Ethylene action can be counteracted by 1-methylcyclopropene (1-MCP), which has been used during postharvest storage to maintain quality. In this work, we evaluated the effect of 1-MCP treatments on eggplant quality and phenolic metabolism during refrigerated storage. Eggplants (cv. Lucía) were harvested at commercial maturity, treated with 1-MCP (1 μL/L, 12 h at 20 °C), stored at 10 °C for 21 d and subsequently held at 20 °C for 2 d. Corresponding controls were stored at 10 °C and then transferred to 20 °C for 2 d. During storage calyx color, damage and chlorophyll content, fruit weight loss and firmness, pulp sugar content, acidity, browning and total phenolics were measured. In addition, polyphenol oxidase (PPO), pyrogallol peroxidase (POD), and phenylalanine ammonia-lyase (PAL) activities were evaluated. Fruit calyxes showed reduced damage and remained greener in 1-MCP treated than in control fruit. 1-MCP treated eggplants showed lower weight loss. Pulp browning was clearly prevented as a consequence of 1-MCP exposure, and this was associated with delayed senescence, lower accumulation of total phenolics and reduced activity of PAL. The activity of the enzymes PPO and POD involved in the oxidation of phenolics compounds was also decreased in 1-MCP treated fruit. Results suggest that 1-MCP treatments delay senescence, prevent browning and are beneficial to complement low temperature storage and maintain quality of non-climacteric eggplant fruit.  相似文献

The SALTMED model is one of the few available generic models that can be used to simulate crop growth with an integrated approach that accounts for water, crop, soil, and field management. It is a physically based model using the well-known water and solute transport, evapotranspiration, and water uptake equations. In this paper, the model simulated chickpea growth under different irrigation regimes and a Mediterranean climate. Five different chickpea varieties were studied under irrigation regimes ranging from rainfed to 100 % crop water requirements, in a dry and a wet year. The calibration of the model using one of the chickpea varieties was sufficient for simulating the other varieties, not requiring a specific calibration for each individual chickpea variety. The results of calibration and validation of the SALTMED model showed that the model can simulate very accurately soil moisture content, grain yield, and total dry biomass of different chickpea varieties, in both wet and dry years. This new version of the SALTMED model (v. 3.02.09) has more features and possibilities than the previous versions, providing academics and professionals with a very good tool to manage water, soil, and crops.  相似文献

Chickpea (Cicer arietinum L.) is one of the most important pulse crops in the world, cultivated on a wide range of environments. In Mediterranean regions, it is traditionally grown as a spring-sown rainfed crop, very dependent on rainfall. In this situation, supplemental irrigation can improve significantly the crop yield. The objective of this study was to evaluate the improvement on chickpea crop yield and water productivity (WP) of five chickpea varieties with supplemental irrigation, in the Mediterranean conditions, with both dry and wet years. Field tests were carried out over two cropping seasons, in Southern Portugal, using three kabuli-type and two desi-type chickpea varieties and four irrigation treatments, corresponding to 100, 50, 25 % of crop irrigation requirements (IR) and rainfed. The results show that all chickpea varieties responded to supplemental irrigation with the increase in grain and biomass yield. However, the magnitude of individual chickpea response depends on the year and the genotype. In 2009, a dry year, the highest WP values were attained at the 50 % IR treatment, whereas in 2010, a wet year, it was the rainfed treatment that showed the highest WP values. The Elixir variety showed the best grain yields and water productivity.  相似文献

Some outbreaks involving highly pathogenic avian influenza viruses (HPAIV) of subtypes H5 and H7 were caused by avian-to-human transmissions. In nature, different influenza A viruses can reassort leading to new viruses with new characteristics. We decided to investigate the impact that the NS-segment of H5 HPAIV would have on viral pathogenicity of a classical avian H7 HPAIV in poultry, a natural host. We focussed this study based on our previous work that demonstrated that single reassortment of the NS-segment from an H5 HPAIV into an H7 HPAIV changes the ability of the virus to replicate in mammalian hosts. Our present data show that two different H7-viruses containing an NS-segment from H5–types (FPV NS GD or FPV NS VN) show an overall highly pathogenic phenotype compared with the wild type H7–virus (FPV), as characterized by higher viral shedding and earlier manifestation of clinical signs. Correlating with the latter, higher amounts of IFN-β mRNA were detected in the blood of NS-reassortant infected birds, 48 h post-infection (pi). Although lymphopenia was detected in chickens from all AIV-infected groups, also 48 h pi those animals challenged with NS-reassortant viruses showed an increase of peripheral monocyte/macrophage-like cells expressing high levels of IL-1β, as determined by flow cytometry. Taken together, these findings highlight the importance of the NS-segment in viral pathogenicity which is directly involved in triggering antiviral and pro-inflammatory cytokines found during HPAIV pathogenesis in chickens.  相似文献

Pedro Ximenez sweet wines obtained following the typical criaderas and solera method for sherry wines and subjected to oxidative aging for 0, 1.3, 4.2, 7.0, or 11.5 years were studied in terms of color and aroma fraction by using the CIELab method and gas chromatography, respectively. The parameters defining the CIELab color space (a*, b*, and L*) were subjected to a multiple-range test (p<0.05) that allowed discrimination of the five wine aging levels studied into five uniform groups according to aging time. The aroma fraction was found to include 15 active odorant compounds with OAV > 1 that enriched the wines with fruity, fatty, floral, and balsamic notes during the aging process. The changes in color parameters and active odorants were not linearly related to aging time, being especially marked during the first 1.3 years and then less substantial up to the 7 years, the oldest wines exhibiting sensorial properties markedly departing from all others. For the wines aged over 1.3 years (minimum aging), 2,3-butanedione, linalool, and decanal can be used as reliable fingerprints of the older wines' quality.  相似文献

Formalin-fixed paraffin-embedded tissues (FFPET) represent the largest source of archival biological material available for genomic studies. In this work we present an advanced protocol for extraction of high quality DNA from FFPET that can be applied in several molecular studies. Although cat mammary tumours (CMT) are the third most frequent tumour in cats the recovery of significant number of samples for molecular studies are in some way restricted to FFPET samples. We were able to obtain high quality DNA from FFPET of thirty six CMT that were subjected to pre-fixation and fixation processes routinely used in the veterinary hospitals. The quality of DNA obtained was tested by PCR amplification using six sets of primers that amplify single-copy fragments. The DNA fragments obtained were further sequenced. This protocol was able to provide FFPET gDNA that can be amplified and sequenced for larger fragments up to 1182bp.  相似文献

In Brazil, increased leaf spot disease (Pyrenophora chaetomioides Speg.) frequency and intensity on cultivated spring oat (Avena sativa) requires that plant pathologists and breeders rapidly identify oat genotypes with greater resistance. Criteria are needed to compare and evaluate oat genotypes to screen large numbers of lines, quantification of resistance components under controlled conditions allowing year-long screening and the rejection of susceptible lines before lengthy field trials. There is a need to determine which resistance components are associated with leaf spot intensity in the field, as estimated from the area under the disease progress curve (AUDPC). We assessed various oat P. chaetomioides resistance components under controlled conditions in seedlings of 26 oat genotypes chosen from recommended varieties and elite breeding lines to determine the association of resistance components with the AUDPC, obtained by evaluating each genotype in the field. The resistance components estimated were: initial lesion size (ILS) and final lesion size (FLS); rate of lesion expansion (r); and area under the normalized and corrected lesion expansion curve (AULECc). All correlations were positive and significant at p?=?0.01 and were distributed into moderate (0.5?<?r?<?0.8) and strong (0.8????r?<?1) correlation classes. The strongest average correlations occurred with the AULECc (0.827), ILS (0.801) and FLS (0.801) components. These results indicate which components may be useful in resistance screening, with FLS possibly being the most useful criterion because it is less laborious to obtain and speeds up the selection process for leaf spot resistance.  相似文献

Growth and feed utilization of juvenile, hatchery‐reared mutton snapper Lutjanus analis (mean weight = 12.2 g) were compared for 40 d in laboratory aquaria on four isonitrogenous diets (45% crude protein) of varying lipid content (6, 9, 12 and 15%) with energy:protein ratios (E: P; kJ/g protein) of 33.9, 36.3, 38.8, and 41.2, respectively. Growth on these diets was compared under temperatures of 25 and 30 C. Final weights (Wtr) and specific growth rates (SGR) were higher (P < 0.05) at lower dietary E: P ratios of 33.9 and 36.3 (Wtr= 20.3–22.0 g; SGR = 1.25–1.35%/d) than at E:P ratios of 38.8 and 41.2 (Wtr= 17.2‐17.7 g; SGR = 0.84–0.85%/d). Growth at 30 C (Wtr= 21.5 g, SGR = 1.35%/d) was higher (P < 0.05) than at 25 C (Wtr= 17.3 g; SGR = 0.82%/d). Feed consumption (FC) was higher (P < 0.05) at a dietary E: P ratio of 33.9 (1.57%/d) than at 36.3 (1.27%/d) or 38.8–41.2 (0.89–0.98%/d). Growth was highly correlated (P < 0.01) to E: P ratio and to feed consumption. Feed conversion ratio (FCR = 2.17–3.98), protein efficiency ratio (PER = 0.58–1.03) and apparent net protein retention (ANPR = 15.8–20.0%) were not significantly (P > 0.05) affected by dietary E: P ratio. Apparent net energy retention (ANER) was higher (P < 0.05) at E: P ratios of 33.9 and 36.3 (9.50–9.98%) than at E: P ratios of 38.8 and 41.2 (7.15–7.10%). Feed utilization parameters were significantly (P < 0.05) better at 30 C (FC = 1.36%/d; FCR = 2.6; PER = 0.88; ANER = 10.2%) than at 25 C (FC = 1.03%/d; FCR = 3.38; PER = 0.69; ANER = 6.72%), with the exception of ANPR (17.0–17.8%). Maximum growth and energy retention in juvenile mutton snapper using a diet containing 45% crude protein was obtained at dietary lipid levels of 6–9% and E: P ratios of 33.9–36.3 kJ/g protein. Studies which determine optimum protein levels and the effects of reducing E: P ratios below 33.9 kJ/g are needed to improve feed conversion and growth.  相似文献

Dietary supplements such as grape seed extract (GSE) enriched in proanthocyanidins (PA) (oligomeric polyphenols) have been suggested to have multiple health benefits, due to antioxidant and other beneficial activities of the PA. However, a systematic analysis of the molecular basis of these benefits has not been demonstrated. Because the brain is vulnerable to age-related oxidative damage and other insults including inflammation, it was hypothesized that rats ingesting GSE would experience changes in expression or modifications of specific brain proteins that might protect against pathologic events. Normal adult female rats were fed diets supplemented with 5% GSE for 6 weeks. Proteomics analysis (2D electrophoresis and mass spectrometry) of brain homogenates from these animals identified 13 proteins that were altered in amount and/or charge. Because many of these changes were quantitatively in the opposite direction from previous findings for the same proteins in either Alzheimer disease or mouse models of neurodegeneration, the data suggest that these identified proteins may mediate the neuroprotective actions of GSE. This is the first identification and quantitation of specific proteins in mammalian tissues modulated by a dietary supplement, as well as the first to demonstrate links of such proteins with any disease.  相似文献
