寒地多年生小麦的选育与细胞遗传学分析

通过杂交方法获得八倍体小偃麦与中间偃麦草杂种后代,对该杂交后代进行了形态学观察和细胞遗传学分析。杂交当代结实率为10%~39%;F₁表现为两亲中间型,多年生,抗小麦多种病害,生长的第2和第3年结少量种子,结实率为2%~3%;F₂分离复杂,出现八倍体小偃麦类型和中间偃麦草类型的多年生材料;F₃和F₄代出现一些普通小麦类型的多年生小麦,表现多分蘖、多小穗、抗病、抗寒。F₁根尖减数分裂中发现49条染色体,在减数分裂中期I形成14~17个二价体和4~21个单价体;而F₂和F₃代减数分裂时形成14~21个二价体和9~17个单价体。杂种后代结实率逐代恢复。F₁植株已在田间自然条件下生长5年。从F₄代中获得了4个植株高大(140 cm)、分蘖丰富(60个以上)、小穗多(25~30个)的饲草型多年生小麦株系,它们不仅具有良好的刈割再生能力,而且兼抗多种病害,抗寒性好,草质与中间偃麦草相似。还获得了一些普通小麦类型的多年生株系,有待进一步改良。这些结果为多年生小麦的遗传研究和利用提供了信息和材料基础。     

BMPR-IB、BMP15、GDF9基因作为闽东山羊多胎性能候选基因的研究

以控制部分绵、山羊品种高繁殖力的BMPR-IB、BMP15和GDF9基因为候选基因,采用PCR-RFLP方法分析闽东山羊BMPR-IB,BMP15和GDF9基因多态性与繁殖性状的关系。研究发现：多胎品种闽东山羊及南江黄羊在BMPR-IB基因的相应位置上并未发生与Booroola Merino羊相同的突变,同时也未检测到BMP15的FecXI,FecXH,FecXB基因及GDF9的FecGH基因,因此排除了这5个突变位点影响闽东山羊高繁殖力性状的可能性。然而,由于闽东山羊的BMPR-IB、BMP15、GDF9的全基因序列信息的缺乏,尚不能完全断定3个基因对闽东山羊高繁殖力性状没有影响。     

PCR-DGGE技术在智能化沼气池微生物多样性研究中的应用

采用变性梯度凝胶电泳(PCR-DGGE)技术研究智能化上流式沼气池微生物群落结构的动态变化。结果表明:沼气池中微生物种类丰富,并且拥有优势种群,主要有:稳杆菌属、热单细胞属、杜檊氏菌属、戈登氏菌属、芽孢杆菌属、拟杆菌门、变形菌门的α类群和厌氧细菌门。各细菌种类不同发酵层、不同发酵时间对微生物多样性存在影响。DGGE图谱中的优势条带16SrDNA基因序列经比对后都属于不可培养细菌。说明利用PCR-DGGE技术研究智能化沼气池中微生物多样性是可行的。     

小花棘豆总黄酮对小鼠游泳运动能力的影响

用不同剂量(10、30、50mg/kg)的小花棘豆总黄酮给小鼠灌胃,观察其对小鼠游泳能力的影响,待小鼠力竭后测定其13项血液生理指标,血液和器官组织的相关酶活性以及中间代谢产物的含量。结果表明,添加小花棘豆总黄酮可延长小鼠游泳至力竭的时间,降低机体白细胞总数和粒细胞百分数,提高血液中淋巴细胞百分数、中值细胞百分数、粒细胞数、血红蛋白浓度、红细胞压积等指标,有效提高了小鼠血清和组织中超氧化物歧化酶、过氧化氢酶和谷胱甘肽过氧化物酶的活性,降低了血清谷草转氨酶和肌酸激酶的活性;降低了脂褐素、丙二醛、乳酸和尿素氮的含量,增加了肌糖原和肝糖原的含量。说明小花棘豆总黄酮具有较好的抗氧化损伤功效。

Expression of Duck Interferon Alpha in BL21（DE3）plysS

To obtain the protein of duck interferon alpha and study its biological activities, the prokaryotic ex-pression vector of DuIFN-α was constructed and expressed in BL21 (DE3) plysS. Using PCR technique, the proteingene of DuIFN-α was cloned from pMD-18-duIFN-α recombinant. The gene was then inserted to pGEM-T vectorand identified by restriction endonuclease analysis and sequencing. DuIFN-α was ligated with the prokaryotic expres-sion vector of pET30 a, then transformed into BL21 (DE3) plysS. The best inducing time and IPTG concentration for the expression of this recombinant protein was tested through the expression of the positive recombinant with differ-ent time span and different IPTG concentration. Lots of the protein of DuIFN-α were expressed in BL21 (DE3)plysS with 1 mmol·L-1 IPTG for 4 hours and its molecular weight for 34 000.     

Expression of a Carrot Antifreeze Protein Gene in Escherichia coli

The recombinant expression vectorpET43. lb-AFP, which contains full encoding region of a carrot 36 kD antifreeze protein (AFP) gene was constructed. The recombinant was transformed into expression host carrying T7 RNA polymerase gene (DE3 lysogen) and induced by 1 mmol. L-1 IPTG (isopropyl-β-D-thiogalactoside) to express 110 kD polypeptide of AFP fusion protein.The analysis of product solubility revealed that pET43. 1b-AFP was predominately soluble, and the expressed amount reached the maximum after the IPTG treatment for 3 h.
