DREB(Dehydrate responsive element binding factor)转录因子是植物非生物逆境适应中的关键调节因子,该类转录因子具有保守的AP2/EREBP结构域,在低温、高盐等非生物逆境胁迫下,可调控下游逆境应答基因的表达,对增强植物的抗逆能力有重要作用。黑果枸杞具有极强的耐干旱、盐碱能力,为研究黑果枸杞DREB类基因在低温、盐碱响应中的功能,本研究以强抗盐灌木黑果枸杞总RNA为模板,基于前期转录组测序拼接序列,利用RT-PCR方法克隆得到一条DREB基因。该基因含有一个1116 bp的开放阅读框,编码372个氨基酸。系统进化树分析显示,Lr DREB1基因属于DREB亚家族A2组成员,与拟南芥AtDREB2B、AtDREB2E具有较高的相似性。荧光定量PCR结果显示,Lr DREB1受盐胁迫、ABA胁迫和低温胁迫诱导表达,可能参与依赖ABA的信号转导途径,调节黑果枸杞抗盐响应。本研究通过对黑果枸杞LrDREB1基因的克隆、序列分析和表达分析,为研究Lr DREB1的功能及黑果枸杞抗盐机理提供了帮助。 相似文献

An experiment was carried out to evaluate fermented soybean meal and squid by‐product blend (1:1) (FP) as replacement of fishmeal (FM) for Japanese flounder, Paralichthys olivaceus. Five isocaloric (19 kJ g?1) diets were prepared by replacing 0 (FP0), 12 (FP12), 24 (FP24), 36 (FP36) and 48% (FP48) FM protein with FP. Triplicate groups of juveniles (mean weight of 3.9 g) were delivered the test diets for 8 weeks in a flow‐through sea water system. The results showed that there were no significant differences (P > 0.05) among the growth rates of fish fed FP0, FP12, FP24 and FP36 diets. Growth and nutrient utilization parameters were significantly reduced in fish fed FP48 diet. Although, whole body proximate composition of fish were not significantly affected by the dietary treatments compared to the control; methionine and phenylalanine contents were significantly decreased in FP48 group. Protein retention was also significantly decreased in the similar group of fish. Dietary treatments did not alter most of the plasma metabolites, while some of the health parameters were improved in the replacement groups. Results suggested that FP is a potential candidate for alternative protein ingredient in aquafeed and can replace 36% FM protein in the diet of Japanese flounder. 相似文献

Objectives of our studies were to quantify effects of salinity on growth and nitrogen metabolism of wheat and to measure variation in response of different cultivars, hybrids, and classes. Methods and criteria for identifying resistance to salinity in wheat, particularly effects on nitrogen metabolism also were tested. Variation in response to salinity was measured by subjecting seedlings of six wheats to one control treatment (‐0.1 bars) and two stress treatments (‐3.5 and ‐10.4 bars) from NaCl, MgSO4, and MgCl2 in hydroponic solutions. Both stress treatments retarded growth; wheats significantly varied at ‐3.5 bars but not at ‐10.4 bars. Stress decreased root and shoot nitrate N and total N contents. Studies with one wheat cultivar showed that salinity decreased activity of nitrate reductase enzyme and stimulated accumulation of proline. Salinity more adversely affected vegetative stages than reproductive stages of plants grown to maturity. We concluded that salinity affected wheat by both osmotic effects and antagonism of nitrate metabolism from chloride. Absolute growth and relative growth at different stress levels were superior to differences in nitrogen metabolism as selection criteria for salinity tolerance. 相似文献

该研究优化了由包裹橡皮的金属小球、压电式加速度传感器、电荷放大器、光电式触发电路、数据采集卡和计算机等组成的西瓜声学特性测试系统。由所获取的声波信号幅值谱计算出声透过率,采用TQ软件中SMLR(逐步多元线性回归)函数选取6个特征频率:752、869、1001、4556、322、3950Hz,由其对应的声透过率值建立了西瓜品质检测的多元线性回归模型。对47个西瓜样本的试验数据分析表明:将敲击点和接收点分别放置在西瓜自然生长状态的中部对侧可获得最佳的测定模型,模型的校正相关系数R、校正均方根误差RMSEC和预测均方根误差RMSEP分别是0.80753、0.646和0.655。实现了西瓜糖度检测目的,为声学无损检测西瓜成熟度提供了参考。 相似文献

水果的缺陷、大小和颜色差异以及光照等因素影响图像背景分割与目标提取精度.以苹果为研究对象,针对4种不同光照强度条件下采集的280幅不同姿态的苹果图像,将彩色图像的R、G、B分量进行算术运算,然后采用形态学开运算进行消噪处理,采用线性空间滤波消除锯齿状边界,采用自动阈值分割方法进行背景分割与目标提取.结果显示,203幅图像的分割偏差小于1%,占总量的72.5%;70幅图像的分割偏差大于1%而小于2%,占总量的25%;偏差大于2%的有7幅,占总量的2.5%.最大分割偏差为2.83%. 相似文献

Curcumin [1,7-bis(4-hydroxy-3-methoxyphenyl)-1,6-heptadiene-3, 5-dione], the main constituent of the rhizomes of the plant Curcume longa L. (turmeric), is a powerful antioxidant in both enzymatic and nonenzymatic systems. The interactions of curcumin with egg and soy phosphatidylcholine were followed by fluorescence spectroscopy. Curcumin had very weak fluorescence in aqueous system, which was enhanced in apolar environments. Curcumin emitted at 490 nm after being excited at 451 nm in phosphatidylcholine micelles. The equilibrium constants for the interaction of curcumin with egg and soy phosphatidylcholine were (3.26 +/- 0.2) x 10(5) and (2.64 +/- 0.2) x 10(5) M(-1), respectively. From the Scatchard plot of the fluorometric data, it was inferred that one molecule of curcumin could bind six molecules of phosphatidylcholine. The equilibrium constant for the phosphatidylcholine-curcumin interaction decreased with temperature, indicating the amphiphilic nature of curcumin. The DeltaG, DeltaH, and DeltaS values obtained for the interaction of egg phosphatidylcholine-curcumin were -7.8 +/- 0.3 kcal/mol, -9.6 +/- 0.4 kcal/mol, and -6.8 +/- 0.2 cal/mol/K, respectively. The fluorescence anisotropy measurements of curcumin with phosphatidylcholine suggested that the anisotropy of the curcumin molecule did not change in phosphatidylcholine. The interaction of divalent metal ions with phosphatidylcholine-curcumin in comparison with phosphatidylcholine-1-anilino-8-naphathalenesulfonic acid complex suggested the strong binding of curcumin to metal ions. 相似文献

Summary There were significant differences among pigeonpea [Cajanus cajan (L.) Millsp] Rhizobium sp. strains (IC 3506, IC 3484, IC 3195, and IC 3087) in their ability to nodulate and fix N2 under saline conditions. Pigeonpea plants inoculated with IC 3087 and IC 3506 were less affected in growth by salinity levels of 6 and 8 dS m-1 than plants inoculated with the other strains. For IC 3506, IC 3484, and IC 3195, there was a decrease in the number of nodules with increasing salinity, while the average nodule dry weight and the specific nitrogenase activity remained unaffected. However, in IC 3087, the number of nodules increased slightly with increasing salinity. Leaf-P concentrations increased with salinity in the inoculated plants irrespective of the Rhizobium sp. strain, and leaf-N concentrations decreased with increasing salinity in IC 3484 and IC 3195 only. Shoot-Na and-Cl levels were further increased in these salt-sensitive strains only at 8 dS m-1. Therefore there may be scope for selecting pigeonpea Rhizobium sp. symbioses better adapted to saline conditions. Objects displaced intermittently across the visual field will nonetheless give an illusion of continuous motion [called apparent motion (AM)] under many common conditions. It is believed that form perception is of minor importance in determining AM, and that AM is mediated by motion-sensitive areas in the "where" pathway of the cortex. However, form and motion typically interact in specific ways when natural objects move through the environment. We used functional magnetic resonance imaging to measure cortical activation to long-range AM, compared to short-range AM and flicker, while we varied stability of structural differences between forms. Long-range AM activated the anterior-temporal lobe in the visual ventral pathway, and the response varied according to the form stability. The results suggest that long-range AM is associated with neural systems for form perception. 相似文献
