AIM: To develop a technique to estimate the volume of epithelial lining fluid (ELF) obtained during bronchoalveolar lavage (BAL) and pleural lavage (PL) in the dog, using the urea dilution method. METHODS: BAL and PL fluids were obtained by saline lavage of pulmonary and pleural cavities of nine clinically healthy mixed-breed dogs immediately after euthanasia. Cell counts in the BAL and PL fluids were measured using standard techniques. The concentration of ELF in each lavage fluid was calculated from the relative concentration of urea in plasma and in each type of lavage fluid. Cell counts in ELF were then calculated. RESULTS: There were substantially higher cell counts in ELF compared to BAL or PF fluid. However, nucleated cell counts in ELF could not be predicted from cell counts in BAL or PL fluid. CONCLUSIONS AND CLINICAL RELEVANCE: These results suggest that accurate assessment of cellular or non-cellular components in lavage fluids should include a calculation of the proportion of ELF recovered, using a method such as urea dilution. 相似文献

This study investigated whether enhanced metabolic responsiveness to beta-adrenergic stimulation might explain the liability of the stress-sensitive pig to develop an acidosis during stressful situations. Blood lactate, pH, PCO2 and heart rate response to an intravenous infusion of isoprenaline were examined in three lines of pentobarbitone anaesthetized, atropinized pigs. The lines were Pietrain X Hampshire which had been selected over eight generations for a positive reaction to the halothane test (stress-sensitive); Pietrain X Hampshire which had been selected over eight generations for no reaction to the halothane test (stress-resistant) and British Landrace which had been selected over four generations for a positive halothane reaction (stress-sensitive). The metabolic responses to isoprenaline were greater in the stress-sensitive Pietrain X Hampshire line than the stress-resistant line. The stress-sensitive Landrace pigs, however, had smaller changes in pH and PCO2 in response to isoprenaline than the stress-resistant Pietrain X Hampshire pigs. This inconsistent trend across the three lines led to the conclusion that enhanced beta-adrenergic responsiveness was not the likely cause of stress sensitivity. 相似文献

Two experiments were carried out to examine the effects of grazing severity on the performance of January/February calved British Friesian dairy cows. In Experiment 1, three groups of cows were rotationally grazed across twenty-four one-day paddocks with high (H), medium (M) or low (L) herbage allowances. Cows on treatment M were offered a daily herbage allowance designed to achieve a residual sward height of 50 mm, assessed by a rising-plate sward stick. The daily herbage allowance (g organic matter (kg live-weight)-1 on treatments L and H were 0.30 below and above that for M, respectively, to give residual sward heights of 42 and 59 mm. In Experiment 2, three groups of cows were grazed across twenty-four one-day paddocks to obtain residual sward heights of 50 mm (severe), 60 mm (moderate) and 80 mm (lax). Average milk yields on the L, M and H treatments in Experiment 1 were 11.8, 14.6 and 14.5 kg d,-1 and in Experiment 2 they were 13.7, 16.0 and 17.0 kg d-1 on the severe, moderate and lax treatments, respectively. The results indicate that the critical herbage height below which milk production per cow declines may vary with the production potential of the animal. There were no significant treatment effects on milk composition. Milk output ha-1 and utilized metabolizable energy ha-1 were greatest with the low herbage allowance in Experiment I and the moderate treatment in Experiment 2. Net herbage accumulation on the severe treatment in Experiment 2 was 30% lower than that on the lax treatment, as a result of treading damage in early season. It is concluded that, in a rotational grazing system, a reasonable compromise between sward utilization and animal performance can be achieved by grazing January/February calved cows to a residual sward height of 60 mm as assessed by a rising-plate sward stick. Distributions of the vector Culicoides brevitarsis Kieffer (Diptera: Ceratopogonidae) (determined from light trap data) and 2 arboviruses (determined from seroconversions in sentinel cattle) were studied in eastern New South Wales in 1993–94. C brevitarsis was recorded progressively from endemic areas on the north coast, to Nowra on the south coast, and westward to Scone, in the Hunter Valley. C brevitarsis also survived through winter at Paterson, in the Hunter Valley. Its apparently focal reappearance in this marginal area had no obvious effect on the broad pattern of its progression or the dispersal of Akabane and bluetongue viruses. These viruses were first recorded from foci near Coffs Harbour, on the mid-north coast. Their first occurrences at different locations were associated with those of C brevitarsis, but not with each other. The viruses were found only within the recorded limits of the vector's distribution. Delays between the initial occurrence of C brevitarsis and first evidence of virus transmissions at locations ranged from 2 to 7 months. The delays decreased away from the points of focus and were negatively associated with the time of initial occurrence of the vector. Seroconversions to the viruses were related to the presence of C brevitarsis. However, the densities of C brevitarsis had no apparent effect on the initial numbers of cattle seroconverting to either virus. The results support the conclusion that the progressions of C brevitarsis and Akabane and bluetongue viruses were the result of gradual movements by the vector. 相似文献

Fossils discovered in Lower Cretaceous (Aptian) rocks in the Tenere Desert of central Niger provide new information about spinosaurids, a peculiar group of piscivorous theropod dinosaurs. The remains, which represent a new genus and species, reveal the extreme elongation and transverse compression of the spinosaurid snout. The postcranial bones include blade-shaped vertebral spines that form a low sail over the hips. Phylogenetic analysis suggests that the enlarged thumb claw and robust forelimb evolved during the Jurassic, before the elongated snout and other fish-eating adaptations in the skull. The close phylogenetic relationship between the new African spinosaurid and Baryonyx from Europe provides evidence of dispersal across the Tethys seaway during the Early Cretaceous. 相似文献

An isotope dilution method using [1‐13C]sodium acetate was applied to determine the effect of feeding ensiled hop (Humulus lupulus L.) residues on plasma acetate turnover rate in six adult crossbred sheep. The sheep were fed 63 g/kg body weight (BW)0.75/day of either mixed hay of orchardgrass (Dactylis glomerata L.) and reed canarygrass (Phalaris arundinacea L.) and round bale silage at 3:1 ratio (Hay‐diet), or another where round bale silage was replaced by ensiled hop residues (Hop‐diet) with a crossover design each of a 3‐week period. The isotope dilution method was performed on day 21 of each dietary treatment. Dry matter digestibility was similar between diets, and nitrogen (N) digestibility was lower (P = 0.001) for Hop‐diet than Hay‐diet. However, N retention did not differ between diets. Plasma acetate concentration was lower (P = 0.04) for Hop‐diet than Hay‐diet, and the turnover rate of plasma acetate did not differ between diets. Plasma concentration of lactate and non‐esterified fatty acids were similar between diets. Hop‐diet was found almost comparable to Hay‐diet on plasma acetate turnover rate in the present experimental conditions. The effect of glyphosate on leaves and basal bulbs of yellow nutsedge (Cyperus esculentus L.) plants was studied. Treatment with glyphosate did not affect leaf anatomy, although light necrotic spots were seen on the veins. In contrast, basal bulb organization was greatly affected with large necrotic zones and disorganization in the vascular cylinder due to herbicide accumulation. Depletion or disappearance of vacuolar phenols was observed in bulbs of plants treated with the highest doses of glyphosate. The most remarkable effect of glyphosate was the appearance of one layer of sclerenchymatic cells between root and rhizome primordia and the cortical tissues which could play an important role in the inhibition of rhizome and root emergence by the herbicide. 相似文献
