The effects of addition of alpha-casein (alpha-CN) to dried egg white (DEW) were investigated by measuring transparency, hardness, and water-holding capacity (WHC) of the heat-induced gels. A DEW concentration of 8% (w/w) was required for formation of a self-supporting gel following heating at 80 degrees C for 20 min at pH 7. Solutions of alpha-CN, even up to a protein concentration of 12% (w/w), did not gel under the same conditions. The addition of alpha-CN (0.5-4%) to 8% DEW caused the increase in gel hardness gels, as compared with DEW gels alone at a total amount of protein concentrations, and the mixed gels became transparent with the increase of added alpha-CN concentrations. The 10% mixed protein solutions of alpha-CN (3-6%) and DEW (4-7%) formed transparent gels, although each protein did not gel individually at their protein concentrations. Mixture with 2:8 mixing ratio of alpha-CN to DEW at a total protein concentration of 10% showed synergistic effects in improving DEW gel properties above pH 7 and below 25 mM NaCl. The improvements (hardness, transparency, and WHC) of DEW gel by alpha-CN seem to be caused mainly by the inhibition of alpha-CN against heat coagulation of DEW protein.  相似文献

A 15-day-old Holstein calf with lethargy and tachypnea presented to the Veterinary Teaching Hospital at Obihiro University of Agriculture and Veterinary Medicine for evaluation of suspected congenital heart defect. A Levine grade 6 systolic murmur was noted at right apical site auscultation and phonocardiogram also recorded systolic a murmur. Electrocardiography findings include increased R and S waves, R wave split, and negative T waves without arrhythmia. Echocardiography revealed a single ventricle with a trace of the right ventricular wall, atrioventricular valve regurgitation, and turbulent in a single ventricle. Arterial blood analysis showed a marked decrease in oxygen saturation of 78% and oxygen partial pressure of 44 mmHg. Post-mortem examination confirmed the diagnosis of a single left ventricle.  相似文献

The decrease in fertility and conception rates of high-producing dairy cows is one of the major negative impacts for today's producers. The recovery of ovarian activity postpartum is affected by the status of immunity, metabolism and reproduction and plays a critical role in subsequent fertility after parturition in the cow. In the present study we investigated the relationships between polymorphisms in genes relating to the above functions and the first postpartum ovulation as a marker of the recovery of ovarian function in the cow. In immune function related-factors, the occurrence of first postpartum ovulation within 3 weeks in the C/C genotypes of tumor necrosis factor α (TNFα) exon (55.4%) and the A/G genotypes of TNFα promoter (55.4%) was significantly higher than that in T/T genotypes of TNFα exon (14.3%) and A/A genotypes of TNFα promoter (14.3%). Moreover, anovulatory cows with the T/T genotype of TNFα exon and the A/A genotype of TNFα promoter tended to have a prolonged days open compared with those of the other genotypes of TNFα polymorphisms. In metabolic function-related factors, ovulatory and anovulatory cows had a different distribution for alleles of the growth hormone receptor, but there were no significant differences in genotype and allele frequency of insulin-like growth factor-I polymorphism. No significant relationships were found between ovarian function after parturition and polymorphisms for reproduction-related genes. In conclusion, polymorphisms of TNFα gene both in exon and promoter regions have a strong association with the early first ovulation within 3 weeks after parturition in the high-producing dairy cow. Taken together, polymorphisms of TNFα gene could be strongly related to early first ovulation after parturition, thus being an effective tool of selection for improving reproductive performance in the high-producing dairy cow.  相似文献

The purpose of this study was to evaluate the effect of protein malnutrition on the skin epidermis of mice. A low protein diet induced thinning of the skin epidermis, a decrease of cell proliferative activity in epidermal cells and a decrease of stratum corneum hydration. Dityrosine was immunostained in the cytoplasm of epidermal cells in the low protein diet group. Plasma advanced oxidation protein product (AOPP) levels were significantly more increased in the low protein diet group than in the control diet group. These results suggest that protein malnutrition adversely affects the structure and water barrier and reservoir functions of the skin epidermis, and these pathological changes are associated with the expressions of protein oxidation markers, dityrosine and AOPP.  相似文献

Because of the increasing use of maize hybrids with genetically modified (GM) stacked events, the established and commonly used bulk sample methods for PCR quantification of GM maize in non-GM maize are prone to overestimate the GM organism (GMO) content, compared to the actual weight/weight percentage of GM maize in the grain sample. As an alternative method, we designed and assessed a group testing strategy in which the GMO content is statistically evaluated based on qualitative analyses of multiple small pools, consisting of 20 maize kernels each. This approach enables the GMO content evaluation on a weight/weight basis, irrespective of the presence of stacked-event kernels. To enhance the method's user-friendliness in routine application, we devised an easy-to-use PCR-based qualitative analytical method comprising a sample preparation step in which 20 maize kernels are ground in a lysis buffer and a subsequent PCR assay in which the lysate is directly used as a DNA template. This method was validated in a multilaboratory collaborative trial.  相似文献

Abstract

Slag silicate fertilizer (SSF) is applied to paddy fields with different soil chemical properties to increase silicon (Si) concentration in rice (Oryza sativa L.) plants. However, the effects of soil chemical properties on Si availability of SSF to rice Si uptake is poorly understood. To investigate the relationships between chemical properties of soils and the effects of SSF application on the Si concentration in rice plants, a field experiment was conducted in 2007 and 2008 at 18 paddy fields on the Shounai Plain, Yamagata, Japan. Two treatments were implemented: SSF applied at 1.5 t ha?1 and a no-SSF control. The Si concentrations of rice tissues were measured at the tillering and ripening stages. The difference in the Si concentrations of rice tissues between treatments (ΔSi concentration) was used to evaluate the effect of SSF. The Si concentrations in the shoots and aboveground parts of the rice plants were significantly increased by the SSF application in six or more of the fields at the two growth stages, whereas the Si concentrations in the panicles of the rice plants at the ripening stage were not increased significantly in most fields. Results of two-way analysis of variance evidenced a significant effect of field on the ΔSi concentrations in the shoots and aboveground parts at both growth stages. Furthermore, the ΔSi concentrations in the same rice plant tissues and at the same growth stages in the first year and in the second year were found to be positively correlated. These results indicate that the effect of SSF on the Si concentration in shoots and aboveground parts of rice plants varies from field to field. The ΔSi concentrations in shoots and aboveground plant parts at both growth stages were also found to be negatively correlated with soil chemical properties, i.e., available Si, Si adsorption capacity, contents of Si adsorbents (acid oxalate-extractable iron and manganese) and the pH under flooded soil conditions. These findings imply that those soil chemical properties of paddy fields should be taken into account for better prediction of ΔSi concentration of rice plants.  相似文献

Even though tetrodotoxin (TTX) is a widespread toxin in marine and terrestrial organisms, very little is known about the biosynthetic pathway used to produce it. By describing chemical structures of natural analogs of TTX, we can start to identify some of the precursors that might be important for TTX biosynthesis. In the present study, an analog of TTX, 5,11-dideoxyTTX, was identified for the first time in natural sources, the ovary of the pufferfish and the pharynx of a flatworm (planocerid sp. 1), by comparison with totally synthesized (−)-5,11-dideoxyTTX, using high resolution ESI-LC-MS. Based on the presence of 5,11-dideoxyTTX together with a series of known deoxy analogs, 5,6,11-trideoxyTTX, 6,11-dideoxyTTX, 11-deoxyTTX, and 5-deoxyTTX, in these animals, we predicted two routes of stepwise oxidation pathways in the late stages of biosynthesis of TTX. Furthermore, high resolution masses of the major fragment ions of TTX, 6,11-dideoxyTTX, and 5,6,11-trideoxyTTX were also measured, and their molecular formulas and structures were predicted to compare them with each other. Although both TTX and 5,6,11-trideoxyTTX give major fragment ions that are very close, m/z 162.0660 and 162.1020, respectively, they are distinguishable and predicted to be different molecular formulas. These data will be useful for identification of TTXs using high resolution LC-MS/MS.  相似文献

In western Japan, Moso bamboo (Phyllostachys pubescens) forests have been expanding by replacing surrounding vegetation such as coniferous plantation forests and natural broadleaved forests. It has been speculated that the replacement of surrounding vegetation by bamboo forests could alter the vegetation water cycle and available water resources. We quantified stand-scale transpiration (E) in a bamboo forest on the basis of sap-flux measurements and compared the E value with values for coniferous forests. The annual E was estimated to be 567 mm. Seasonal trends in E primarily corresponded to seasonal trends in the vapor pressure deficit. Annual E for the bamboo forest was higher than that for the coniferous forests by 12% of annual precipitation (P). This difference in annual E is comparable with the difference in annual interception evaporation (I) relative to P between bamboo and coniferous forests; previous studies reported lower I for bamboo forests by ∼10% of P. Thus, the sum of E and I was comparable for bamboo and coniferous forests. As this study is the first measuring E of bamboo forests, further studies are required to examine the generality of our results.  相似文献

Seagrasses are known to have allelopathic activity to reduce growth of phytoplankton. We found growth-inhibiting bacteria (strains E8 and E9) from Zostera marina possessing strong activity against the toxic dinoflagellate Alexandrium tamarense. Strain E9 markedly inhibited growth of A. tamarense even with initial inoculum size as small as 2.9 cells ml?1. This bacterium also had growth-inhibiting effects on the red-tide raphidophytes Chattonella antiqua and Heterosigma akashiwo, the dinoflagellate Heterocapsa circularisquama, and the diatom Chaetoceros mitra. Small subunit (SSU) ribosomal DNA (rDNA) sequencing analysis demonstrated that the most probable affiliation of these strains was Flavobacteriaceae, and proved that another inhibitory bacterial strain (E8) was the same species as strain E9. Two other bacterial strains (E4-2 and E10), showing different colony color and isolated from the same seagrass sample, revealed no growth-inhibiting activity. Interestingly, strain E4-2 showed the same sequences as E8 and E9 (100 %), and strain E10 matched E8 and E9 with 99.80 % similarity. Growth-inhibiting bacteria against the toxic dinoflagellate Alexandrium tamarense associated with seagrass, such as Flavobacterium spp. E8 and E9, are able to repress shellfish poisoning besides the allelopathic activity of seagrass itself.  相似文献

In mass culture of Pacific bluefin tuna Thunnus orientalis, a marked growth variation is observed after they start feeding at 6–7 mm in body length (BL) on yolk‐sac larvae of other species, and the growth variation in tuna larvae is a factor leading to the prevalence of cannibalism. To examine the relationship between prey utilization and growth variation, nitrogen stable isotope ratios (δ15N) of individual larvae were analysed. A prey switch experiment was conducted under two different feeding regimes: a group fed rotifers (rotifer fed group), and a group fed yolk‐sac larvae of spangled emperor, Lethrinus nebulosus (fish fed group) from 15 days after hatching (6.87 mm BL). The fish fed group showed significantly higher growth than the rotifer fed group. Changes in the δ15N of the fish fed group were expressed as an exponential model and showed different patterns from those of the rotifer fed group. The δ15N of fast‐growing tuna larvae collected in an actual mass culture tank after the feeding of yolk‐sac larvae was significantly higher than those of the slow‐growing larvae, indicating that slow glowing larvae depended largely on rotifers rather than the yolk‐sac larvae.  相似文献
