A procedure, based on XAD-4 adsorption resin, which permits the obtainment of enriched polyphenolic extracts from Sesamum indicum perisperm (coat) has been developed. Chemical analysis of the obtained extracts led to the identification of 16 lignans. Among them, two new lignans, (+)-saminol and (+)-episesaminone-9-O-beta-D-sophoroside, have been isolated. Additionally, the relative stereochemistry of (-)-sesamolactol, previously reported as todolactol A epimer, has been unequivocally defined using X-ray crystallography. The structures of all new compounds were determined by spectroscopic methods, mainly by the concerted application of 1D and 2D NMR techniques (HMQC, HMBC, and NOESY) and mass spectroscopy. 相似文献

The olive (Olea europaea) is one of the most important oleaginous crops of the Mediterranean basin. Increased demand for olive oil creates a need for new olive varieties to help meet the requirements of the global market. However, olive breeding has been handicapped by such varied challenges as a prolonged juvenile period, agrotechnical problems and insufficient genetic knowledge. The use of DNA markers has the potential to overcome these problems and increase the effectiveness of classical breeding programmes. In this study, co‐dominant polymorphic simple sequence repeats (SSRs) were used as markers to analyse the genetic relationships between several local and other ‘non‐native’ olive cultivars. Cluster analysis revealed four major groups among the 15 cultivars examined in this study. Table and oil cultivars were clustered in different groups. However, the clusters did not differentiate between cultivars of different geographical origins. In addition, we used the data gathered to analyse genetic relationships to evaluate the effects of heterosis in agricultural traits. Genetic distances between cultivars were determined based on the SSR genotype data and were used for evaluating the possible effects of heterosis in various F1 populations. Interestingly, phenotypic data of F1 progenies from crosses between different cultivars indicated the potential effects of heterosis as expressed in several traits. Genetic distance between parents was significantly correlated to F1 performance for three traits: percentage of dry fruit weight, oil content and commercial oil production. Thus, crosses between olive cultivars exhibiting relatively extensive genetic distances one from the other are expected to result in better progeny performance in future Olea breeding programmes. Our study linked assessment of biodiversity of commercial olive cultivars with the application of this information in olive breeding programmes for selection of specific parents to generate superior new cultivars. 相似文献

In several laboratory and production species, the establishment of a proteome reference map of a specific tissue has been accomplished. The rabbit is widely used as both a production and experimental animal. A lot of physiology research involving the gastrocnemius muscle of rabbit is described, although no reference proteome map is available. In this work, the first reference map of the rabbit’s gastrocnemius muscle using 2D gel electrophoresis and the identification of proteins through peptide mass fingerprinting (PMF) was established. A total of 45 proteins were localized and identified with three major roles: cell structure and contractile apparatus; metabolic and cell defense proteins. Abstract— Most marine fish larviculture is dependent on the culture and nutritional manipulation of live prey, of which the rotifers Brachionus sp. are one of the most commonly used. The particular reproductive characteristics of gilthead seabream Spus aurata L. lead to an egg overproduction in commercial hatcheries, where the excess is normally wasted. This study was developed with the objective of testing seabream eggs as an enrichment product for Brachionus sp., using as control the commercial product Algamac 2000×. In view of the need to preserve and store the eggs produced in excess, freezedrying was examined as a possible technical solution for this purpose. The fatty acid profiles of the tested enrichment products (fresh seabream eggs, freeze-dried eggs and Algamac 2000×), as well as of the enriched rotifers at 0 h and after 3,6,12, 15, 18 and 24 h of enrichment, were analyzed. Freeze-dried eggs were stored for 14 and 58 wk prior to analysis, in order to examine a potential deterioration of the fatty acid nutritional quality. A potential use of seabream eggs as an enrichment product for Brachionus sp. was demonstrated. Rotifers enriched with eggs, fresh and freeze-dried, presented relatively high levels of polyunsaturated fatty acids (PUFA), highly unsaturated fatty acids (HUFA; × C20 and 2 double bonds), and fatty acid methyl esters (FAME), a docosahexaenoic acid (DHA)/eicosapentaenoic acid (EPA) ratio of about 2 and a fairly high (n-3)/(n-6) ratio, especially during the first 3-6 h of enrichment. When freeze-dried eggs were stored in a dry atmosphere during 14 and 58 wk, only a slight and nonsignificant decrease was noted in the fatty acid content over this period of time. Cataracts are among the most common ocular diseases, and are a leading cause of vision loss in humans and dogs. Jack Russell Terriers (JRT) and Labrador Retrievers (LR) are among the most popular canine breeds in the United Kingdom, and also among the most affected by cataracts.This study aimed to analyze the clinical features and the surgical outcome of cataracts in JRT and LR in an ophthalmologic reference Veterinary Hospital in the United Kingdom. For that purpose, medical records from JRT and LR diagnosed with cataracts between January 2015 and December 2016 were retrospectively evaluated. Data related to identification, clinical history, preoperative features, and surgical outcomes were analyzed.Forty-four dogs (81 eyes), including 26 JRT and 18 LR, were enrolled in the study. Mean ages were 10.2 ± 3.2 years in JRT and 8.5 ± 3.7 years in LR. Twenty-eight (63.6%) were females and 16 (36.4%) were males. Most dogs (84.1%) presented with bilateral cataracts. Nuclear and cortical cataracts were the most prevalent type in both breeds (JRT: n?=?30, 61.2%; LR: n?=?16, 50.0%), although subcapsular cataracts were also frequent in LR (n?=?10, 31.3%). Significant differences in cataract location within the lens were detected between the two breeds (P?=?.002).Senile in JRT (n?=?7) and genetic in LR (n?=?7) were the most common etiologies. Concomitant ocular lesions were more frequent in dogs presented with cataracts in advanced stages, and included lens (n?=?18; JRT: n?=?15; LR: n?=?3) and retinal alterations (n?=?8; JRT: n?=?2; LR: n?=?6), and glaucoma (n?=?6; JRT: n?=?5; LR: n?=?1). Thirty-three animals (75.0%, 51 eyes) were submitted to phacoemulsification with intraocular lens placement. Of these, 28 eyes (54.9%; JRT: n?=?21; LR: n?=?7) were visual, 19 eyes (37.3%; JRT: n?=?11; LR: n?=?8) presented impaired vision and four eyes (7.8%; JRT: n?=?0; LR: n?=?4) were blind at last clinical record. Postoperative complications were detected in 11 eyes (21.6%), and were more frequent in dogs presented with cataracts in advanced stages.These results and the multifactorial nature of cataracts call for further studies to identify and characterize the variables in a broader assessment, including other breeds and influencing factors. 相似文献

In cultural landscapes there are often negative biodiversity consequences of agricultural abandonment and subsequent scrub and forest encroachment, due to homogenization and the loss of early-successional habitats. The common forestry practice of removing understory vegetation to prevent fire hazard (fuel management) probably has the side-effect of ameliorating these consequences, but it is uncertain whether it effectively restores habitats for early-successional species. Here we examine the influence of time since fuel management and management frequency on butterfly assemblages, using a chronosequence of cork oak (Quercus suber) stands spanning about 70 years. Overall species richness increased immediately after management and abundances peaked about 2–3 years later, while both declined thereafter for about 10–20 years to pre-disturbance levels. Richness and abundances were also much higher in recurrently managed stands. Most life history groups showed successional trends similar to the overall species richness and abundances, though consistent positive effects of fuel management were only observed for species with univoltine life cycle, herbaceous layer feeding, larval overwintering, and intermediate body size. Individual species were largely associated with recent and recurrent management, though a few specialists occurred most often in undisturbed stands. These findings suggest that fuel management at <10 years intervals is strongly positive for butterfly assemblages in landscapes under land abandonment. However, to maintain the overall forest biodiversity it is critical that patches of undisturbed habitat are also retained at the landscape scale. 相似文献

Health problems in pedigree dogs have recently been highlighted in the media worldwide and verified internationally by cynological organisations. Collaborative actions are now needed to deal with both existing and future issues. In this article, potential roles for various stakeholders are discussed and the value of national and international platforms for collaborations is stressed. Development of specific strategies for action must be based on criteria of significance, such as severity, prevalence and inheritance, as well as availability of effective preventive measures. Assessment of options should be founded on evidence from appropriate populations-at-risk and consider broader issues, such as demographics and human-animal interactions. Existing data, such as those from insurance statistics and health surveys, should be used as a reference until representative national/international population-level breed-specific data are available. Key issues and challenges, as well as possible strategies to address them, are discussed. 相似文献

The detection of ChIFN production after ex vivo antigenic-stimulation of T-lymphocytes has been evaluated for the first time, as a tool to assess cell-mediated immunity (CMI) after avian influenza (AI) infection in 10-day-old SPF chickens. Preliminarily, recall antigens have been produced either by concentrating and inactivating the whole virus or by dissociating the viral proteins. Biologically and structurally intact forms of the viral proteins were isolated by non-ionic detergents while heating, chemical agents and ionic detergent used for virus inactivation altered the antigenic viral components. The n-octyl-B-D-gluco-pyranoside treatment at low temperature was very efficient to produce AI antigenic proteins used for evaluation of ChIFN production after ex vivo antigenic-stimulation of splenic and peripheral lymphocytes. In addition, protocols to isolate lymphocytes from the respiratory tract - the trachea and the lung - have been adapted for local CMI evaluation after similar ex vivo recall assay. Specific AI CMI in the spleen, the blood and the lung was detected for 5 weeks after low pathogenic AI (LPAI) infection in chickens, while further development is needed for tracheal CMI measurement. 相似文献

In this cross-sectional study, we assessed and mapped the seroprevalence of brucellosis in small-scale dairy farming in an urban and peri-urban area of Tajikistan and investigated factors associated with seropositivity. As urban and peri-urban farming is both an opportunity to improve the livelihood for small-scale farmers and a potential public health hazard, studies are warranted to reveal possible peculiarities in the epidemiology of brucellosis in this type of dairy farming. In total, 904 cows of breeding age belonging to 443 herds in 32 villages were serologically tested with indirect enzyme-linked immunosorbent assay (ELISA) and positive samples confirmed with competitive ELISA. Two logistic regression models were used to investigate an association between seropositivity and risk factors at herd and individual level. The herd and individual seroprevalences were 4.1 and 2.0 %, respectively. Herds with a history of abortions were found to be associated with seropositivity [odds ratio (OR)?=?5.3; 95 % confidence interval (CI), 1.3–21.3]. Large herds with more than eight cattle were more likely to be seropositive compared to smaller herds with one to two cattle (OR?=?13.9; 95 % CI, 1.6–119). The number of calves produced per cow (indicating age) was found to be associated with seropositivity. Younger cows with one to two produced calves were less likely to be seropositive compared to older cows with more than six produced calves (OR?=?0.24; 95 % CI, 0.06–1.0). Neither introduction of new cattle to the herd nor communal grazing was associated with seropositivity. This study shows that infection with Brucella (1) is present in small-scale urban and peri-urban dairy farming in Tajikistan and (2) has significant negative effects on reproductive performance in this farming system and (3) that some previously known risk factors for seropositivity in rural farming system were absent here. 相似文献
