suppressed in CSD-affected rootstock bark; these were identified as miraculin-like proteins, potential trypsin inhibitors. Down-regulation of chitinases and proteinase inhibitors in CSD-affected plants is relevant since chitinases are well-known pathogenesis-related protein, and their activity against plant pathogens is largely accepted. 相似文献

Corn samples and different commercial dry‐milled fractions collected from an industrial mill in Argentina were surveyed for fungal contamination. The percentage of Fusarium isolates in whole corn kernels among all fungi recovered was 2.0–97.0%; in corn grits, it was 2.6–50.0%. Maximum levels in the other fractions were 5.2 × 105 colony forming units per gram (CFU/g) in germ and bran, 5.0 × 103 CFU/g in C flour, and 2.7 × 103 CFU/g in corn meal. The high initial contamination from whole corn is reflected in germ and bran, which is destined for animal consumption, but not in corn meal. F. verticillioides and Aspergillus flavus were the most frequent species in the whole corn kernel, but F. verticillioides was prevalent in all the other industrial fractions. Other potentially toxigenic fungi that were isolated included Aspergillus parasiticus, Alternaria alternata, Penicillium citrinum, and P. funiculosum. In this first report about mold contamination in corn industrial dry‐milled fractions in Argentina, the high fungal contamination level observed in the stored corn could indicate the necessity to improve the hybrid quality and the storage conditions to diminish the risk of mycotoxin occurrence. 相似文献

The objective of this research was to determine whether computer‐analyzed (objective) mixograph parameters could replace conventional mixograph parameters in the evaluation of flour quality. The 642 hard winter wheat flours, collected from federal regional performance nurseries in 1995 and 1996, were analyzed by a conventional and computerized mixograph. Mixograph bandwidths at 6 min (BW6) showed the most significant linear correlation with subjective mixing tolerance scores (r = 0.81, P < 0.1%, n = 642). Prediction models of conventional and experimental baking parameters were developed by continuum regression using computer‐analyzed mixograph parameters of a calibration set (n = 282). The developed models could estimate conventional mixograph mixing time and tolerance scores, baking water absorption and mixing time, and bread loaf volume, showing R2 values of 0.86, 0.74, 0.68, 0.80, and 0.51, respectively, for a validation set (n = 380). These results indicated that computer‐analyzed mixograph parameters could be applied to develop prediction models to be used for flour quality evaluation in wheat breeding programs. 相似文献
