An enzyme-linked immunosorbent assay (ELISA) was evaluated for detection of antibodies (Ab) against Mycoplasma hyopneumoniae and M. flocculare in sera from swine experimentally infected with these agents. In addition, the ELISA was compared with the complement fixation test (CFT), and radial immunodiffusion enzyme assay (RIDEA) for the demonstration of Ab against M. hyopneumoniae. Twenty two 6-week-old swine from a respiratory disease-free herd were divided into five groups. Two or three pigs from each of the four groups were inoculated, respectively, with M. hyopneumoniae or with M. flocculare while two pigs in each group were contact exposed to the inoculated penmates. A fifth group, consisting of three pigs, served as inoculated controls. Pigs inoculated with M. hyopneumoniae began coughing 13 days post inoculation (PI). Antibodies were first detected 2 weeks PI with the CFT, 3 weeks PI with the ELISA, and 5 weeks PI with the RIDEA. With the ELISA and RIDEA, Ab were still detectable one year PI at a very low level. With the CFT, Ab were not detectable in sera from any swine beyond 5 months PI. At necropsy 1 year PI, no lesions were detected in lungs of any of the animals nor were mycoplasmas detected. M. flocculare inoculated or contact-exposed pigs never evidenced clinical signs. Antibodies against M. flocculare were first detected 5 to 12 weeks PI with CFT, and 6 to 12 weeks PI with the ELISA. Peak optical density (OD) values obtained in the ELISA with M. flocculare Ab were as high as the values obtained with peak M. hyopneumoniae Ab titers. Levels of Ab against M. flocculare were at relatively higher OD at 1 year PI than Ab against M. hyopneumoniae. Sera with high levels of Ab against M. flocculare cross-reacted slightly with M. hyopneumoniae antigen in immunoblotting and ELISA.  相似文献

The pin-hole collimator is used to improve spatial resolution and magnify areas of interest. The pin-hole collimator has many applications in small animal veterinary scintigraphy. The principles of image formation for the pin-hole and parallel hole collimators are reviewed. The effects of distance on resolution and sensitivity are presented for the pin-hole and parallel hole collimators. Specific application of the pin-hole and parallel hole collimators. Boran (Bos indicus) heifers were immunised by infection with local Theileria parva parva, T p lawrencei and T mutans stocks and treated with parvaquone and later exposed to natural tick and tick-borne disease challenge in the Trans-Mara Division of Kenya. The Theileria species parasites in the challenge were maintained in African buffalo and cattle and the tick vectors were supported by several species of wild Bovidae and domestic livestock present in the area. Thirty immune cattle were observed for 30 weeks while grazing on a ranch in the Trans-Mara Division. Of these, 15 were immersed in toxaphene at weekly or twice-weekly intervals while 15 cattle remained without acaricide application. Cattle which became pregnant were withdrawn from the experiment. There was no evidence of any clinical tick-borne disease in either group of cattle during the experiment. Five species of ixodid ticks infested the cattle during the experiment and cattle which were not treated with acaricide had far more ticks. Rhipicephalus appendiculatus was the most abundant tick species, with a mean infestation of 46 adults per animal in the undipped cattle. This tick also appeared to be the cause of the observed reduction in weight gains. Major haematological parameters did not differ significantly between the groups. Behavioural studies showed that the undipped cattle spent less time grazing and ruminating. This study has shown that, at the expense of some loss in productivity, zebu cattle, immunised against ticks and theileriosis, can be kept despite tick infestation.  相似文献

Summary A series of international trials with the potato crop was conducted in eight countries representing widely differing environments. The purpose of the experiments was: (1) to develop a method for obtaining environmental indexes which measure productivity of diverse potato-growing areas, and (2) to construct performance models for individual genotypes based on the environmental indexes. Yield data of 32 genotypes obtained from a series of 13 experiments conducted in the eight countries in 1983 and 1984 were used in the study. The response pattern of the genotypes was studied by a multivariate analysis. Five genotypes were chosen as representatives for different response patterns to the environments and their data were used for calculating a series of independent indexes for each of the experimental sites based on principal component analysis. Performance models for each of the genotypes were constructed by stepwise regression analysis of yields of a genotype on the environmental indexes based on 1983 data. Yield data of 1984 experiments were used to verify the models. Satisfactory correspondence between observed and predicted yields was obtained for most of the genotypes.  相似文献

A genetic map was constructed using DNA‐based markers in a barley mapping population derived from the cross ‘Tankard’בLivet’, that was developed to explore the genetic control over grain damage in spring barley cultivars. Quantitative trait loci (QTL) were located for husk skinning, gape between the lemma and palea and splitting of the fused pericarp/testa/aleurone tissues. The QTL accounted for 70% of the genetic variation in Split and 60% of the genetic variation in Gape and Skinning. The QTL were clustered on chromosomes 1H, 4H, 5H, 6H and 7H. QTL analysis indicates the possibility of transgressive segregation for grain splitting and so the breeding of lines with more extreme splitting. This is of concern to the malting industry as, without extensive phenotypic assessment, such lines could be commercialized, as was the case of Landlord, and put malting barley supplies at risk. These findings are discussed in relation to the genetic control over traits including grain length and width.  相似文献

Summary Effect of sterilizing (WA) cytoplasm on heterosis and combining ability for days to flowering, plant height and grain yield in rice was studied in 70 crosses and their reciprocals produced by 10 cytoplasmic male sterile (A), their maintainer (B) and seven restorer (R) lines following line x tester design. The materials consisting of 140 hybrids (70 A/R and their reciprocal 70 R/B) plus 17 parental lines (10 B + 7 R) were evaluated under six environments, created by growing in three fields with different fertilizer doses (0, 60, 120 kg N/ha) and 2 seasons (dry and wet) during 1986 at IRRI farm. Reciprocal cross effect (A/R vs R/B) were highly significant for all the three traits. Interaction of reciprocal cross effects with environments were also highly significant for yield and days to flowering. Cytoplasmic effect for yield, days to flowering and plant height were estimated by comparing A/R and R/B combination and testing the significance of difference with LSD value. In order to avoid confounding effect of spikelet sterility on yields, twenty crosses showing normal spikelet fertility were selected. Both positive and negative cytoplasmic effects were observed for the three traits. The effects were modified by environments, except for plant height. Heterosis for all three traits was also affected by cytoplasm, however, manifestation of cytoplasmic effects was higher for heterosis for days to flower than in heterosis for yield and plant height. Effect of cytoplasm was more pronounced on general combining ability effects of parents than specific combining ability effects of crosses. Among the parents two CMS lines A4 (IR54752A) and A8 (IR22107-113-3-3A) and two R lines: R2 (IR46) and R7 (IR9761-19-1) showed consistent positive effect of cytoplasm on general combining ability. These lines have given several good heterotic combinations. The study indicated the usefulness of evaluating diverse cytoplasmic sources in various nuclear genotypes bred for hybrid rice breeding program.  相似文献

Summary Four selection strategies were used on four sets of oat lines to select for grain yield. Two of these used uniform environments whereby sequential selection of the high-yielding lines occurred in continuous high- or continuous low-productivity environments. These are referred to as high and low uniform selection strategies, respectively. The other two selection strategies were conducted by sequential selection of the high-yielding lines in alternating high- and low-productivity environments. They are referred to as high and low shuttle selection strategies, respectively, with high and low designating the productivity of the first environment in the sequence. After three or four cycles of selection, the surviving lines and a random sample from each set were evaluated for mean grain yield, grain yield response to improving environments, and stability of grain yield, in a range of environments typical of oat production on Iowa farms.Grain yield and regression response for all selection strategies, when calculated across all sets of lines, were significantly greater than corresponding values for random samples. Stability was unchanged. The uniform-high and uniform-low strategies gave the greatest and the smallest gains in mean grain yield, respectively, with the shuttle strategies giving intermediate gains. Shuttle selection in predominantly high-productivity environments increased grain yield more than shuttle selection in predominantly low-productivity environments. The uniform-strategy followed by the shuttle-high strategy identified entries with superior performance in high productivity environments. The effect of protein and peptide addition on the oxidation of eicosapentaenoic acid ethyl ester (EPE) encapsulated by maltodextrin (MD) was investigated. The encapsulated lipid (powder lipid) was prepared in two steps, i.e., mixing of EPE with MD solutions (+/- protein and peptides) to produce emulsions and freeze-drying of the resultant emulsions. EPE oxidation in MD powder progressed more rapidly in the humid state [relative humidity (RH) = 70%] than in the dry state (RH = 10%). The addition of soy protein, soy peptide, and gelatin peptides improved the oxidation stability of EPE encapsulated by MD, and the inhibition of lipid oxidation by the protein and the peptides was more dramatic in the humid state. Especially, the oxidation of EPE was almost perfectly suppressed when the lipid was encapsulated with MD + soy peptide during storage in the humid state for 7 days. Several physical properties such as the lipid particle size of the emulsions, the fraction of nonencapsulated lipids, scanning electron microscopy images of powder lipids, and the mobility of the MD matrix were investigated to find the modification of encapsulation behavior by the addition of the protein and peptides, but no significant change was observed. On the other hand, the protein and peptides exhibited a strong radical scavenging activity in the powder systems as well as in the solution systems. These results suggest that a chemical mechanism such as radical scavenging ability plays an important role in the suppression of EPE oxidation in MD powder by soy proteins, soy peptides, and gelatin peptides.  相似文献

Plants tolerant to polluted environments were selected, based on several criteria, to restore a coastal forest ecosystem severely damaged by air pollutants discharged from an industrial complex. In addition, a restoration plan was prepared synthesizing these results and the diagnostic ecological indicators in the area for which restoration is required. Pollution-tolerant plants of 11 tree and subtree species, 10 herb species and one shrub species were selected from field surveys in the vicinity of two representative industrial complexes in Korea, Ulsan and Yeocheon. Nine species were selected for tolerance to SO2 fumigation and six species were selected for tolerance to Al3+. Growth and photosynthetic responses of sample plants transplanted into polluted and unpolluted sites showed that 15 species out of the 26 sample plants showed a disposition for tolerance. Most of these are endemic plants and they are composed of diverse species in structure and function. This result implies that these tolerant species could play important roles in the restoration of the study area, which has several specific features. On the other hand, results from transplant tests indicate that a field survey is the most reasonable method for selection of tolerant plants to restore a pollution-damaged ecosystem, as was shown in another restoration program. Results of ecological analysis on vegetation map indicate that the spatial range within the first ridge is the sector for which restoration is required. This sector was classified into four zones on the basis of topographic conditions: lower and upper slopes of both slopes facing and opposite the pollution source. Guidelines for soil amelioration and arrangement of tolerant plants were prepared considering the degree of vegetation degradation, leaf damage of major plant species and soil pollution in each zone under the restoration plan.  相似文献
