Dichelobacter nodosus is the principal cause of ovine footrot and strain virulence is an important factor in disease severity. Therefore, detection and virulence determination of D. nodosus is important for proper diagnosis of the disease. Today this is possible by real-time PCR analysis. Analysis of large numbers of samples is costly and laborious; therefore, pooling of individual samples is common in surveillance programs. However, pooling can reduce the sensitivity of the method. The aim of this study was to develop a pooling method for real-time PCR analysis that would allow sensitive detection and simultaneous virulence determination of D. nodosus. A total of 225 sheep from 17 flocks were sampled using ESwabs within the Swedish Footrot Control Program in 2014. Samples were first analysed individually and then in pools of five by real-time PCR assays targeting the 16S rRNA and aprV2/B2 genes of D. nodosus. Each pool consisted of four negative and one positive D. nodosus samples with varying amounts of the bacterium. In the individual analysis, 61 (27.1%) samples were positive in the 16S rRNA and the aprV2/B2 PCR assays and 164 (72.9%) samples were negative. All samples positive in the aprV2/B2 PCR-assay were of aprB2 variant. The pooled analysis showed that all 41 pools were also positive for D. nodosus 16S rRNA and the aprB2 variant. The diagnostic sensitivity for pooled and individual samples was therefore similar. Our method includes concentration of the bacteria before DNA-extraction. This may account for the maintenance of diagnostic sensitivity. Diagnostic sensitivity in the real-time PCR assays of the pooled samples were comparable to the sensitivity obtained for individually analysed samples. Even sub-clinical infections were able to be detected in the pooled PCR samples which is important for control of the disease. In this study, the expression patterns of inhibins, activins, insulin-like growth factor-I (IGF-I) and steroidogenic enzymes in equine placentae recovered during the latter two-thirds of gestation were examined. Concentrations of inhibin A and inhibin pro-alphaC in endometrial and fetal placental tissue homogenates were very low during the period examined, whereas these tissues contained high concentrations of activin A. In both maternal endometrial and fetal placental tissues, activin A levels decreased as pregnancy progressed. Expression of inhibin alpha-subunit was not observed in the placenta using either immunohistochemistry or in situ hybridization. Inhibin/activin betaA-subunit and its mRNA were confined to maternal endometrial glands, whereas immunopositive betaB-subunit was not detected in either endometrial glands or microcotyledons. Cytochrome P450 side chain cleavage enzyme was detected by immunohistochemistry in both endometrial glands and microcotyledons, whereas cytochrome P450 17alpha-hydroxylase/lyase was absent in these tissues. Immunopositive signals for 3beta-hydroxysteroid dehydrogenase and cytochrome P450 aromatase were localized in microcotyledons but not in endometrial glands. Immunohistochemistry revealed that IGF-I was highly expressed in microcotyledons around Day 130, and decreased as pregnancy progressed. Changes in the expression of IGF-I were correlated with the number of PCNA positive cells in the placenta. The present study demonstrated the presence and localized the site of expression of activin, IGF-I and steroidogenic enzymes in equine placental tissues during the latter two-thirds of gestation; the results suggest that activin and IGF-I may be involved in the regulation of placental development. 相似文献

A tumor sized in 2.0x2.0x2.5 cm developed in the cerebellum of a female Beagle was pathologically investigated. Histopathologically, the tumor grew by compression and partially by infiltration into the adjacent cerebellar parenchyma. There were a large number of necrotic lesions and proliferation of collagen fibers. The tumor cells had oval nucleus showing cellular atypia and a high mitotic index. The tumor cells were reacted with vimentin antibody on immunostain. Electron microscopic examination revealed the tumor cells interdigitated with cytoplasmic processus where the desmosomes developed on cell junction. This tumor was diagnosed as anaplastic meningioma, which is rarely observed in dogs. 相似文献

Between January, 2013 and December, 2014, there was a lumpy skin disease (LSD) outbreak that affected cattle in different localities of Zimbabwe. The outbreak resulted in severe economic losses to the livestock industry. A retrospective study was conducted by examining stored veterinary records of the LSD outbreak at the Central Veterinary Laboratory (CVL) in Harare, Zimbabwe. Over the 2-year period, a total of 10,038 cases and 880 deaths (8.77 %) were recorded. LSD was reported from all regions of the country, with the highest incidence occurring in Mashonaland West (30.95 %) and Midlands province (14.59 %). The frequency of reported outbreaks was highest in March and April, with the lowest reported cases occurring in November. A total of 25 representative specimens (skin biopsies) were collected from nodular skin lesions of infected cattle, and after viral DNA isolation, the P32 gene was successfully amplified, by using PCR, in 88 % (22/25) of all assayed specimens. Out of the 22 samples that showed amplification, 16 (73 %) were selected for DNA sequencing, and from these, 13 sequences were submitted to GenBank and assigned accession numbers: KX033494, KX033495, KX033496, KX033497, KXO33498, KX033499, KX033500, KX033501, KX033502, KX033503, KX033504, KX033505 and KX033506. Phylogenetic analyses of the 13 sequences was done by using MEGA 7 and showed that the viruses formed two major clusters implying that at least two strains of LSDV are in circulation in Zimbabwe. Forty-five Pelibuey sheep were experimentally infested with nematodes to evaluate the effect of three free condensed tannin (FCT) levels of Lysiloma acapulcensis on fecal egg counts (FECs), packed cell volumes (PCV), ocular mucosa colors (OMC), average daily gain (ADG), and adult nematode count. Five treatments were used: 12.5, 25.0, and 37.5 mg of FCT kg?1 of body weight (BW); sterile water (control); and ivermectine (0.22 mg kg?1 of BW) as chemical group. The data were processed through repeated measurement analysis. Even though the three FCT doses decreased (P?<?0.05) the FEC, the highest reduction was obtained with 37.5 mg kg?1 of BW. No differences were observed in PCV and OMC. Higher ADG (P?<?0.05) was observed with 37.5 mg kg?1 of BW of FCT. The count of adult nematodes (females and males) in the higher dose of FCT was similar to chemical treatment. Dose of 37.5 mg kg?1 of BW decreased the parasite infection and improved the lamb performance. Therefore, this dose could be used as a nutraceutic product in sheep production. 相似文献

A study was conducted on 37 randomly selected broiler poultry farmers in Rwanda to characterize the production system using pre-tested semi-structured questionnaires. The data were processed in SPSS and presented as means, percentages and ranges in tables and text. All respondents kept Cobb breed and young stock was mainly (73 %) imported from abroad. The majority of respondents were males (68 %) and most farmers had attended only primary level of education (40.5 %). Most of the farms were in the peri-urban (48.6 %) and urban (37.8 %) areas and hired male youth (62.2 %) mainly aged 19–35 years. The majority of respondents (68 %) kept less than 500 birds per batch. Recordkeeping was well practiced (91.9 %) and (62.6 %) had permanent poultry houses and all farmers used deep litter system. Purchased feedstuffs were reportedly (92 %) mixed at farm level as the main feed resource. Maize bran was reported (97.06 %) the main, basal feedstuff. The mortality rates of chick and growers were 12.3 and 9.4 %, respectively. The slaughter age was reportedly 60 days with an average dressing percentage of 75.67 %. The main challenges reported were scarcity and unaffordability of quality feeds (59.5 %), lack of market access (45 %) and lack of credit (21 %). The farmers had various views on improving broiler production in Rwanda ranging from establishing feed processing industries 62.2 %, improving marketing facilities 35.1 %, increasing availability of day-old chick and access credit 27 %, to intensification of farmer training 16.2 %. 相似文献

Understanding the reasons why animals of similar performances have different feed requirements is important to increase profits for cattle producers and to decrease the environmental footprint of beef cattle production. This study was carried out aiming to identify the associations between residual feed intake (RFI) and animal performance, nutrient digestibility, and blood metabolites related to energy balance of young Nellore bulls during the finishing period. Animals previously classified as low (n?=?13) and high RFI (n?=?12), with average initial body weight of 398 kg and age of 503 days were used. Cattle were fed a high energy diet and were slaughtered when rib fat thickness measured by ultrasound between the 12th and 13th ribs reached the minimum of 4 mm. A completely randomized design was adopted, being data analyzed with a mixed model that included the random effect of slaughter group, the fixed effect of RFI class, and linear effect of the covariate feedlot time. No differences were found (p?>?0.10) between RFI classes for performance, dry matter, and nutrients intake. However, dry (p?=?0.0911) and organic matter (p?=?0.0876) digestibility tended to be lower, and digestibility of neutral detergent fiber corrected for ash and protein (p?=?0.0017), and total digestible nutrients (p?=?0.0657) were lower for high RFI animals, indicating lesser capacity of food utilization. Difference between low and high RFI animals was also found for blood cortisol at the end of the trial (p?=?0.0044), having low RFI animals lower cortisol concentrations. This study was conducted to evaluate the feeding value of whole date palm (WDP) with and without enzyme supplementation on aged laying hen’s performance. Apparent metabolizable energy value of WDP was determined by the total collection method using ten adult leghorn cockerels. WDP was substituted with a corn-soybean meal basal diet at 40% level, and then the metabolizable energy of this experimental diet and basal diet was determined. After that, a total number of 256 Bovanz 95-week-old hens were randomly allocated into eight groups consisting of four replicates of eight birds each, based on a 4 × 2 factorial arrangement of the treatments. Eight iso-caloric and iso-nitrogenous experimental diets including one corn-soybean meal-based control diet and two, three, and four corn-soybean meal-based diets included 70, 140, and 210 g/kg ground WDP, respectively. Each of the diets supplemented with two levels of an enzyme (0.0 and 0.07 g/kg Natozim Plus). There was no significant difference in feed intake, feed conversion ratio, egg production, egg mass, eggshell thickness, and Haugh unit among the treatments (P > 0.05). However, yolk color score significantly decreased as dietary WDP level increased. The serum biochemical metabolites were not significantly affected by WDP and enzyme supplementation (P > 0.05). There were no significant differences in the relative weight of different organs except for abdominal fat. Our findings show that using WDP up to 21% of the diet was more economic and had no adverse effect on productive performance and serum metabolites of laying hens. However, WDP had an adverse effect on yolk color which can be ameliorated by carotenoid supplementation. 相似文献

In the recent past, pig performance and sow management on smallholder farms in the Southeast Asian Massif have been extensively researched. But the objective of this study was to investigate the lifetime efficiency of breeding sows of the local Vietnamese Ban (n = 89) and Mong Cai (n = 131) breeds in a production system context. The impact of sow longevity was considered in particular. In addition, predictors of early pre-weaning mortality in piglets were investigated. In total, nine villages of Son La province, situated in the uplands of north-western Vietnam, were selected, differing in access to markets, production intensity, and pig management level. Data was collected over a period of 10 years through an on-farm performance testing system. It was shown that sows of both breeds that achieved five or more parities farrowed and weaned significantly more piglets per life year compared to sows with only one or two parities. The conception rate (Spearman’s rho = 0.374) and age at first farrowing (Spearman’s rho = ?0.424) were significantly correlated with the number of weaned piglets per life year of stayable (≥4 parities) Mong Cai sows. The risk of early pre-weaning death of piglets born to Mong Cai sows was affected more by litter size, while for piglets born to Ban sows, the risk was influenced more by the season. Therefore, interventions to improve the sow lifetime efficiency and piglet survivability must match the sow breed and management level. 相似文献
