Thymelaeaceae plants produce dextrorotatory dibenzylbutyrolactone lignans, which are opposite enantiomers to the lignans isolated from other plants (e.g.,Forsythia spp.). In our previous paper, (–)-pinoresinol (74% enantiomer excess), (+)-matairesinol (optically pure), and (+)-wikstromol (optically pure) were isolated fromWikstroemia sikokiana (Thymelaeaceae). In the present investigation, a survey of lignans and the determination of their enantiomeric compositions were continued. Four lignans, (–)-lariciresinol, (–)-secoisolariciresinol, (+)-kusunokinin, and (+)-methyltrachelogenin, were isolated from MeOH extracts ofW. sikokiana stem. To our knowledge, we have isolated (+)-methyltrachelogenin from plants for the first time. Chiral high-performance liquid chromatographic analysis showed that (+)-kusunokinin and (+)-methyltrachelogenin were optically pure, whereas (–)-lariciresinol and (–)-secoisolariciresinol were not (39% and 45% enantiomer excess, respectively). Feeding experiments with deuterium-labeled substrates demonstrated conversion of coniferyl alcohol to the lignans and interconversion of lignans. These reaction sequences are similar to the sequence catalyzed byForsythia enzymes. The effects of a higher concentrate feed during the raising period on the growth and carcass of Japanese Black cattle being fattened at a younger age were investigated using three pairs of artificially bred identical twins and three pairs of full‐sibs. The animals were fed the concentrate at 2.4%/day (2.4% group) and 1.2%/day (1.2% group) comparative bodyweight (BW) during the raising period. The BW and subcutaneous fat thickness were measured every month using ultrasonic equipment. The animals were slaughtered at the age of 24 months. The BW of the 2.4% group from the age of 6–10 months appeared to be higher than that of the 1.2% group (P < 0.1). The subcutaneous fat of the 2.4% group from the age of 7–11 months was thicker than that of the 1.2% group (P < 0.1). After those ages, the BW and subcutaneous fat of the two groups did not differ and there was no difference in the carcass characteristics between the two groups. Using a higher concentrate feed during the raising period for younger fattening of Japanese Black cattle influenced growth from the late half of the raising period to the early part of the fattening period. 相似文献
A total of 20 cases of disproportionate calves on Japanese Brown cattle were examined morphologically. Investigation of 5 affected calves revealed that the insufficiency of endochondral ossification was confined only to the long bones of the limbs and was not accompanied by other associated changes. On the basis of the histological changes of epiphyseal plate and the affected site, this condition may be called chondrodysplasia of the rhizomelic type. Therefore, this disorder was defined as a bovine dwarfism. From three dimensional image analysis and histological changes of the tibial proximal portion in the other 15 cases, deformity and shortness of the tibia were related to the state of distribution and the degree of damages of the epiphyseal plate. In the affected cartilage matrix, cystoid degeneration, fibrous striation, bone spicule, necrotic foci and rent were found. Inadequate metachromasia suggests the abnormal staining of sulfated glycosaminoglycans and alcian blue stainability may be attributable to the abnormal interactions between proteoglycan and other matrix components. At the interface of cartilage and bone, irregular calcification, fibrotic scar and sealing by osseous tissue with relation to vascularity were found. These changes in this study indicated the failure of modeling on the epiphyseal plate, showing disturbance of chondrocytic differentiation and abnormal formation of the matrix. 相似文献
Four lignans — pinoresinol, lariciresinol, secoisolariciresinol, matairesinol — were isolated from each ofDaphne odora andDaphne genkwa (Thymelaeaceae). Matairesinol isolated from both plants was optically pure (>99% e.e.) and dextrorotatory. Pinoresinol and lariciresinol isolated from the plants were not optically pure, and their enantiomeric compositions ranged from 88% to 95% e.e. in favor of (–)-enantiomers. As for secoisolariciresinol, the one fromD. odora was optically pure [(+)-enantiomer, >99% e.e.], and that fromD. genkwa was 97% e.e. in favor of the (+)-enantiomer. Lignan-synthesizing enzyme activity was detected from a Thymelaeaceae plant for the first time; cell-free extracts fromD. genkwa catalyzed the formation of (–)-lariciresinol (23% e.e.) from racemic (±)-pinoresinols. 1. Progressive alterations in oxidative phosphorylation of liver mitochondria were followed for 14 d in growing chickens fed on either semi‐purified low (7%) or high (61%) protein‐energy diet. Hepatic mitochondrial oxidative phosphorylation rates were assessed polarographically with pyruvate + malate as substrates.
2. The ADP:O values were reduced significantly 4 d after the feeding of a high‐protein‐energy diet, when compared with those in chickens fed on a low‐protein‐energy diet, whereas the state 3 oxidation rates in chickens fed on a high‐protein‐energy diet from day 6 to 14 were significantly lower than those in low‐protein‐fed chickens.
3. No changes in sensitivity of mitochondrial ATPase activity to oligomycin, expressed as % of total ATPase activity, were observed among chickens fed for 21 d on diets with various protein concentrations though the FoF1‐ATPase activity, expressed per mg protein, tended to decrease in chickens fed on high‐protein‐energy diet.
4. These results suggest that the reduced ADP:O values for liver mitochondria in the high‐protein‐fed chickens may not be involved in the degrees of integrity of the FoF1‐ATPase. 相似文献
Prebiotic fructooligosaccharides are noted for their intestinal immunodulating effects, and the identification of markers for the effects is a matter of great concern. This study aimed to identify marker genes for physiological effects of a particular fructooligosaccharide (FOS) on a host animal and also to define the target of its function in the small intestine. DNA microarray technology was used to screen candidate marker genes, and comprehensive changes in gene expressions in the ileum of mice fed with FOS were investigated. One of the major physiological effects of FOS was intestinal immunomodulation. Marker genes were then identified for major histocompatibility complex classes I and II, interferon, and phosphatidylinositol metabolites. Also, the ileum was segmented into Peyer's patch (PP) and the other ileal organ (DeltaPP), and these were analyzed by quantitative RT-PCR method, with the result that the site for recognizing the FOS function was the DeltaPP rather than the PP. This is the first paper showing the markers for the physiological effects of FOS in the small intestine at gene expression level. Applying these marker genes would make it possible to clarify the mechanisms of how the administration of dietary FOS and associated changes in the intestinal environment are recognized by host organisms as well as how its immunomodulating effects are expressed in the body. 相似文献
