The present work aimed to investigate the presence of T. vivax DNA in the semen of experimentally infected goats. Twelve male goats native to the Brazilian Northeast, adults, were randomly assigned to two experimental groups: the infected group consisting of six goats infected intravenously with 0.5 mL of blood containing approximately 1.25?×?105 trypomastigotes of T. vivax, and a control group composed of six uninfected goats. After the infection, clinical examinations aiming to evaluate rectal temperature, parasitemia and hematocrit were performed. Semen samples were collected from goats by electroejaculation on the 7th, 14th and 21st days post-infection (dpi). The recombinant DNA-encoding gene encoding the L-like-specific gene for T. vivax. The infection was characterized by increased rectal temperature, high parasitemia and significant reduction of hematocrit values. Results for T. vivax DNA detection using TviCatL-PCR were positive in all semen samples from the infected group collected on 7th, 14th and 21st dpi. The presence of T. vivax DNA in 7th dpi suggests the early invasion of the parasite in the reproductive organs. Also, the finding of T. vivax DNA in all periods analyzed may suggest the continued elimination of the parasite in the semen, which may increase the chances of sexual transmission. Thus, T. vivax DNA is recorded for the first time in the semen of infected goats. Thus, these data are of great importance, since the detection of the T. vivax genetic material in the semen may point to the possibility that the parasite may be transmitted through the sexual pathway. 相似文献

Haemonchus contortus is responsible for severe economic losses in sheep and goat breeding in the Northeast of Brazil. However, the effectiveness of control is compromised due to anthelmintic resistance and misuse. In the search for natural anthelmintics, Melia azedarach L., a plant indigenous to India but now distributed throughout Brazil, was selected due to the reported anthelmintic properties of its seeds. The aim of this study was to evaluate the anthelmintic activity of the seed and leaf extracts of the Brazilian adapted plant and investigate the type of organic chemical compounds present in the most active extracts. The ovicidal and larvicidal activity of M. azedarach extracts on H. contortus was evaluated through egg hatching and larval development tests. Hexane and ethanol extracts of seeds and chloroform and ethanol extracts of leaves of M. azedarach were used in the tests. To perform the larval development test, feces of an animal free from parasites were mixed with third instar H. contortus larvae and extracts in several concentrations. The coprocultures were incubated for 7 days at 30 degrees C, then the larvae were recovered and counted. LC50 was calculated by probits using the SPSS 8.0 program. The seed ethanol extract was the most active on eggs (LC50=0.36mgmL(-1)) and the leaf ethanol extract showed the best inhibition of larval development (LC50=9.18mgmL(-1)). Canine ehrlichiosis and babesiosis have a worldwide distribution with geographic variation in prevalence and main clinical manifestations. We prospectively determined seroprevalence of canine babesiosis and ehrlichiosis, and risk factors for seropositivity. Three hundred and eighty-one dogs were randomly selected to represent the canine population at a Veterinary Teaching Hospital in south Brazil (latitude 23° S). Dogs were tested with a point-of-care ELISA for Ehrlichia canis antibodies and IFA to confirm previous exposure to Babesia vogeli. Multiple logistic regression analysis was then used to estimate adjusted odds ratio (OR) and their 95% confidence intervals. One hundred and thirty-six (36%) dogs were seropositive for B. vogeli antibodies, whereas 87 (23%) dogs were seropositive to E. canis antibodies. Fifty-four (14%) dogs seroreacted to both agents. Adult dogs previously infested with ticks were more likely to seroreact to B. vogeli or E. canis. Superficial bleeding (OR = 12.4) was more common in dogs exposed to B. vogeli, whereas neurological signs (OR = 7.7) were more common in dogs seropositive to E. canis. Neurological signs (OR = 12.0) and lameness (OR = 12.8) were more prevalent in dogs that seroreacted to both organisms. Owners of dogs with ticks were more likely to have been exposed to ticks themselves (OR = 3.2). Canine babesiosis and ehrlichiosis appear to be highly prevalent in this hospital population. Clinical signs differed from the most common signs in other regions with bleeding occurring more in dogs seropositive to babesiosis, but not ehrlichiosis; neurologic signs in dogs with E. canis antibodies; and lameness in dogs that seroreacted to both organisms. 相似文献

Gastrointestinal nematode control has been performed through use of anthelmintics. However, the development of resistant populations has required research into new alternatives. There are popular reports about anti-parasitic activity of Azadirachta indica in animals and plants. The aim of this study was to evaluate the anthelmintic activity of A. indica after feeding sheep with the dried leaves. In this experiment, 40 sheep were allotted into four treatment groups. Group I received a treatment of A. indica dry leaves mixed in a concentrate at a rate of 0.1 g/kg dose for 3 months. Group II was treated with double the dose of Group I. Group III was treated with closantel (Diantel) at the manufacturer-recommended dose once at the beginning of the study and Group IV was not treated. To compare treatment effects, the following parameters were evaluated: egg count per gram of feces (EPG), worm burden, weight gain and haematocrit. EPG and worm burden results were statistically evaluated using the Kruskal-Wallis test. Haematocrit and live weight gain were submitted to analysis of variance (ANOVA) and the means evaluated by Tukey's test with 95% probability. None of the evaluated parameters of the treatment groups were statistically different when compared to the control group, demonstrating that, with the protocol used, A. indica has no anthelmintic effect. 相似文献
