Deep Freezing of Boar Semen Packaged in Plastic Bags and Straws

For practical reasons, a large volume (i.e. 5 ml) of frozen boar semen per insemination dose is desirable, but successful freezing has not been achieved, since optimal cooling rates have not yet been established. Post-thaw motility and the acrosome intep'ty of semen from four boars frozen with a programmable freezin machine, in mini-(0.25 ml), maxi-(5 ml) plastic straws and in 10 × 5 cm PVC- or Teflon FEP-plastic bags (0.35 – 0.12 mm thick, 5 ml) was studied. The freezing of the semen was monitored using thermocouples placed in the straws and the bags. The freezing curve started from +5°C, at a rate of −3°C/min, to – 6°C, it was held for 1 min at −6°C, and was followed by further drop to −100°C at a rate of −20°C/min, with subsequent storage in LN₂. The bags had a much shorter freezing point plnteau, compared to the maxi-straws. Post-thaw sperm motility was significantly higher when semen was frozen in mini-straws or in bags than in maxi-straws. The freezing procedure did not cause major acrosomal damages, significantly more normal apical ridges being present in the bags and mini-straws than in the maxi-straws. This in vitro evaluation indicates that the freezing method employed is satisfactory for freezing large volumes of boar semen into plastic bags .     

Anaerobes in periodontal disease in the dog: a review

As anaerobic sampling and culture techniques improved, the documented prevalence of anaerobic bacteria in periodontal disease has increased. The anaerobic bacteria have become more well-known in humans and consequently in dogs, since this species is a major model in periodontal studies. A review of the literature related to anaerobic flora is described.     

Changes in the concentration of mRNAs for the inhibin subunits in ovarian follicles after administration of gonadotropins to progestin treated ewes.

RNA was extracted from single or small groups of ovine ovarian follicles after treatment of ewes with FSH and/or LH. The content of mRNA for the alpha-inhibin and beta A-inhibin subunits was analyzed by hybridization with specific cDNA probes. All ewes were treated with progestin vaginal pessaries to suppress spontaneous preovulatory follicle maturation and ewes were given three intramuscular injections of gonadotropins at 8-hr intervals starting 24 hr prior to collection of ovaries. In experiment I, both Schering-FSH and NIDDK-oFSH-17 (oFSH) significantly increased alpha- and beta A-inhibin mRNA per ewe in 2-5 mm follicles and tended to increase alpha- and beta A-inhibin mRNA in large (greater than 5 mm) follicles. In experiment II, oFSH and NIDDK-oLH-25 (oLH) were administered in a 2X2 factorial arrangement. Separate administration of oFSH or oLH increased (P less than .05) the alpha-inhibin mRNA concentration in large follicles. alpha-inhibin mRNA concentration in 4-5 mm follicles was also increased by oFSH but was decreased by oLH. Concomitant treatment with oFSH and oLH did not change alpha-inhibin mRNA concentrations from those measured in oFSH treated ewes. In experiment II, beta A mRNA concentrations followed a pattern similar to that of alpha A mRNA, but the differences were not statistically significant. We conclude that, in the ewe, exogenous FSH increases the concentration of inhibin mRNA in the whole follicle. The ability of exogenous oLH to alter expression of the inhibin subunit genes may depend upon the stage of follicle maturation.     

Characterization of and radioimmunoassay for canine thyroglobulin

Canine thyroglobulin (cTg) has been isolated and purified. It has similar electrophoretic patterns as Tg from other mammalian species. The main fraction had a MW of 660,000, whereas also fractions of a MW of approximately 1,300,000 (dimer) and 330,000 (subunit) were present. The iodine content was 0.8 to 1.0 % (w/w). cTg did not cross-react with antibodies against human Tg to a degree that would allow the use of a radioimmunoassay for human Tg for the determination of cTg in serum or plasma. Therefore a polyclonal antiserum was raised against cTg and a homologous radioimmunoassay was developed, which was sensitive (0.4 μg/l) and specific (cross-reactivity in cTg assay of human Tg, goat Tg, T₄, T₃, and DIT < 0.01 %).

Plasma Tg levels in normal dogs of both sexes and aged 3–15 years amounted to 192 ± 73 μg/l (mean ± SD, n=30). There was no relation between plasma Tg and T₄ levels.     

A new virus disease of watercress in England1

In 1983 plants showing symptoms typical of watercress chlorotic leafspot agent (WCLSA) were found in Kent (GB). Later, plants showing similar symptoms were found in Dorset (1986) and Hampshire (1987), the main watercress-producing regions of England. These plants, unlike those infected with WCLSA, contained virus particles which were isometric with diameters of 37-38 nm. An antiserum to the virus was produced, with a titre in immunodiffusion tests of 1/128 and this gave good results in immunosorbent electron microscopy (ISEM) and ELISA tests on crude plant sap. Purified preparations of the virus reacted positively in ISEM and immunodiffusion tests with an antiserum to watercress yellow spot virus, a partially described virus occurring in France. Tests suggest that the crook root fungus Spongospora subterranea f.sp. nasturtii is probably the vector of this virus but other means of transmission cannot be excluded. Virus incidence within watercress beds varied, with little or no infection in the water inlet region of the bed where there are also low levels of crook root infection, higher levels in the central region and highest levels at the outlet region where crook root infection is also highest.     

Efficacy of nemadectin, a new broad-spectrum endectocide, against natural infections of canine gastrointestinal helminths

Nemadectin, a new broad-spectrum endectocide, was highly efficacious against natural infections of all the major canine gastrointestinal helminths. At single oral dosages of 0.2-0.4 mg kg-1 body weight (BW), a liquid formulation administered in gelatin capsules was 100% effective in eliminating natural infections of Toxocara canis, Toxascaris leonina, Ancylostoma caninum and Uncinaria stenocephala. Tablets (267 mg) containing 4.6% nemadectin given at a rate of 1/3 tablet per 20 kg BW (0.2 mg nemadectin kg-1) were 100% active against T. canis, A. caninum and U. stenocephala. With both formulations, an increase in the dose rate to 0.6-0.8 mg kg-1 BW resulted in 99-100% elimination of Trichuris vulpis infections. No adverse reactions were observed in any of the treated dogs.
