Objective The aim of this study was to define and evaluate a combined inhalation?intravenous anaesthetic protocol for use in equine anaesthesia. Study design Prospective, randomized clinical trial. Animals Twenty‐eight horses (body mass 522 ± 82; 330–700 kg [mean ± SD; range]) with a mean age of 6 ± 4 years (range: 2–18 years) presented to the university hospital for various surgical procedures requiring general anaesthesia. Materials and methods Animals were randomly allocated to one of two treatment groups. Anaesthesia was maintained in 14 horses with halothane alone (H group). The mean end‐tidal halothane concentration was 1.24%. In the second group (n = 14) anaesthesia was maintained with both halothane (end‐tidal concentration 0.61%) and a continuous infusion of a ketamine–guaiphenesin mixture (HKG group). The two techniques were compared in terms of qualitative differences and cardiopulmonary effects. Results The stability of anaesthesia was significantly greater in group HKG and the need for dobutamine to maintain blood pressure was significantly less. Recovery times and quality were acceptable in all cases. There were no significant differences between the groups. Conclusions The infusion of ketamine and guaiphenesin in horses receiving low inspired concentrations of halothane provides suitable surgical anaesthesia and lowers the risk of hypotension. Clinical relevance The anaesthetic technique described in this study is a useful and practical alternative to inhalation anaesthesia using halothane alone. 相似文献

Context
Light pollution is a global change affecting a major proportion of global land surface. Although the impacts of Artificial Light At Night (ALAN) have been documented locally for many taxa, the extent of effect of ALAN at a landscape scale on biodiversity is unknown.Objectives
We characterized the landscape-scale impacts of ALAN on 4 insectivorous bat species Pipistrellus pipistrellus, Pipistrellus kuhlii, Eptesicus serotinus, Nyctalus leisleri, and compared the extent of their effects to other major land-use pressures.Methods
We used a French national-scale monitoring program recording bat activity among 2-km car transect surveys, and extracted landscape characteristics around transects with satellite and land cover layers. For each species, we performed multi-model averaging at 4 landscape scales (from 200 to 1000 m buffers around transects) to compare the relative effects of the average radiance, the proportion of impervious surface and the proportion of intensive agriculture.Results
For all species, ALAN had a stronger negative effect than impervious surface at the 4 landscape scales tested. This effect was weaker than the effect of intensive agriculture. The negative effect of ALAN was significant for P. pipistrellus, P. kuhlii and E. serotinus, but not for N. leisleri. The effect of impervious surface varied among species while intensive agriculture had a significant negative effect on the 4 species.Conclusion
Our results highlight the need to consider the impacts of ALAN on biodiversity in land-use planning and suggest that using only impervious surface as a proxy for urbanization may lead to underestimated impacts on biodiversity.353.

Reservoir siltation because of water erosion is an important environmental issue in Mediterranean countries where storage of clear surface water is crucial for their economic and agricultural development. The high density of gully systems observed in Mediterranean regions raises the question of their contribution to reservoir siltation. In this context, this study quantified the absolute and relative contributions of rill/interrill and gully/channel erosion in sediment accumulation at the outlet of small Tunisian catchments (0·1–10 km2) during the last 15 years (1995–2010). To this end, a fingerprinting method based on measurements of caesium‐137 and total organic carbon combined with long‐term field monitoring of catchment sediment yield was applied to five catchments in order to cover the diversity of environmental conditions found along the Tunisian Ridge and in the Cape Bon region. Results showed the very large variability of erosion processes among the selected catchments, with rill/interrill erosion contributions to sediment accumulated in outlet reservoirs ranging from 20 to 80%. Overall, rill/interrill erosion was the dominant process controlling reservoir siltation in three catchments whereas gully/channel erosion dominated in the other two catchments. We identified the presence of marly gypsum substrates and the proportion of catchment surface covered by soil management/conservation measures as the main drivers of erosion process variability at the catchment scale. These results provided a sound basis to propose guidelines for erosion mitigation in these Mediterranean environments and suggested to apply models simulating both rill/interrill and gully/channel erosion in catchments of the region. Ozone, one of the major atmospheric pollutants, alters tree growth, mainly by decreasing carbon assimilation and allocation to stems and roots. To date, the mechanisms of O3 impact at the cellular level have been investigated mainly on young trees grown in controlled or semi-controlled conditions. In the context of climate change, it is necessary to introduce a valuable defence parameter in the models that currently predict O3 impact on mature trees and the carbon sequestration capacity of forest ecosystems.Context
Air pollution is an important factor that affects negatively forest ecosystems. Among oxidative air pollutants, ozone is considered as the most toxic in terms of impact on vegetation.Aims
This paper focuses on the negative impacts of ozone on trees in controlled conditions or in their natural environment. The current knowledge of the responses at cell level is presented and ways to improve their use for ozone risk assessment of forest stands are discussed.Methods
Information was collected from original papers or reviews, providing an overview of the research conducted over the last 60 years.Results
The negative effects of ozone on carbon assimilation and tree biomass production were reviewed and discussed, with a focus on effects on cell processes implied in cell defence, including stomatal regulation, detoxification, signalling, and biosynthesis of wood compound.Conclusion
In the context of increasing significance of O3 flux approach, this review intends to shed light into the black box of defence processes, which are playing a crucial part within the effective O3 dose modelling. European water legislation enforces increasingly restrictive measures with regards to reduction of water consumption and waste emission in order to minimise the potential environmental impact of the agro industry sector. Fish farms are particularly concerned, but legislation covering effluent discharge varies significantly from country to country. However, recommendations and directives from institutional, national or regional bodies suggest the enforcement of increasingly strict waste reduction measures and the development of waste treatment. Before treatment, it is necessary to evaluate waste production in terms of composition and quantity. The waste quantification methods used today for fish culture systems are either based on direct measurements of nutrient and suspended solid fluxes or on indirect evaluation based on the digestibility coefficients of the feed constituents. The objective of the present study is to evaluate the waste of a freshwater flow through farm using both approaches and to discuss their applicability, drawbacks and advantages from the viewpoints of fish farmers and control authorities. Waste production on the farm was monitored during several 24 hour cycles in order to characterise the effluents of the system. The predictions and measurements for the total nitrogen (TN) parameter were well correlated, but measured and predicted suspended solids (SS) and total phosphorus (TP) values presented a weaker correlation coefficient. The hydrobiological method gives details on the N and P forms of waste but this method is heavy and it is difficult to obtain representative samples and flow rate measurements. The in vivo feeding value of 118 early maize (Zea mays L) hybrids, registered in France between 1958 and 1994, has been estimated
from digestibility measurements with sheep at INRA Lusignan (France). There were great variations in the digestibilities of
organic matter and crude fibre, and in the energy value of these hybrids, ranging respectively between 65.1 and 73.5%, 45.4
and 60.0%, and 0.79 and 0.95 UFL. There was a regular decrease in the average feeding value, regarding the year of registration,
for each of the feeding value traits investigated. From 1958 to date, the frequency of hybrids lower than Mammouth increased,
while the number of hybrids higher than LG11 or Brutus clearly decreased. Across the years, the increase in variations among
hybrids occurred because of the introduction of hybrids with a poorer feeding value, particularly since the end of the 80's.
The extremely efficient genetic pressure in breeding for whole plant yield and stalk standability could explain the drift
in feeding value. A silage maize hybrid with a high yield, a high lodging resistance, and a good feeding value can be available
only if breeders use a digestibility assessment, proved as representative of the digestion in cattle. In addition to a digestibility
evaluation, the prediction of maize ingestibility could be taken into account as soon as a criterion is established.

A summary is given of the results of three sets of electrophysiological experiments on the American cockroach (Periplaneta americana). The effects of DDT and S-bioallethrin [bioallethrin (S)-cyclopentenyl isomer] on single giant axons were studied using the voltage-clamp technique. Whereas both molecules induced long tails of inward (sodium ion) current, the voltage and time-dependency of these tails differed. With DDT, the tail was proportional to the activation of the peak current and decreased with the duration of the pulse, suggesting that the molecules were bound to open sodium channels and delayed their closing. With S-bioallethrin, the voltage dependency of the tail was different from that of the peak current, and the tail current increased exponentially with the duration of the depolarisation, suggesting that the pyrethroid insecticide modified resting (or silent) sodium channels into slowly activating channels. Modified action potentials, mimicking those produced by the two molecules, were computed on the basis of these results. Deltamethrin, one of the most potent pyrethroid insecticides, was applied topically on a leg mechanoreceptor and was found to have little effect on the local ‘receptor potential’ but to inhibit action potential production. The effects of topical applications of deltamethrin on the dorsal part of the abdomen, on nervous activity in the abdominal connectives, were studied under different experimental conditions. The results suggest that the insecticide molecules diffused rapidly through the cuticle, were concentrated in the haemolymph, and eventually reached the central nervous system, where they inhibited nerve activity. 相似文献

A PCR-RFLP test based on a pectate-lyase encoding gene permits the detection of several Erwinia carotovora subspecies, but requires complete DNA extraction. This paper reports on the suitability of a simplified PCR-RFLP protocol to characterise E. carotovora strains and on the performance of PCR, using the same primers, to detect the atroseptica subspecies in substrates of epidemiological significance. A collection of 140 strains from various hosts and geographical origins was characterised for biochemical traits and PCR-RFLPs. PCR performed on boiled bacterial suspensions yielded an amplification product of 434 bp in 109 of the 140 strains. None of the E. carotovora subsp. betavasculorum strains was amplified, even after complete DNA extraction. RFLPs of the PCR product yielded 24 groups, 3 of which were new. Twenty one groups were specific to one subspecies. Several strains biochemically similar to E. carotovora subsp. atroseptica, but growing at 37 °C, showed PCR-RFLP profiles characteristic of E. carotovora subsp. carotovora. Phenetic and cladistic analyses gave three main domains, not strictly related to hosts or geographical origins. The atroseptica (RFLP groups 1 and 2) and wasabiae (group 21) subspecies constituted one of the domains, despite clustering distantly from one another. Host specialisation and molecular homogeneity suggest a clonal structure within these subspecies. Conversely, E. carotovora subsp. odorifera, despite its limited host range and geographical distribution, and E. carotovora subsp. carotovora showed great molecular diversity, spreading respectively across five and 19 RFLP groups. These two subspecies shared RFLP groups 4, 5 and 6. The tree nodes in the phenograms showed a low robustness when bootstrapping the data matrix. PCR coupled with a 48h enrichment step in a polypectate-rich medium improved detection thresholds of E. carotovora subsp. atroseptica (1.5.102- 1.5.103 bacteria/ml in leaves, stems, and tuber peel extracts to 4.107 bacteria/ml in wash water) relative to either immunomagnetic separation coupled with PCR or DAS-ELISA (2.105 in plant samples to 2.107 bacteria/ml in wash water). 相似文献

Severe feed restriction decreases serum insulin-like growth factor I (IGF-I) concentration in animals, and this decrease is thought to be due to reduced IGF-I production in the liver. The objective of this study was to determine whether feed deprivation also increases degradation of serum IGF-I and serum levels of IGF binding protein 3 (IGFBP-3) and acid-labile subunit (ALS), which inhibit IGF-I degradation and increase IGF-I retention in the blood by forming a ternary complex with IGF-I, in cattle. Five steers had free access to pasture, and another five were deprived of feed for 60 h. Serum concentration of IGF-I and liver abundance of IGF-I mRNA at the end of the 60-h period were 50% and 80% lower, respectively, in feed-deprived steers than in fed steers. Less 125I-labeled IGF-I remained intact after a 45-h incubation in sera of feed-deprived steers than in sera of fed steers, suggesting that serum IGF-I is more quickly degraded in feed-deprived animals. Serum levels of IGFBP-3 and ALS were decreased by 40% and 30%, respectively, in feed-deprived steers compared with fed steers. These decreases were associated with more than 50% reductions in IGFBP-3 and ALS mRNA in the liver, the major source of serum IGFBP-3 and ALS. Taken together, these results suggest that feed deprivation reduces serum concentration of IGF-I in cattle not only by decreasing IGF-I gene expression in the liver, but also by increasing IGF-I degradation and reducing IGF-I retention in the blood through decreasing IGFBP-3 and ALS production in the liver. 相似文献
