In this study, the possible influence of temperature on infectious pancreatic necrosis virus (IPNV)-induced apoptosis in a zebrafish liver epithelium (ZLE) cell line was investigated. At a lower temperature (18 degrees C), there was expression of viral proteins VP2 and VP3 at 4 h post-infection (p.i.). At this time no expression was found in the high temperature group at 28 degrees C. The cell survival ratio was 52 and 18% at 24 and 48 h p.i., respectively, during IPNV infection at 18 degrees C. In addition, we assayed for apoptosis in IPNV-infected cells with terminal deoxynucleotidyl transferase (TdT)-mediated end labelling (TUNEL) of DNA at different dosages of virus. We found a ratio of apoptotic cells of 8 and 25% at 12 and 18 h p.i., respectively, in the multiplicity of infection (MOI) 1 group. The MOI 10 group had 20 and 45% apoptotic cells at 12 and 18 h, respectively. Furthermore, at 18 degrees C IPNV activated the caspase-8 and 3 from 1.5 to 2 times at 12 and 18 h p.i., respectively. Taken together, these findings suggest that successful virus replication occurs at the low temperature (18 degrees C) compared with the non-permissive temperature of 28 degrees C. Thus, IPNV replication is capable of activating caspase-8 and -3 and inducing host apoptosis.  相似文献

宁德三都澳海区褐菖鲉(Sebasticus marmoratusCuvier)经375 d不同方式的网箱养殖比较,与大黄鱼混养的褐菖鲉个体平均体重增重量达80.9 g,成活率72.8%;单养Ⅱ网箱底部投放石莼等活体藻类,褐菖鲉个体增重量为71.0 g,成活率72.5%;单养Ⅰ只放养褐菖鲉,个体增重量为61.6 g,成活率67.1%。生长速度和成活率:混养单养Ⅱ单养Ⅰ,可知褐菖鲉与大黄鱼混养效果最好。水温高于26℃,褐菖鲉极易死亡,采取网箱底部投放大型活体藻类、不投饵或少投饵、不更换网衣等度夏措施是养殖成功的关键。  相似文献

The effects of salinity on hemolymph osmotic pressure, gill Na+/K+‐ATPase activity and dopamine (DA), norepinephrine (NE), serotonin (5‐HT) in the gills, and hemolymph of the adult Charybdis japonica were studied. DA levels increased significantly (P < 0.05), while the NE and 5‐HT revealed contrary change in hemolymph and gills. The iso‐osmotic point of C. japonica (911.4 mOsm/kg) was at salinity of 27.87 ppt. The Na+/K+‐ATPase activity of gill showed negative correlation with salinity in the hypotonic environment (<27.87 ppt). The results of this experiment indicated that C. japonica had great capability to acclimate to low salinity.  相似文献

A 63‐day growth trial was undertaken to estimate the effects of supplemented lysine and methionine with different dietary protein levels on growth performance and feed utilization in Grass Carp (Ctenopharyngodon idella). Six plant‐based practical diets were prepared, and 32CP, 30CP and 28CP diets were formulated to contain 320 g kg?1, 300 g kg?1 and 280 g kg?1 crude protein without lysine and methionine supplementation. In the supplementary group, lysine and methionine were added to formulate 32AA, 30AA and 28AA diets with 320 g kg?1, 300 g kg?1 and 280 g kg?1 dietary crude protein, respectively, according to the whole body amino acid composition of Grass Carp. In the groups without lysine and methionine supplementation, weight gain (WG, %) and specific growth rate (SGR, % day?1) of the fish fed 32CP diet were significantly higher than that of fish fed 30CP and 28CP diets, but no significant differences were found between 30CP‐ and 28CP‐diet treatments. WG and SGR of the fish fed 32AA and 30AA diets were significantly higher than that of fish fed 28AA diets, and the performance of grass carp was also significantly improved when fed diets with lysine and methionine supplementation (P < 0.05), and the interaction between dietary protein level and amino acid supplementation was noted between WG and SGR (P < 0.05). Feed intake (FI) was significantly increased with the increase in dietary protein level and the supplementation of lysine and methionine (P < 0.05), but feed conversion ratio (FCR) showed a significant decreasing trend (P < 0.05). Two days after total ammonia nitrogen (TAN) concentration test, the values of TAN discharged by the fish 8 h after feeding were 207.1, 187.5, 170.6, 157.3, 141.3 and 128.9 mg kg?1 body weight for fish fed 32CP, 32AA, 30CP, 30AA, 28CP and 28AA diets, respectively. TAN excretion by grass carp was reduced in plant‐based practical diets with the increase in dietary protein level and the supplementation of lysine and methionine (P < 0.05). The results indicated that lysine and methionine supplementation to the plant protein sources‐based practical diets can improve growth performance and feed utilization of grass carp, and the dietary crude protein can be reduced from 320 g kg?1 to 300 g kg?1 through balancing amino acids profile. The positive effect was not observed at 280 g kg?1 crude protein level.  相似文献

在室内可控条件下,对尼罗罗非鱼[初始体质量(62.50±3.44)g]进行饥饿28d和随后再投喂21d的处理,于饥饿第0、7、14、21、28天和再投喂第14、21天进行采样分析,研究饥饿和再投喂期间尼罗罗非鱼生长、血清生化指标和肝胰脏生长激素(GH)、类胰岛素生长因子-Ⅰ(IGF-Ⅰ)和胰岛素(IN)mRNA表达丰度的变化。结果显示,与饥饿第0天相比,饥饿超过7d鱼体体质量显著降低(P<0.05),再投喂21d显著增加(P<0.05);肝体比随饥饿时间延长显著降低(P<0.05),恢复投喂后较饥饿时升高,但显著低于饥饿前水平(P<0.05)。在血清指标上,甘油三酯、血糖、碱性磷酸酶、谷草转氨酶和谷丙转氨酶均随饥饿时间延长而逐渐降低,恢复投喂后均有不同程度提高,但转氨酶活性显著低于饥饿前水平(P<0.05);饥饿和再投喂对血清总胆固醇、高密度脂蛋白胆固醇和低密度脂蛋白胆固醇无显著影响(P>0.05)。在激素方面,与饥饿第0天相比,饥饿使血清GH含量及其肝胰脏mRNA表达丰度显著升高,血清IGF-Ⅰ及其肝胰脏mRNA表达丰度降低,恢复投喂后两者均显著升高(P<0.05);INmRNA表达丰度在饥饿7~21d显著升高(P<0.05),饥饿第28天时无显著差异(P>0.05),再投喂后显著降低(P<0.05)。  相似文献

Argopecten irradians is one of the most important commercial species of Pectinidae family in China. The internal defense system of mollusks consists of circulating haemocytes. In order to characterize the haemocytes of the scallop A. irradians, light and electron microscopical studies were carried out. Four types of haemocytes were recognized: type I small hyalinocytes (2.38 ± 0.08 μm, 30-35%), type II large hyalinocytes (4.41 ± 0.33 μm, 15-20%), type III small granulocytes (4.15 ± 0.26 μm, 20-25%), and type IV large granulocytes (8.26 ± 0.52 μm, 25-30%). Granulocyte types showed smaller N/C ratios than hyalinocytes. The mean haemocyte concentration was about (3.75 ± 0.65) × 107 cells ml− 1 of haemolymph. Among haemocytes, 44.7% are granular and 55.3% are agranular. These gave a relatively systematic classification scheme for haemocytes of A. irradians. Three types of granules were identified: type I, with high electron-density; type II, with low electron-density; and type III, with a middle level of electron-density, based on TEM studies. Different haemocyte types were not separated with DDGC of Percoll in this study. Both granulocytes and hyalinocytes showed a phagocytic response to the two strains of bacteria, Escherichia coli and RLOs. The phagocytic ability of granulocyte was significantly higher (41-48%) than that of hyalinocyte (9.2-11.2%).  相似文献

Research on the little loligo squid (Uroteuthis chinensis) protein hydrolyzed by papain and Flavourzyme and characterization of hydrolysates was carried out in this article. The optimum hydrolysis conditions of papain were obtained by orthogonal experiments, and the influence factors on the degree of hydrolysis such as enzyme/substrate ratio, hydrolysis time, pH, and temperature were investigated. The effect of Flavourzyme on the degree of hydrolysis was also studied. The characteristics of freeze-dried hydrolysates such as yield, moisture, nitrogen recovery, molecular mass distribution, and amino acid composition were determined. The results showed that papain and Flavourzyme could hydrolyze the little loligo squid protein efficiently, and the degree of hydrolysis was up to 25.82% under the optimum hydrolysis conditions. The average molecular mass of hydrolysates obtained with papain and Flavourzyme was 1,246 Da, and the low molecular mass peptides were the main fraction of hydrolysates. The yield, moisture, and nitrogen recovery of freeze-dried hydrolysates was 19.8, 4.4, and 74.2%, respectively. Compared with suggested patterns of amino acid requirements by the Food and Agriculture Organization of the United Nations/World Health Organization (FAO/WHO), the hydrolysates of the little loligo squid had high nutritional value, and was a potential nutritious supplement used in various food products.  相似文献

Abstract

The objectives of this study are to analyze the interdependent behavior of lead‐lag milkfish price adjustments between vertically‐related markets (from production to consumption) as well as between five major horizontally‐related wholesale markets. Empirical results show that there is a two‐way lead‐lag cointegrating price relationship between milkfish wholesale and retail markets. Since the fish wholesale market cannot completely and quickly assimilate the retail market information, the speed of response of wholesale prices to retail price changes is relatively slow, inducing low pricing efficiency. The Taipei wholesale market appears to cause the greatest leadership impact on the milkfish price formation compared to other wholesale markets in both the long‐run and short‐run. The results also reveal that there is a high degree of integration relationship between the horizontally integrated wholesale markets (Chiayi, Changhwa and Taichung) for milkfish in the southwestern region within a short distance.  相似文献
