The aim of this trial was to assess the effect that calcium gluconate priming of 468 broilers has on the antibacterial activity of a standard dose of enrofloxacin. Hence, a series of oral pharmacokinetic studies were carried out in four groups of broilers medicated individually through an oral cannula as follows: group A, medicated only with enrofloxacin 10mg/kg; group B, receiving immediately one after the other, calcium gluconate (200mg/kg) and enrofloxacin 10mg/kg; group C, dosed first with calcium gluconate (200mg/kg) and 1h later enrofloxacin (10mg/kg); and group D, dosed first with calcium gluconate (200mg/kg) and 2h later enrofloxacin (10mg/kg). Broilers were bled at different times after the dose of enrofloxacin and antibacterial activity, measured as concentration of enrofloxacin, was measured by an agar diffusion assay. Results revealed that group D the greatest values of maximum serum concentration (Cs(max)), area under the concentration vs. time curve (AUC) and area under the moment curve (AUMC). These values were statistically higher than the corresponding ones derived from groups A, B and C (P<0.05). Taking Cs(max) and AUC values of group A as reference baseline, an increase of 24% and 50%, respectively, was obtained in group D. Group B had the lowest Cs(max), AUC, AUMC and elimination half life (T(1/2)beta) and these values were statistically different from groups A, C and D (P<0.05). The T(1/2)beta was statistically longer in groups C and D as compared with A and B, and the former groups were also different between each other (P<0.05). These results show that if calcium gluconate is first dosed to broilers and 2h later enrofloxacin is administered (as in group D), a more pronounced antibacterial activity of enrofloxacin can be obtained. A challenge of this sequential dosing scheme in a field trial may reveal its clinical value. 相似文献

Calcium uptake by bell pepper (Capsicum annuum L. cv. ‘California Wonder') varied by stage of plant development and N form supplied (NO3 ‐NH4 + ratios: 1:0, 3:1, 1:1, 1:3, and 0:1) in a hydroponic study. Uptake of Ca++ was highest at bloom and during fruit expansion, making the fruit development stage the highest demand period. Calcium uptake declined with each increasing increment of NH4 + relative to NO3 ‐ supplied, although fruit yield was not significantly reduced until the ratio of NH4 + to NO3 ‐ exceeded 50%. Tissue Ca++ levels in the blossom‐end of the fruit were reduced whenever NH4 + was included with N supplied. Vegetative yield of plants followed the same trend as that observed for total fruit dry weights. Our results indicate that pepper yields are higher when NO3 ‐ is the predominant form of N. Also, these results strongly suggest that Ca++ fertilizer applications should precede the bloom period and continue during fruit development to ensure adequate Ca++ availability for fruit development. 相似文献

A methodology for ranking and classifying performance of irrigation systems through multidimensional performance indicators is developed using fuzzy set theory. The procedure uses the concepts of fuzzy resemblance and fuzzy dominance. Preference levels reflecting management priorities are incorporated into the analysis using appropriate weighting factors. The application of the procedure is demonstrated through two case studies: the Shi-Jin irrigation district in Hebei Province, China and the Goulburn irrigation region in Victoria, Australia. The classification of performance periods for the Australian system shows three clusters indicating the predominant effect of the increase in waterlogged area when higher priority is given to this indicator. No clear trend appeared when equal weight was assigned to all indicators. The effect of economic reforms implemented in 1977–78 in China is clearly reflected in the resulting ranking and clustering of the performance periods in the Shi-Jin irrigation district. Including Baytril, in various parts of the world many commercial preparations of enrofloxacin for parenteral administration are being employed for the treatment of bacterial diseases in cows. To optimize clinical responses and to minimize development of bacterial resistance to this agent, the copied pharmaceutical preparations must comply with some key pharmacokinetic features when bioequivalence studies are performed. To assess whether or not there was bioequivalence among nine commercial preparations of enrofloxacin and the original one, a controlled pharmacokinetic study was carried out. These was done utilizing the microbiological agar-diffusion test as quantitative/qualitative analytical method. A non-compartmental model defined kinetic variables. Results for Baytril revealed that maximal serum concentration (Csmax) was only matched by one preparation while area under the curve (AUC) of the serum concentration/activity of enrofloxacin and metabolites in time was not matched by any preparation. Time to Csmax (Tmax), elimination half-life, and shape of the time-serum concentrations of enrofloxacin profiles obtained for the nine generic preparations differ significantly somehow from the corresponding data obtained for the reference enrofloxacin. The need for studies to demonstrate bioequivalence becomes mandatory if similar preparations of enrofloxacin become commercially available. Enrofloxacin should be used selectively and cautiously to limit development of bacterial resistance. Non-bioequivalence of relevant pharmacokinetic values, such as Csmax and bioavailability (AUC) would facilitate development of bacterial resistance and limit the useful life span of this antibacterial agent. 相似文献

The confirmatory diagnosis of Mycobacterium bovis (M. bovis) in animal samples is carried out by culture in Stonebrink media. However, culture is very slow because of the extremely long duplication time of the bacillus and difficult because of the scarcity of bacilli in diagnostic samples. This study describes the development of a single-tube touch-down polymerase chain reaction (PCR) protocol for the detection of M. bovis using primers that target the IS6110 element. Spiked water and milk as well as routine diagnostic samples (milk and nasal swabs) from M. bovis-positive cattle were tested. This protocol allows the rapid and sensitive detection of M. bovis in bovine samples by enhancing the sensitivity of standard PCR amplification. 相似文献

Immunity to Taenia solium infection was investigated using an experimental intramuscular oncosphere infection assay (IMOA) model in pigs. Three naturally infected pigs with cysticercosis were treated with oxfendazole (OFZ), a drug demonstrated to kill cysts in porcine muscle. These animals were then challenged with oncospheres but did not develop any cysts while three uninfected pigs that were similarly challenged, did develop intramuscular cysts. In another study, two groups of three pigs each were immunized with crude T. solium oncosphere and metacestode antigens, respectively, and tested with the IMOA. Immunization with crude oncosphere antigens (OAs) induced 100% protection, while metacestode antigens provided only partial protection. Immunoblots showed that pigs with complete immune protection to oncosphere intramuscular challenge had antibodies to two OAs at 31.3 and 22.5 kDa, respectively. Antibody to these two antigens was absent in pigs immunized with metacestodes or in uninfected control pigs. This study demonstrated the presence of two antigens that are unique to the oncosphere. Although, antibody to these two antigens is consistently present in pigs that are protected from an oncosphere intramuscular challenge their role in preventing infection by T. solium larval cysts is still hypothetical. 相似文献

Based on favorable experiences with acupuncture for the treatment of hind limbs paralysis, lumbo-sacral alterations, and other spinal cord problems; a clinical trial, involving 40 dogs affected with wobbler syndrome was carried out. Patients were graded in three categories according to the severity of each case and then randomly divided in two groups. Both groups contained all three grades. Group 1 (20 dogs) was treated using orthodox medical and surgical interventions, while Group 2 (20 dogs) were treated mainly with electroacupuncture, and in few cases with surgical intervention as well. The study was carried out in a three-year period. Acupuncture treatments were given every other day delivering 150 to 300 mVolts at 125 Hz, equivalents to approximately 20 microAmps, in ten acupuncture points per treatment. Deep needle insertion was used. Overall per cent success in Group I was only 20%, while in group II the corresponding value was 85%. The number of acupuncture treatments required to achieve full recovery in Group II was dependent upon the severity of the case, as follows: Grade I: 18.5 +/- 2.5; Grade II: 25 +/- 5.4; and Grade III: 34 +/- 6.7 (r = 0.962). A model to predict the bending modulus of elasticity (MOE) of oriented strand board (OSB) panels produced by batch processing
is presented. The approach developed herein is unique in its comprehensiveness since the MOE is determined from information
on the panel structure, temperature and moisture profiles and vertical density profiles obtained from the mat formation and
compression models presented in Part 1. Comparison of predicted MOE values with those measured from 24 commercially produced
panels shows good agreement considering some of the uncertainties involved. Simulations show that the MOE can be increased
by any of the following changes: reduced fines content, increased panel density, better flake alignment in each of the three
layers within a panel, increased flake length and a larger difference between the density of the face and core layers. The
model was also used in a genetic algorithm to carry out an optimization study of batch OSB manufacturing. This analysis showed
that by combining the appropriate reduction in the amount of flakes used, increase in fines content, improvement in flake
alignment within each of the face and core layers and shortening of the batch time, a significant theoretical profit increase

Based on its ideal PK/PD ratios, doxycycline hyclate (DOX‐h), a time‐dependant antibacterial, is ideally expected to achieve sustained plasma drug concentrations at or slightly above the MIC level for as long as possible between dosing intervals. Pursuing this end, a poloxamer‐based matrix was used to produce a 10% long‐acting injectable preparation (DOX‐h‐LA) and its serum concentrations vs. time profile investigated after its injection to pigs in the pericaudal s.c. by parallel design. Results were compared with the forced oral bolus dose and i.v. pharmacokinetics of DOX‐h. For this study, 12 recently weaned pigs per group were included in this trial, and a dose of 20 mg/kg was injected in all cases. DOX‐h‐LA showed the greatest values for bioavailability (115.38%); maximum serum concentration (Cmax) value was 1.5 ± 0.2 with a time to reach Cmax of 3.41 ± 0.04 h and an elimination rate constant of 70.93 ± 0.87 h. Considering minimum effective serum concentration of 0.5 μg/mL, a dose interval of at least 5 days can be achieved for DOX‐h‐LA, whereas p.o. and i.v. dosing of DOX‐h may only last 11 and 15 h, respectively. Pigs were slaughtered on day 30 after this trial, and no visible remnants of the preparation were detected neither fibrosis was observed after a thorough macroscopic and histopathological analysis. 相似文献
