Changes in immune factors expressed by milk somatic cells from Holstein cows with hypocalcemia after calving were investigated in this study. Fourteen multiparous Holstein cows after their 3rd or 4th calving in one farm were used. The cows were divided into 2 groups: 7 cows needing treatment due to onset of hypocalcemia (hypocalcemia group; age = 5.53 ± 0.27 years, parity = 3.14 ± 0.14) and 7 cows without health problems (control group; age = 5.88 ± 0.31 years, parity = 3.57 ± 0.26). Milk samples were collected aseptically using a cannula and mRNA of immune factors expressed by milk somatic cells were analyzed. Milk samples (50 mL) were collected from the right rear mammary gland of cows before milking at day 1 and weeks 1, 2, 4, and 8 after calving. All milk samples showed a negative reaction to the California Mastitis Test. Levels of relative interleukin (IL)-6 and cathelicidin in the hypocalcemia group were lower than those in the control group in weeks 1 to 8. A significant difference in relative IL-6 levels was found in week 4 (P < 0.05). These results suggest that levels of IL-6 expressed by milk somatic cells may be affected by hypocalcemia in dairy cows. 相似文献

This paper presents a study carried out by a group of Japanese researchers to critically review past researches and discussions on the multifunctionality of paddy rice cultivation, with the aim of describing the current status of the subject and providing options and potentials for future research and practices. The review was of over 200 documented articles on the major external functions. The results of this study revealed that the importance of multifunctionality is well perceived in Japan. The methodologies to measure and estimate the magnitude have been established for most functions. However, there is a need for more cases, to integrate all the functions at the local or regional scale to represent site-specific characteristics of multifunctionality. There is also a need to develop management practices to sustain and enhance the positive multifunctionality of paddy rice cultivation while minimizing negative effects to the environment, which may be brought into the agricultural policies. Except for Korea, Taiwan, and Japan, there are slightly different notions on multifunctionality in other monsoon Asia countries, but the gap in this notion is reducing following recent international activities. 相似文献

Water quality in the watershed mainly used for agriculture is degraded by nutrients from fertilizers and animal wastes. These diffuse (non-point) sources have accumulated in soils and been released into the river system for the long-term. In this paper, a water quality tank model classified by land use is modified for the accumulation, and total nitrogen (T-N) is simulated over 40 years in the Yamada River basin within the Lake Kasumigaura watershed, Japan. For considering the nitrogen balance, the model has a database comprising statistical data such as population, land use, rain, fertilizer, and animal waste; it calculates cumulative load in soils and dissolved load separately. The model simulated the measured data with relative error of 15% for runoff and 7% for T-N. Five scenarios for the reduction of cumulative load were developed as a countermeasure plan: soil washing, slow-release fertilizer, fertilizer application reduction, cover crop, and animal waste reduction. The results show that T-N first decreases and then increases after 30 years in the case of the slow-release fertilizer scenario and that T-N is reduced by 30% after 40 years in the case of the 100% animal waste reduction scenario. 相似文献

Background
The rice interactome, in which a network of protein-protein interactions has been elucidated in rice, is a useful resource to identify functional modules of rice signal transduction pathways. Protein-protein interactions occur in cells in two ways, constitutive and regulative. While a yeast-based high-throughput method has been widely used to identify the constitutive interactions, a method to detect the regulated interactions is rarely developed for a large-scale analysis.Results
A split luciferase complementation assay was applied to detect the regulated interactions in rice. A transformation method of rice protoplasts in a 96-well plate was first established for a large-scale analysis. In addition, an antibody that specifically recognizes a carboxyl-terminal fragment of Renilla luciferase was newly developed. A pair of antibodies that recognize amino- and carboxyl- terminal fragments of Renilla luciferase, respectively, was then used to monitor quality and quantity of interacting recombinant-proteins accumulated in the cells. For a proof-of-concept, the method was applied to detect the gibberellin-dependent interaction between GIBBERELLIN INSENSITIVE DWARF1 and SLENDER RICE 1.Conclusions

The effect of genome composition and cytoplasm on petal size was studied in Brassica. Two accessions of Brassica rapa (2n = 20, AA) were reciprocally crossed with three accessions of Brassica oleracea (2n =18, CC) to produce resynthesized B. napus (2n = 38, AACC or CCAA) and sesquidiploids (2n = 29, AAC or CAA). Petal size was measured and compared among diploids (AA and CC), sesquidiploids (AAC and CAA) and amphidiploids(AACC and CCAA). The results showed that petal size is a genome‐dependent and highly heritable character. The heritability of petal length is as high as 96.3%. The addition of each C‐genome to the AA genomic background increased the petal length by 4‐5 mm. Cytoplasm of B. oleracea showed a positive effect on petal length by about 1.3 mm over that of B. rapa. Petal width was positively correlated with petal length at a highly significant level (r= 0.806, df = 81). Resynthesized B. napus (AACC) showed significantly larger flower petals than natural rapeseed cultivars (AACC). 相似文献

Genetic structure and phylogenetic relationships in East and South Asian melons were analyzed, based on the geographical variation
of five isozymes. The analysis of Indian melon accessions showed a continuous variation in seed length, ranging from 4 to
13 mm. Most of the East Asian melons, vars. makuwa and conomon, were classified as the small seed type with seed length shorter than 9 mm. The frequency of the small seed type increased
from the west to the east in India. Allelic variation was detected at a total of nine loci of five isozymes among 114 melon
accessions. Gene diversity calculated for the nine loci indicated that Indian melon was rich in genetic variation, which decreased
from India towards the east. Clear geographical variation was detected in two enzymes, APS and6-PGDH. Pgd-1
1 and Ap-3
1 were frequent in India and Myanmar, while most of the melons in Laos, China, Korea and Japan carried Pgd-1
3 and Ap-3
3, except var. inodorusin China. Among the latter two alleles, the frequency of Ap-3
3 was more than 50% in the small seed type in north and east India, indicating that vars. makuwa and conomon were related to the small seed type in these areas. It was also suggested that the small seed type with wet tolerance originated
in central India and was selected under wet condition in the east.

Adaptation and ecological differentiation in wheat with special reference to geographical variation of growth habit and Vrn genotype 总被引:1,自引:0,他引:1
Geographical variation of growth habit was studied for 749 landraces from various parts of the world, with special reference to their adaptation and ecogeographical differentiation. The total frequency of spring‐type landraces was 49.9%, and varied between localities. Spring‐type landraces were frequent in two distinct areas where the average January temperature was either below ‐7°C or above 4°C, with winter‐type landraces in areas from ‐7°C to 4°C. These results indicated that geographical variation of growth habit is closely related to the degree of winter coldness. An analysis of the Vrn genotype for 216 spring‐type landraces demonstrated the uneven distribution of four Vrn genes, with Vrn4 being the least frequent. The adaptive Vrn genotype was different between localities. Genotypes carrying Vrn‐A1 and additional Vrn gene(s) were frequent in two distinct areas where the average January temperature was either below ‐7°C or over 10°C, while genotypes with any of three Vrn genes, except Vrn‐A1, adapted to areas with temperatures from 4°C to 10°C. Therefore, it was concluded that the adaptability of wheat landraces differed depending on their growth habit and Vrn genotype, and that ecotypes with different Vrn genotypes were allopatrically distributed as a result of adaptation to different winter temperature. However, the differential distribution of Vrn‐B1, Vrn‐D1 and Vrn4 could not be explained by their adaptability, and might reflect the polyphyletic origin of common wheat. 相似文献

Mitochondrial DNA (mtDNA) wascharacterized by RFLPs in 94 strains offoxtail millet, Setaria italica (L.)P. Beauv. Three RFLP patterns wereobserved by using rice atp6 as aprobe, and were designated as types I-III. Difference between types I and II seems tobe attributed to recombination between twoatp6 genes. In East and SoutheastAsia and Afghanistan, both of types I andII were found, while type I was predominantin India, Central Asia and Europe. InChina, type III was also found.
