Experimental studies about the recovery, survival and migration to pasture of cyathostomin infective larvae (L(3)) from fresh feces depositions were conducted from February 2005 to March 2007 in a tropical region of southeast Brazil. Grass and feces were collected weekly at 8 a.m., 1 and 5 p.m. and processed by the Baermann technique. Multivariate analysis (principal components method) showed the influence of time and environmental variables on the number of infective larvae recovered from the feces and pasture. In the rainy period (October-March), more infective larvae were recovered on the feces and grass apex. In contrast, in the dry period (April-September), the recovery was higher only on the grass base, as well as the L(3) survival on feces and grass. More larvae were recovered at 8 a.m., except from the grass apex, where the highest recovery was at 1 p.m. Few studies investigating the seasonal transmission of equine cyathostomin have been conducted in South American tropical climates. These results demonstrate that in tropical conditions L(3) are available on feces and pasture throughout the year. Knowledge of climatic influences on the development and survival of L(3) is crucial to designing integrated parasite control programs that provide effective protection while slowing the development of anthelmintic resistance.  相似文献

The present work aimed to evaluate the endectocide activity of a new injectable long-action formulation, containing ivermectin (IVM) and abamectin (ABA). In each one of the four experiments performed, the following groups were formed: group I: 2.25% IVM (450 microg/kg)+1.25% ABA (250 microg/kg), group II: 3.15% IVM (630 microg/kg) and group III: control. Eighteen bovine naturally infected by gastrointestinal nematoda were selected for anthelmintic evaluation and necropsied on posttreatment day (PTD) 14 to estimate the total parasitic burden. For the Rhipicephalus (Boophilus) microplus field trial, 30 bovine were selected by means of counts of semi-engorged R. (B.) microplus and the therapeutic and residual efficacy evaluated by tick counts on PTDs 1, 3, 7, 14, 21, 28, 35, 42, 49, 56, 63, 70, 77, 84 and 91. In the stall test, 15 calves were artificially infested with 5000 R. (B.) microplus (Mozzo strain) larvae three times a week and daily collections of all the engorged female ticks detached from each calf were performed until the PTD 80. Forty bovine naturally infected with Dermatobia hominis larvae were selected and the number of larvae was counted by visual and tactile inspection on PTDs 3, 7, 14, 28, 35, 49, 63, 77, 91 and 105. In this trial, a formulation containing 1% doramectin (200 microg/kg) was also used. IVM+ABA formulation and 3.15% IVM eliminated four of the eight species of nematode identified. The anthelmintic efficacy of the avermectins association against Haemonchus placei, Cooperia spatulata and C. punctata was 89.64%, 98.84% and 97.69%, while 3.15% IVM achieved 30.98%, 84.79% and 75.56%, respectively. The two formulations evaluated showed reduced acaricide action on the PTD 1 and 3, reaching high efficacy percentages from PTD 14 onward. The IVM+ABA showed efficacy above 95% in the period between PTDs 21 and 49. In the stall test, it observed no difference (P>0.05) between the two formulations regarding the R. (B.) microplus counts during the entire evaluation period. IVM+ABA reduced the number of ticks from the PTD 1 to 77 (P<0.05) and 3.15% IVM reduced (P<0.05) the tick number from PTD 4 up to PTD 80. The three endectocides showed no difference (P>0.05) regarding the number of D. hominis larvae and prevented this parasite reestablishment until PTD 105. These results indicate that the IVM+ABA association showed higher anthelmintic activity and similar efficacy against arthropods to the formulation containing 3.15% IVM.  相似文献

Clostridium difficile was investigated as a possible cause of enteritis in calves. The organism and its toxins (TcdA and TcdB), respectively, were found in 25.3% and 22.9% of stool samples from diarrheic calves. Culture positive samples were more likely than culture negative samples to be toxin positive. However, toxin positive stools were more common among nondiarrheic calves, but diarrheic calves were nearly twice as likely to be culture positive. Ribotype 078 was dominant among isolates. Salmonella sp. was isolated from both diarrheic and nondiarrheic calves, but large numbers of E. coli were found more commonly in diarrheic calves than in nondiarrheic animals. Prevalence rates for coronavirus and Cryptosporidium sp. were substantially higher in nondiarrheic calves than in diarrheic, but rates of detection of rotavirus and Giardia sp. were more nearly equal between groups. Lesions in naturally infected calves included superficial mucosal erosion with associated fibrinous exudates. Neutrophils and eosinophils infiltrated lamina propria. Large Gram-positive rods morphologically compatible with C. difficile were abundant in the colonic lumen and the organism was isolated by bacteriologic culture. Toxins were found throughout the colon. Purified toxins A and B (individually and conjointly) caused comparable lesions, as well as fluid accumulation, in ligated intestinal loops. The dissipation rate of endosulfan isomers (α and β) in seawater and sediment was studied. The disappearance rate of both isomers from seawater and pure water was compared, and the same measurements were made in both sterile and unsterile marine sediment. Flasks of water and sediment, fortified with a dispersion of a commercial endosulfan 350 g litre−1 EC, Protodan 35®, were incubated under laboratory light at room temperature for 82 days. A micro on‐line extraction method and GC‐ECD was used to determine the pesticide and its metabolites. The dissipation of endosulfan (in two phases of first‐order kinetics) occurred more rapidly in seawater than in pure water. At the end of the experiment, the concentration of α‐endosulfan in sterile sediment was four times greater than in unsterile sediment, while the dissipation rate of β‐endosulfan in unsterile sediment was approximately double that observed in sterile sediment. The dissipation of both forms in sediment occurred in a single stage. Endosulfan β‐isomer was more persistent than α‐isomer in both sterile and unsterile sediment. Dissipation of endosulfan sum of α‐ and β‐isomers in sediment at the end of the experiment ranged from 80% (sterile) to 95% (unsterile). Honey bees (Apis mellifera L. Hymeoptera: Apidae) use hydrogen peroxide (synthesized by excreted glucose oxidase) as an important component of social immunity. However, both tolerance of hydrogen peroxide and the production of glucose oxidase in honey is costly. Hydrogen peroxide may also be encountered by honey bees at high concentrations in nectar while foraging, however despite its presence both in their foraged and stored foods, it is unclear if and how bees monitor concentrations of, and their behavioral responses to, hydrogen peroxide. The costs of glucose oxidase production and the presence of hydrogen peroxide in both nectar and honey suggest hypotheses that honey bees preferentially forage on hydrogen peroxide supplemented feed syrups at certain concentrations, and avoid feed syrups supplemented with hydrogen peroxide at concentrations above some tolerance threshold. We test these hypotheses and find that, counter to expectation, honey bees avoid glucose solutions supplemented with field-relevant hydrogen peroxide concentrations and either avoid or don’t differentiate supplemented sucrose solutions when given choice assays. This is despite honey bees showing high tolerance for hydrogen peroxide in feed solutions, with no elevated mortality until concentrations of hydrogen peroxide exceed 1% (v/v) in solution, with survival apparent even at concentrations up to 10%. The behavioral interaction of honey bees with hydrogen peroxide during both within-colony synthesis in honey and when foraging on nectar therefore likely relies on interactions with other indicator molecules, and maybe constrained evolutionarily in its plasticity, representing a constitutive immune mechanism.  相似文献

If we are to breed common bean (Phaseolus vulgaris L.) for durable resistance to diseases, we must understand pathogenic variation and find sources of resistance. Our first objective was to determine the patterns of pathogenic variation found among isolates of Phaeoisariopsis griseola (PG), the fungus that causes angular leaf spot (ALS) in common bean. We characterized 433 PG isolates from 11 Latin American and 10 African countries, using differential cultivars, isozymes, and/or random amplified polymorphic DNA (RAPD) markers. We also systematically screened, for ALS resistance, common bean accessions from the world collection held at CIAT, and assessed the progress so far made in breeding for resistance to ALS. Despite their great diversity within and between countries on both continents, the PG isolates were classified into two major groups: Andean, and Middle American. Although each group had internal differences for virulence, and biochemical and molecular characteristics, the ‘Andean’ PG isolates were more virulent on common beans of Andean origin, than on those of Middle American origin, thus, suggesting a host-pathogen co-evolution. The ‘Middle American’ PG isolates, although more virulent on common beans from Middle America, also attacked Andean beans, thus, exhibiting a much broader virulence spectrum. To find sources of resistance, we tested 22,832 common bean accessions against naturally occurring PG isolates in the field at CIAT's Experiment Station, Quilichao, Colombia, between 1985 and 1992. The resulting 123 intermediate (scores of 4 to 6) and resistant (scores of 1 to 3) accessions were then tested in the greenhouse against selected 14 PG isolates of diverse origins. Nineteen accessions were intermediate or resistant to at least 13 of 14 PG isolates. Similarly, of 13,219 bred lines tested in the field between 1978 and 1996, 89 were intermediate or resistant. Of these, 33 bred lines proved intermediate or resistant to at least eight of nine PG isolates to which they were challenged in the greenhouse. We suggest that, to breed for durable resistance to ALS, common bean populations should be developed from crosses between Andean and Middle American gene pools. The populations should then be systematically evaluated and selected against the broadest range of the most virulent PG isolates of diverse evolutionary origins. Classically, selection for superior genotypes in cacao has been based on the successive harvest records across a number of years. Little information on the minimum duration of these harvest periods is available in the literature. The repeatability coefficient (ρ) was used to estimate this period. Twenty five cacao genotypes were assayed in a randomized block design with four replications and 16-plant plots. The following yield components were studied: number of healthy fruits per plant, number of collected fruits per plant, weight of humid seeds per plant and per fruit, and percentage of diseased fruits per plant, over 5 years (1986–90). Repeatability estimates were higher than 0.84 for all components, except percentage of diseased fruits per plant (^ρ - 0.41). With such estimates, it is possible to select genotypes on the basis of only two years of successive harvests, with a determination coefficient of 90%. The advantages of applying the repeatability coefficient to the cacao breeding program are discussed. In small volcanic islands the local scale climate is influenced by the regional scale climate and by the orography and orientation of air masses movement over the islands. A model was developed in a GIS environment to generate local scale climate variables from those observed at the synoptic scale, from coastal weather stations. An advective submodel, based on the Foehn effect and assuming the conservation of mass and energy, computes local scale air temperature, relative humidity, clouds occurrence and precipitation. A radiative submodel, using information generated by the advective submodel, computes local scale global radiation. A rotational terrain model allows that computations be performed according to the direction of wind. Because the model works within a GIS, results concern the spatial distribution of all climatic variables on the island territory. Results of the validation of temperature, relative humidity, global radiation and rainfall are presented. For agro-meteorological purposes, an application of generated data to perform the sequential water balance is also analysed by comparing results from computations using simulated and observed data at a control weather station located at medium altitude. Results support assumptions utilised in the model and the further use of generated local climate fields for water management and environmental studies in small island environments.  相似文献

This study investigates the plastic responses after shoot removal of Amaranthus cruentus L. and A. hypochondriacus L. This practice is common in the Sierra Norte de Puebla, Mexico, where both species are cultivated and used as a food source during the vegetative stage. Empirical observations indicate that biomass increases in clipped plants. The experimental study carried out on both species of Amaranthus imitated local farm management. Three levels of shoot removal were applied to plants of both species: Control, 10% and 40%. In A. cruentus plants with 10% of the shoot removed produced more stalk biomass, more leaves, greater leaf area, greater length of primary branches and a larger number of secondary branches than the control plants. In A. hypochondriacus, there were no differences among levels of shoot removal in stem biomass; in general, the number of primary and secondary branches in control plants was greater than that in clipped plants. Patterns of biomass allocation revealed important differences between species, as clipped plants allocated more biomass to seeds in A. cruentus, while control plants allocated more to leaves in A. hypochondriacus. Furthermore, changes in response at the primary branch (secondary branches, leaf biomass, and area) level were observed between species. In general, clipped plants of A. cruentus overcompensate for the damage, while this response was not evident in clipped plants of A. hypochondriacus. Instead, this species showed high tolerance to damage. The phenotypically plastic responses expressed for both species can be used as a guide to improve management of these plants by local farmers from the Sierra.  相似文献
