A highly conserved neutralizing epitope on group 2 influenza A viruses   总被引:1,自引:0,他引:1  
Current flu vaccines provide only limited coverage against seasonal strains of influenza viruses. The identification of V(H)1-69 antibodies that broadly neutralize almost all influenza A group 1 viruses constituted a breakthrough in the influenza field. Here, we report the isolation and characterization of a human monoclonal antibody CR8020 with broad neutralizing activity against most group 2 viruses, including H3N2 and H7N7, which cause severe human infection. The crystal structure of Fab CR8020 with the 1968 pandemic H3 hemagglutinin (HA) reveals a highly conserved epitope in the HA stalk distinct from the epitope recognized by the V(H)1-69 group 1 antibodies. Thus, a cocktail of two antibodies may be sufficient to neutralize most influenza A subtypes and, hence, enable development of a universal flu vaccine and broad-spectrum antibody therapies.  相似文献

It is difficult to obtain fossil data from the 10% of Earth's terrestrial surface that is covered by thick glaciers and ice sheets, and hence, knowledge of the paleoenvironments of these regions has remained limited. We show that DNA and amino acids from buried organisms can be recovered from the basal sections of deep ice cores, enabling reconstructions of past flora and fauna. We show that high-altitude southern Greenland, currently lying below more than 2 kilometers of ice, was inhabited by a diverse array of conifer trees and insects within the past million years. The results provide direct evidence in support of a forested southern Greenland and suggest that many deep ice cores may contain genetic records of paleoenvironments in their basal sections.  相似文献

In mango (Mangifera indica) cv. Nam Dok Mai fruit, stored at 4 °C, peel browning occurred within 9 d, while no browning was found in cv. Choke Anan fruit stored at 4 °C for 30 d. During 6 d of shelf life at 27-28 °C, following various periods of low temperature storage, the peel browning in cv. Nam Dok Mai (if not yet maximal) became worse, whereas little browning was observed in cv. Choke Anan fruit. The pulp of the fruit of both cultivars did not show browning during the 4 °C storage, but the pulp of cv. Nam Dok Mai exhibited some browning during shelf life if the fruit had been stored at 4 °C for more than 18 d. Peel and pulp color were not correlated with total free phenolics. A high correlation coefficient was observed between peel browning and PAL activity in the peel, while a very low correlation was found with peel catechol oxidase activity. The browning in the pulp was not correlated with the measured enzyme activities. The data therefore show a relation between PAL activity in the peel and low temperature-induced peel browning.  相似文献

Thinning of fruit-tree blossoms is used to regulate the yearly tree bearing and to increase the fruit yield and quality. While this is still mostly done by hand, the increasing costs of manual labor have created a demand for mechanization. This has recently led to the development of several prototype thinning machines. The main disadvantage of these machines is that they are not selective, while the fruit bearing capacity of different floral buds is not equal. On-line information about the position and distribution of the floral buds on the tree can improve the efficiency of mechanized thinning. Therefore, the aim of this study was to identify the most informative wavebands to develop a multispectral vision sensor for detection of the floral buds of the pear cultivar Conference. Hyperspectral scans were taken from tree samples in five early phenological stages to create a database of reflectance spectra for the different tree features. A stepwise algorithm was then applied to this training set to select the best combination of wavebands having the highest discriminating power between the components of interest. Subsequently, canonical correlation analysis was used to create discriminant functions out of the selected wavebands. It was possible to correctly classify 95 % of the (pixel) observations with six selected wavebands. The discrimination performance was also tested as a function of the number of used wavebands. Analysis showed that when only the two most important wavebands were used, still over 90 % of the (pixel) observations could be correctly classified.  相似文献

Cruciferous vegetables contain glucosinolates, which are degraded to isothiocyanates. These are easily absorbed, conjugated to glutathione, and excreted into the urine as their corresponding mercapturic acids. We have developed and validated a solid phase extraction-high-performance liquid chromatography-electrospray ionization mass spectrometry/mass spectrometry method for the specific analysis of individual isothiocyanate mercapturic acids in urine. The range of reliable analysis was 1.0-310 microM in urine. Urine samples fortified with three different levels of isothiocyanate mercapturic acids were measured on six different days by three independent technicians. The relative standard deviation (RSD) of repeatability was 12, 6, and 3%; the RSD of reproducibility was 19, 14, and 8%, and spike recoveries were 103, 104, and 103%, respectively, for 1.04, 10.5, and 313 microM levels. In 24 h urine collected from two volunteers after they consumed broccoli and cauliflower, clearly sulforaphane mercapturic acid (133 micromol) and allyl isothiocyanate mercapturic acid (4.7 micromol) were found. This procedure demonstrates a reliable and efficient method to study the intake and mode of action of isothiocyanates in animal studies and clinical trials.  相似文献

The stability of creatine monohydrate (CrMH), crystallised guanidinoacetic acid (GAA‐C) and granulated GAA (GAA‐G) in a moist retorted and a dry extruded dog food formulation during production and storage was investigated. Commercial food mixtures were supplemented with CrMH, GAA‐C or GAA‐G. Uniformity after mixing and retorting or extrusion was determined based on replicate samples (moist n = 8, dry n = 10). Storage stability was evaluated at 25°C/60% relative humidity for 15 months and 40°C/75% for 6 months. Foods with CrMH were analysed for creatine (Cr) and creatinine (Crn), whereas GAA‐C and GAA‐G foods were analysed for GAA concentrations. Coefficients of variation (CV) for uniformity of the additives after mixing of moist and dry pet food formulations were below 15%, and the CV was lower in processed mixtures. Recoveries after retorting and extrusion were higher for GAA‐G (79 and 99%) and GAA‐C (89 and 86%) compared to CrMH (36 and 85%) foods. In moist CrMH food, Cr concentrations re‐increased by 54% whilst Crn concentrations decreased by 39% after storage at 25°C for 15 months. With total molar Cr + Crn remaining stable throughout storage, Crn and Cr appeared to effectively interconvert. Storage of the extruded CrMH food at 25°C for 15 months resulted in a 63% decrease in Cr and a 39% increase in Crn concentration. The decrease in Cr concentration was larger at 6 months storage at 40°C compared to 15 months storage at 25°C. Both GAA‐C and GAA‐G moist and dry foods were stable during storage (<10% decrease). This study showed that GAA is highly stable during production and storage of moist and dry canine foods whilst CrMH is relatively unstable, particularly during storage. The latter makes it difficult to establish a guaranteed Cr content in finished moist retorted and dry extruded foods with CrMH.  相似文献

The present study was conducted with privately owned dogs and cats to investigate whether a relationship exists between the dietary AGEs and the urinary excretion of AGEs, as indication of possible effective absorption of those compounds in the intestinal tract of pet carnivores. For this purpose, data were collected from both raw fed and dry processed food (DPF) fed to dogs and cats, through spot urine sampling and questionnaires. Raw pet food (RF, low in AGE diets) was fed as a primary food source to 29 dogs and DPF to 28 dogs. Cats were categorized into 3 groups, which were RF (n = 15), DPF (n = 14) and dry and wet processed pet food (DWF, n = 25). Urinary-free carboxymethyllysine (CML), carboxyethyllysine (CEL) and lysinoalanine (LAL) were analysed using ultrahigh-performance liquid chromatography (UHPLC)—mass spectrometry, and were standardized for variable urine concentration by expressing the AGE concentrations as a ratio to urine creatinine (Ucr) concentration (µg/µmol Ucr). Urinary excretion of CML, CEL and LAL in dogs fed with DPF was 2.03, 2.14 and 3 times higher compared to dogs fed with RF (p < .005). Similar to the dogs, a significant difference in CML:Ucr, CEL:Ucr and LAL:Ucr between the three diet groups was observed in cats (p-overall < 0.005, ANOVA), in which the RF fed group excreted less AGEs than the other groups. Linear regression coefficients and SE of CML:Ucr, CEL:Ucr and LAL:Ucr showed that body weight and neuter status were significantly correlated with CML and CEL excretion, but not to LAL excretion. Our results revealed a significant correlation between dietary AGEs and urinary excretion of free CML, CEL and LAL, and also showed that endogenous formation of these AGEs occurs in both dogs and cats under physiological conditions.  相似文献

Fern leaves, also called fronds, are often used in bouquets. Leaves of the sword fern (Nephrolepis cordifolia) consist of a central vascular tissue, with numerous leaflets (pinnae) at each side. Leaves that have been cut and immediately placed in water show abscission of the pinnae, starting from about day 4 of vase life, with 50% pinnae abscission on day 13. The onset of pinnae abscission was hastened by a period of dehydration (3, 6, 9 or 12 h at 25 °C). The time to 50% pinnae abscission was between 7.0 and 4.7 days after 3 h and 12 h of dehydration, respectively. Dehydration treatments might induce air emboli in the xylem, but in these experiments did not inhibit water uptake. Dehydration did increase the rate of ethylene production of the cut leaves throughout vase life. A 3 h treatment with 1-MCP at concentrations of 200 or 300 nL L?1 prior to the period of dehydration reduced the rate of ethylene production and reduced the rate of abscission. 1-MCP treatments were also effective if given after the period of dehydration. The data show that pinnae abscission limits the vase life of cut leaves of the sword fern, and that a short period of water stress drastically increases the rate of abscission. The increase in pinnae abscission was correlated with an increase in ethylene production. As 1-MCP alleviated the effect of dehydration on pinnae abscission, the dehydration effect involved ethylene perception. The data suggest that a small water stress induced an autocatalytic rise in ethylene production which was the direct cause of the increase in pinnae abscission.  相似文献

The unambiguous recognition of a species’ habitat is a matter of debate. For terrestrial species, habitat is often defined as physical patches of a certain vegetation type in a matrix of non-habitat. Ecological resources that make up the habitat of a species may, however, only cover subsets of vegetation types or can be spatially dispersed in a complex way over different vegetation types. Here we present and test a procedure to recognize and delineate habitat according to a resource-based approach instead of a vegetation-based approach. We used the green hairstreak butterfly (Callophrys rubi) in a heathland landscape as a study case. Our resource-based habitat approach selects those zones that comprise essential resources and conditions within an appropriate spatial window. Variables that were retained in a logistic regression model were used to calculate larval, adult and combined habitat indices in a GIS, taking into account thermal constraints on resource-use, as this is a key habitat aspect for this heliothermous insect. To group different (and sometimes scattered) ecological resources into functional habitat zones, we derived a measure of space-use from mark-release-recapture data. By least-cost modelling this spatial window was adapted to the nature of the vegetation between sets of resources. The habitat zones that were delineated using this approach matched the observed distribution of butterflies significantly better than did a classic approach based on vegetation types with host plants only. Our approach provides concrete output for conservation purposes, like recognizing zones with the highest potential for habitat restoration.  相似文献
