A continuous source of Bose-Einstein condensed sodium atoms was created by periodically replenishing a condensate held in an optical dipole trap with new condensates delivered using optical tweezers. The volatile production characteristics of ring rot (Corynebacterium sepedonicum) and soft rot (Erwinia carotovora) infected potatoes were investigated under controlled conditions in order to assess the monitoring of volatiles as a technique for differentiating between the infections in stored potatoes. Total volatile production/unit time was much greater from soft rot due to more rapid development after infection. Total volatile production during the period required for the development of the soft rot infection was similar to the output detected during the longer period required for the development of the ring rot infection. The volatile profiles of the two pathogens featured a number of commonly shared metabolites, but one compound was unique to the ring rot infections and two compounds were unique to the soft rot infections. There were also consistent disease-specific changes in the relative concentration of many of the metabolites detected in the volatile profiles.  相似文献

Mefluidide {N-[2,4-dimethyl-5-[[(trifluoromethyl) sulfonyl] amino] phenyl] -acetamide}, a chemical with plant growth regulator properties, was applied to the foliage of potatoes (Solanum tuberosum L.) in field experiments to determine its effects on tuber development, maturity, and storage of the cultivars Russet Burbank and Norland. The rate of sucrose loss before harvest was reduced in mefluidide-treated plants. Total and marketable yields generally were lower in treated plants. Specific gravity tended to increase in Russet Burbank with application rates up to 1.0 kg ai/ha mefluidide. Preharvest treatments of mefluidide resulted in an increase in periderm thickness and russeting. However, weight loss during storage was greater in tubers from treated plants. Less sprouting occurred in storage in tubers from mefluidide-treated plants. Mefluidide resulted in a lower anthocyanin content and consequently an undesirable color change in Norland tubers.  相似文献

Amisk was registered in Canada as a clonal variant of Ranger Russet in 1993. Amisk is similar to Ranger Russet in foliage and tuber morphology and DNA analysis did not detect a polymorphism. However, under Canadian prairie conditions, four years of field data indicate that the yield and number of tubers >88 mm is higher for the Amisk clone than for the 1982 clone of Ranger Russet, second growth occurs less frequently and Amisk is more resistant to tuber invasion by verticillium and fusarium wilt pathogens.  相似文献

Prunus africana, from the moist highlands of sub-Saharan Africa, is subject to great exploitation because its bark is used in the treatment of prostate-related diseases. A better understanding of the biology of its seed could help the ex situ and in situ conservation of the species. Fifty per cent of fresh seeds (46.8% MC) from mature purple fruits harvested in Kenya germinated after 10 days incubation at 25°C. The germination rate was linearly related to constant temperature, giving a prediction for the minimum temperature for germination at ca. 0.6°C. All seeds germinated at 5°C during 4 months fully hydrated storage. P. africana seed is, thus, neither dormant nor chilling sensitive. The relationship between equilibrium moisture content and relative humidity was used to construct a sorption isotherm, against which seed desiccation and storage responses were assessed. Seventy six per cent of mature seeds survived drying to 4.5% MC (22% RH) compared to 17% of seeds from immature purple-green fruits. Although, 25–45% of mature seeds survived at 8–12% MC after 2 months, only 10% remained viable after 6-month storage at 5°C. This study proposes optimum conditions for the artificial regeneration of P. africana seeds and highlights the need for further experiments storing dry seeds at sub-zero temperatures.  相似文献

Root and hypocotyl elongation, water status and solute accumulation were studied in osmotically stressed seedlings of the tropical tree, Colophospermum mopane (Kirk ex Benth.) Kirk ex J. Léonard, which grows in hot arid areas of southern and central Africa. Seeds were imbibed for 24 h and then subjected to a polyethylene-glycol-generated osmotic stress of -0.03 (control), -0.2, -0.8, -1.6 or -2.0 MPa for 60 h. Seedlings subjected to moderate water stress (-0.2 MPa) had higher root growth rates (2.41 +/- 0.24 mm h(-1)), greater final root lengths (111 +/- 3.8 mm) and longer cells immediately behind the root elongation zone than control seedlings (1.70 +/- 0.15 mm h(-1) and 93 +/- 3.9 mm, respectively). Root lengths of seedlings in the -0.8 and -1.6 MPa treatments were similar to those of control seedlings, whereas the -2.0 MPa seedlings had significantly shorter roots. Both root and hypocotyl tissues exhibited considerable osmotic adjustment to the external water potential treatments. Seedlings in the -0.03, -0.2, and -0.8 MPa treatments had similar cell turgor pressures (0.69 +/- 0.10, 0.68 +/- 0.07 and 0.57 +/- 0.04 MPa, respectively), whereas the -2.0 MPa treatment lowered cell turgor pressure to 0.17 +/- 0.04 MPa. Root vacuolar osmotic pressures were generally similar to sap osmotic pressures, indicating that the increased root elongation observed in moderately water-stressed seedlings was not caused by increased turgor pressure difference. Neutral-fraction solute concentrations, including the osmoticum pinitol, increased approximately two-fold in root sap in response to a low external water potential. In hypocotyl sap of seedlings in the -2.0 MPa treatment, pinitol more than doubled, sucrose increased from about 2 to 75 mol m(-3) but glucose and fructose remained unchanged and, as a result, total sugars increased only slightly. The benefits of rapid early root elongation and osmoticum accumulation under conditions of water stress are discussed in relation to seedling establishment.  相似文献

The fatigue and creep performance of MDF, OSB and chipboard have been examined in two environments, namely 65%RH (standard environment) and 85%RH (high humidity). Parallel fatigue and creep tests have been performed in four-point bending on the three wood-based panel products in the two environments. The constant 65%RH environment is service class 1 and the constant 85% RH environment is indicative of a service class 2 environment as detailed in Eurocode 5. The non-interruptive technique of stress-strain hysteresis loop capture has been utilised to follow property changes of the fatigue samples during cyclic loading at a stress ratio of R equal to 0.1. Loop parameters such as loop area, dynamic modulus, and fatigue modulus have been used to characterise the response of these materials to fatigue loads in the two environments. Creep microstrains for the creep samples were recorded in parallel with the fatigue parameters. Fatigue and creep results at 85%RH were more variable than those reported at 65%RH for MDF, OSB and chipboard. In general, at R=0.1 and 85%RH, fatigue and creep microstrains were higher, dynamic stiffnesses were lower and hysteresis loop areas were higher than corresponding properties measured at 65%RH. MDF and chipboard were less moisture tolerant than OSB, this is reflected in the large changes in fatigue and creep parameters.  相似文献

To identify emerging Escherichia coli that have the potential to cause diarrhea in pigs, the prevalence of E. coli pathotypes was determined among 170 and 120 isolates from diarrheic and nondiarrheic piglets, respectively. The isolates were tested for F4, F5, F6, F18, and F41 fimbriae, for E. coli attaching and effacing (EAE), porcine attaching and effacing-associated (Paa), and adhesin involved in diffuse adherence (AIDA-I) factors, for LT, STa, STb, and enteroaggregative heat-stable (EAST1) enterotoxins, and for Shiga toxins (Stxl, Stx2, and Stx2e), using DNA hybridization and polymerase chain reaction. All isolates were O-serotyped and tested for antibiotic resistance against 10 drugs. Seventeen different pathotypes, accounting for 40.0% of the isolates, were recovered from diarrheic piglets. The main pathotypes included EAST1 (13.5%), F4/LT/STb/EAST1 (6.5%), AIDA-I/STb/EAST1 (4.1%), F5/STa (2.9%), EAE/EAST1 (2.9%), and AIDA-I/F18 (2.3%). Only 3 pathotypes, EAE (11.7%), EAST1 (10.8%), and EAE/EAST1 (3.3%), were recovered from nondiarrheic piglets. Paa factor was detected in 8.8% and 7.5% of isolates from diarrheic and nondiarrheic piglets, respectively, and always was associated with other virulence determinants. Overall, 22.9% of isolates from diarrheic piglets appeared to be enteropathogens: enterotoxigenic E. coli (11.7%), enteropathogenic E. coli (3.5%), and E. coli isolates (3.0%) for which none of the above adherence factors was detected. Pathotypes AIDA-I/STb/EAST1 and AIDA-I/STb were isolated only from diarrheic piglets and accounted for 4.7% of isolates. Strains of these pathotypes induced diarrhea when inoculated into newborn colostrum-deprived pigs, in contrast to an isolate positive only for EAST1, which did not induce diarrhea. Antibiotic sensitivity test showed that isolates of the AIDA-I/STb/EAST1 and AIDA-I/STb pathotypes were the only strains sensitive to enrofloxacin, gentamicin, neomycin, and trimethoprim-sulfamethoxazole. This study showed that at least 20.5% of isolates from diarrheic piglets appeared to be associated with AIDA-I/STb pathotype and that EAST1 pathotype is probably not an important marker for diarrhea in piglets.  相似文献

The effect of low storage temperature (4 to 8 C) and maleic hydrazide (MH) treatment on sprouting, sucrose and reducing sugar (glucose and fructose) concentrations in tubers, and fry color-sugar relationships of Shepody were determined and compared to Russet Burbank. Sprouting of Shepody was delayed by 3 to 15 weeks when the temperature was lowered from 8 to 6 or 5 C, respectively. Shepody accumulated more sugars at all storage temperatures than Russet Burbank. The reducing sugars accumulated in Shepody at 5 and 6 C could be lowered to levels acceptable for processing for about 24 weeks after harvest by reconditioning at 18 C for at least two weeks. MH increased glucose in Shepody stored at 5 C compared to untreated tubers. Excessive sprouting (> 10 cm sprout length) of nonsprout inhibited Shepody occured late in storage with four weeks reconditioning. Although not statistically significant, MH tended to cause darker fry color per unit of reducing sugar in Shepody compared to tubers from untreated plants. Storage of Shepody tubers not treated with MH could be extended a few weeks by temperatures < 8 C only if there is precise control over temperature, sugars are closely monitored, and reconditioning is rapid to minimize sprouting.  相似文献
